Ogawa, Hiroshi;Kodera, Yasutake;Kanoh, Shigeyoshi;Ueyama, Akihiko;Motoi, Masatoshi research published 《 Preparation and Characterization of Side-Chain Liquid-Crystalline Polyoxetanes Anchoring a Pendant Spacer-Separated Mesogen at the Tertiary-Like C-3 Carbon of the Oxetane Unit》, the research content is summarized as follows. Polyoxetanes anchoring a pendant spacer-separated mesogen at the tertiary-like C-3 carbon of the oxetane unit were prepared by cationic ring-opening polymerization of the corresponding oxetane derivatives The resulting polymers were liquid-crystalline substances showing textures assignable to the nematic or smectic mesophase over a wide temperature range from about 260 °C to room temperature These mesogens also behaved in the manner similar to that of the mesogens in the analogous polyoxetanes attached by a Me side chain at the quaternary C-3 carbon, although the less bulky polymer backbone in the present work indicated a somewhat higher isotropic phase-transition temperature than that of the methyl-substituted analogs. In both types of the polyoxetanes, their mesophase patterns were influenced by the core structure and alkoxy tail length in the pendant mesogen, but not by the bulkiness of the second side chains, H and CH3, attached at the C-3 carbon of the polyoxetane unit.

This paper describes our continued efforts in the area of small-mol. apelin receptor agonists. Recently disclosed compound 2 showed an acceptable metabolic stability but demonstrated monodemethylation of the dimethoxyphenyl group to generate atropisomer metabolites in vitro. In this article, we extended the structure-activity relationship at the C2 position that led to the identification of potent pyrazole analogs with excellent metabolic stability. Due to the increased polarity at C2, the permeability for these compounds decreased. Further adjustment of the polarity by replacing the N1 2,6-dimethoxyphenyl group with a 2,6-diethylphenyl group and reoptimization for the potency of the C5 pyrroloamides resulted in potent compounds with improved permeability. Compound 21 (I) displayed excellent pharmacokinetic profiles in rat, monkey, and dog models and robust pharmacodynamic efficacy in the rodent heart failure model. Compound 21 also showed an acceptable safety profile in preclin. toxicol. studies and was selected as a backup development candidate for the program.

The reaction of hydroxide with epichlorohydrin in the gas phase was studied with ab initio calculations Three mechanisms were examined, each corresponding to nucleophilic attack at a different carbon atom of the substrate. Through attack at the ring methylene position was favored, whereas attacks at the alkyl methylene or ring methine positions have larger activation energies. These results are consistent with exptl. results found in the literature.

Merrill, G. N. research published 《 A computational study into the reactivity of epichlorohydrin and epibromohydrin under basic conditions in the gas phase and solution》, the research content is summarized as follows. Ab initio MO calculations were carried out on epibromohydrin (EBH) and epichlorohydrin (ECH) in an attempt to elucidate their reactivity with respect to a hard nucleophile, hydroxide. These systems were modeled in both the gas phase and a polar solvent under basic conditions. In the gas phase, a direct displacement mechanism (nucleophilic attack at the C1 position) was operative for EBH, while an indirect pathway (nucleophilic attack at the C3 position and subsequent intramol. displacement) was followed for ECH. In an acetone solution, only the indirect displacement mechanism occurs. An electrostatic argument is advanced to account for this behavior in polar solution

Muthukrishnan, Sivaramakrishnan;Shete, Vivekanand S.;Sanan, Toby T.;Vyas, Shubham;Oottikkal, Shameema;Porter, Lauren M.;Magliery, Thomas J.;Hadad, Christopher M. research published 《 Mechanistic insights into the hydrolysis of organophosphorus compounds by paraoxonase-1: exploring the limits of substrate tolerance in a promiscuous enzyme》, the research content is summarized as follows. We designed, synthesized, and screened a library of analogs of the organophosphate pesticide metabolite paraoxon against a recombinant variant of human serum paraoxonase-1 (PON1). Alterations of both the aryloxy leaving group and the retained alkyl chains of paraoxon analogs resulted in substantial changes to binding and hydrolysis, as measured directly by spectrophotometric methods or in competition experiments with paraoxon. Increases or decreases in the steric bulk of the retained groups generally reduced the rate of hydrolysis, while modifications of the leaving group modulated both binding and turnover. Studies on the hydrolysis of phosphoryl azide analogs as well as amino-modified paraoxon analogs, the former being developed as photoaffinity labels, found enhanced tolerance of structural modifications when compared with O-alkyl-substituted mols. Results from computational modeling predict a predominant active site binding mode for these mols., which is consistent with several proposed catalytic mechanisms in the literature and from which a mol.-level explanation of the exptl. trends is attempted. Overall, the results of this study suggest that while paraoxonase-1 is a promiscuous enzyme, there are substantial constraints in the active site pocket, which may relate to both the leaving group and the retained portion of paraoxon analogs. The three pan-genotypic HCV NS3/4A protease inhibitors (PIs) currently in clin. use-grazoprevir, glecaprevir, and voxilaprevir-are quinoxaline-based P2-P4 macrocycles and thus exhibit similar resistance profiles. Using our quinoxaline-based P1-P3 macrocyclic lead compounds as an alternative chem. scaffold, we explored structure-activity relationships (SARs) at the P2 and P4 positions to develop pan-genotypic PIs that avoid drug resistance. A structure-guided strategy was used to design and synthesize two series of compounds with different P2 quinoxalines in combination with diverse P4 groups of varying sizes and shapes, with and without fluorine substitutions. Our SAR data and cocrystal structures revealed the interplay between the P2 and P4 groups, which influenced inhibitor binding and the overall resistance profile. Optimizing inhibitor interactions in the S4 pocket led to PIs with excellent antiviral activity against clin. relevant PI-resistant HCV variants and genotype 3, providing potential pan-genotypic inhibitors with improved resistance profiles.

In order to study the impact of CO2 during n-heptane ignition delay under O2/CO2 conditions, this study presents a numerical and exptl. study in four cases (40 mol.% O2/60 mol.% CO2, 50 mol.% O2/50 mol.% CO2, 57 mol.% O2/43 mol.% CO2 and 65 mol.% O2/35 mol.% CO2). Firstly, a third body effect (TBE) model is presented, which considers the third body efficiency of CO2. Secondly, the TBE model adds the third body efficiency into a one-step global mechanism and obtains an ignition delay time formula according to the auto-ignition theory. Thirdly, experiments are carried out in a constant volume combustion chamber (CVCC) test system. Furthermore, the experiment and simulation results are compared and discussed. In addition, two parameters (α and β) are defined to study the third body effect and the chem. effect of CO2 resp. Finally, chem. kinetic analyses are used to study the CO2 effect. The results show that the TBE model is in agreement with the experiment and the maximum difference is 8.94% under the 57 mol.% O2/43 mol.% CO2 condition. The third body effect is the main effect of CO2 and it can decrease small mol. intermediates and radicals. Besides, C3H6O is the most affected species by the third body effect of CO2 among four prominent intermediate species. Furthermore, the third body effect of CO2 inhibits most paths of OH production H+ O2→O+ OH (R11) is the most affected pathway whose peak rate of production (ROP) decreases by 51.5%.
