Objective To explore the effect of oxytocin combined with carboprost tromethamine on postoperative hemorrhage, recovery of gastrointestinal function, and the first average urination time after cesarean section in full-term delivery. Methods A total of 130 women after full-term delivery were chosen from the hospital from Oct. 2014 to Oct. 2016, then they were divided into control group and observation group according to random number table, 65 women in each group. The women in observation group were treated by oxytocin combined with carboprost tromethamine. The amounts of postoperative hemorrhage at 2 and 24 h after operation, the time of gastrointestinal function recovery, the first average urination time, and the incidence rates of adverse effects in the two groups were compared. Results The amounts of postoperative hemorrhage at 2 and 24 h after operation in observation group were statistically significantly less than those in control group (t = 3.14, 3.30, P<0.05). The time of gastrointestinal function recovery in observation group was statistically significantly shorter than that in control group (t = 2.74, P<0.05). The first average urination time in observation group was statistically significantly better than that in control group (F = 3.78, P<0.05). There was no statistically significant difference in the incidence rates of adverse effects between the two groups (χ2 = 0.97, P>0.05). Conclusion Oxytocin combined with carboprost tromethamine used for full-term delivery after cesarean section can effectively reduce the amount of postoperative hemorrhage, promote recovery of gastrointestinal function and urination as soon as possible, and it doesn′t increase the incidence rates of adverse effects, the value is superior to oxytocin.

Using a high-throughput screening (HTS) approach, we have identified and validated several small-mol. Mcl-1 inhibitors (SMI). Here, we describe a novel selective Mcl-1 SMI inhibitor, 2 (UMI-77), developed by structure-based chem. modifications of the lead compound 1 (UMI-59). We have characterized the binding of UMI-77 to Mcl-1 by using complementary biochem., biophys., and computational methods and determined its antitumor activity against a panel of pancreatic cancer cells and an in vivo xenograft model. UMI-77 binds to the BH3-binding groove of Mcl-1 with Ki of 490 nmol/L, showing selectivity over other members of the antiapoptotic Bcl-2 family. UMI-77 inhibits cell growth and induces apoptosis in pancreatic cancer cells in a time- and dose-dependent manner, accompanied by cytochrome c release and caspase-3 activation. Coimmunoprecipitation experiments revealed that UMI-77 blocks the heterodimerization of Mcl-1/Bax and Mcl-1/Bak in cells, thus antagonizing the Mcl-1 function. The Bax/Bak-dependent induction of apoptosis was further confirmed using murine embryonic fibroblasts that are Bax- and Bak-deficient. In an in vivo BxPC-3 xenograft model, UMI-77 effectively inhibited tumor growth. Western blot anal. in tumor remnants revealed enhancement of proapoptotic markers and significant decrease of survivin. Mcl-1, an antiapoptotic member of the Bcl-2 family of proteins, is a validated and attractive target for cancer therapy. Overexpression of Mcl-1 in many cancers results in disease progression and resistance to current chemotherapeutics. Utilizing high-throughput screening, compound I was identified as a selective Mcl-1 inhibitor and its binding to the BH3 binding groove of Mcl-1 was confirmed by several different, but complementary, biochem. and biophys. assays. Treatment of human leukemic cell lines with compound II resulted in cell death through activation of caspase-3 and induction of apoptosis.

Totally 90 cesarean section women were involved and randomly divided into normal saline (N) group, ondansetron (O) group and midazolam (M) group. SBP, DBP, HR and SpO2 were observed before, 0 and 5 min after carboprost tromethamine injection (T0, T1 and T2, resp.). SBP and DBP were significantly higher, HR was faster and SpO2 was lower at T2 compared with those at T1 in 3 groups (P<0.05). SBP and DBP was lower, HR was slower and SpO2 was higher in M group than those in N group (P<0.05). Midazolam prevents adverse reactions in cesarean section women induced by carboprost tromethamine, and the effect is better than ondansetron.

To explore the clin. efficacy and safety of carboprost tromethamine combined with Barkri balloon in the treatment of postpartum hemorrhage. A total of 90 patients with postpartum hemorrhage admitted to our hospital from Jan. 2015 to Dec. 2016 were selected and randomly divided into a control group and an observation group with 45 cases each. In the control group, a Bakri balloon was injected from the vagina or uterine incision during cesarean section, and 300mL of normal saline was injected. Patients in the observation group were injected with carboprost tromethamine for combination therapy on the basis of the control group. Both groups of patients were treated for 24 h. The clin. efficacy of the two groups of patients was compared, systolic blood pressure (SBP), diastolic blood pressure (DBP) and blood oxygen saturation (SaO2), blood loss and adverse reactions were compared for safety evaluation of the combination. Results: After treatment, the total effective rates of the observation group and the control group were 93.33% and 80.00%, resp., and there was a statistical difference between the two groups (P < 0.05). There was no statistical difference in the amount of bleeding before treatment between the two groups of patients with postpartum hemorrhage (P > 0.05). After 2h and 24h of hemostatic treatment, the bleeding volume of the observation group was significantly less than that of the control group (P < 0.05). There was no significant difference in SBP, DBP and SaO2 between the two groups of patients with postpartum hemorrhage before and after treatment (P > 0.05). There was no significant difference in the occurrence of adverse reactions between the two groups (P > 0.05). There was no statistically significant difference in the incidence of adverse reactions between the two groups. Carprost tromethamine combined with Bakri balloon has good clin. efficacy and high safety in the treatment of postpartum hemorrhage, and it is worthy of clin. application.

Motherwort (YiMuCao), a traditional Chinese herb, has been shown beneficial effects for women’s diseases. This meta-anal. aimed to evaluate the efficacy and safety of motherwort injection add-on therapy to carboprost tromethamine for prevention of post-partum blood loss. A systematic literature search was conducted in PubMed, Embase, Cochrane Library, CNKI, VIP and Wanfang from their inception to Dec. 2017. Randomized controlled trials that determined the add-on effects of motherwort injection to carboprost for prevention of post-partum blood loss were eligible. Pooled risk ratio (RR) and mean difference (MD) with 95% confidence interval (CI) were used to summarize the effect sizes. Eight trials including 1276 pregnant women fulfilled the inclusion criteria. Prophylactic use of motherwort injection add-on therapy significantly reduced the post-partum 2 h (MD -127.5 mL; 95% CI -149.13 to -105.88) and 24 h (MD -146.85 mL; 95% CI -179.77 to -113.94) blood loss and incidence of post-partum hemorrhage (RR 0.28; 95% CI 0.17-0.45) than carboprost. Moreover, adjunctive treatment with motherwort injection significantly decreased the length of the third stage of labor (MD -3.41 min; 95% CI -4.33 to -2.49) and duration of lochia (MD -7.13 days; 95% CI -8.49 to -5.76). There was no statistical significant difference in the incidence of adverse events (RR 0.76; 95% CI 0.50-1.16). VProphylactic use of motherwort injection add-on therapy to carboprost tromethamine could reduce post-partum blood loss. However, more well-designed trials are necessary to confirm the findings of this study due to the methodol. flaws of the included trials.

The amounts of blood loss and urine volumes during operation and at 2 and 24 h after operation in the two groups were recorded, the incidence rates of adverse events in the two groups were compared. Results The amount of blood loss during operation and at 2 and 24 h after operation in study group were statistically significantly lower than those in control group (P<0.05). Urine volumes during operation and at 2 and 24 h after operation in study group were statistically significantly higher than those in control group (P<0.05). The incidence rate of adverse reactions in study group was statistically significantly lower than that in control group (P<0.05). Conclusion For the high-risk pregnant women receiving cesarean section, the clin. efficacy and safety of hemabate in prevention and treatment of postpartum hemorrhage are better than those of oxytocin combined with carboprost methylate suppositories, which is worthy of further promotion and application in clin. practice.

Objective: To observe the effect of carboprost tromethamine (hemabate) and oxytocin in cesarean section for preventing and reducing the intraoperative bleeding and compare their adverse reaction. Methods: From Nov. 2013 to March 2014, a total of 40 parturient women underwent cesarean section were included in the study. They were randomly divided into group I (20 cases using carboprost tromethamine injection) and group II (20 cases using oxytocin) according to different drugs for the injection into their uterine corpus immediately after fetal disengagement. There were no significant differences in maternal age, gestational age, body weight in general between two groups (P > 0.05). The study protocol was approved by the Ethical Review Board of Investigation in Human Being of Shanghai First Maternity and Infant Hospital. Informed consent was obtained from each participants. Results: The amount of blood loss during cesarean section in group I was obviously lower than that of group II (P < 0.05). There were significant differences in vital signs at 10 min after treatment between two groups (P < 0.05). The incidence rate of adverse reaction in group I was significantly higher than that of group II (P < 0.01). Conclusion: Carboprost tromethamine injection in caesarean operation can effectively decrease the intraoperative bleeding, but more attention should be paid on its adverse reaction.

Objective To observe the preventive effect of Guyuan Fujiu decoction combined with carboprost tromethamine on postpartum hemorrhage caused by uterine inertia in vaginal delivery. Methods A total of 110 cases of postpartum hemorrhage due to uterine inertia in the hospital from Apr. 2018 to June 2020 were randomly divided into control group and observation group, 55 cases in each group. The control group was given carboprost tromethamine injection after the delivery of the fetus, and the observation group was given Guyuan Fujiu decoction on the basis of the control group. The incidence of postpartum hemorrhage, amount of postpartum hemorrhage, hemostatic time, use of other hemostatic measures, changes of Hb and hematocrit were compared between the two groups. Results The postpartum hemorrhage rate of the observation group (3.64%) was significantly lower than that of the control group (14.55%), the difference was statistically significant (χ2=3.960, P<0.05). Postpartum hemostasis time [(23.56±3.14) min] was significantly shorter than that in the control group [(40.05±5.74) min], the difference was statistically significant (t=18.691, P<0.05). Compared with the control group, the amount of bleeding in the observation group decreased significantly at 30 min, 2 h and 24 h postpartum (t=8.654, 13.495 and 15.496, P<0.05). The application rate of other hemostatic measures in the observation group (7.27%) was significantly lower than that in the control group (21.82%) (χ2=4.681, P<0.05). Compared with the control group, the decrease of maternal Hb and hematocrit in the observation group were significantly reduced (t=22.817, 28.939, P<0.05). Compared with the control group, the serum D-dimer level in the observation group was significantly lower at 2 h postpartum, and the FIB level was significantly higher (t=19.492, 17.256, P<0.05). The level of serum antithrombin-III in the observation group was significantly higher than that in the control group (t=6.090, 10.092, P<0.05). The adverse reaction rate of the observation group (7.27%) was lower than that of the control group (16.36%), but the difference was not statistically significant (χ2=2.181, P>0.05). Conclusion Guyuan Fuguo decoction combined with carboprost tromethamine can be used to prevent uterine hypostasis postpartum hemorrhage, reduce postpartum hemorrhage, improve coagulation function, promote uterine contraction and drug safety.

Amperometric organic- and aqueous-phase peroxide biosensors were prepared with native and N-hydroxysuccinimide ester (NHS)-modified horseradish peroxidase (HRP). The ε-amino functions of the six free lysine residues of HRP were selectively modified with homobifunctional suberic acid bis(N-hydroxysuccinimide ester) (SA-NHS) or ethylene glycol bis(succinic acid N-hydroxysuccinimide ester) (EG-NHS). The biosensors were based on electrostatic complexation of the HRP variants with an osmium bis(bipyridyl)poly(4-vinylpyridine) polymer. The enzyme electrodes were operated in both aqueous (0.1 M phosphate buffer, pH 7.0) and organic (90% CH3CN) phases, for the detection of hydrogen peroxide and Me isothiocyanate. Kinetic anal. of the steady-state amperometry data showed that chem. modification of HRP about up to a ∼5-fold enhancement of the biosensor sensitivity for H2O2 and an improvement in both sensor stability and organotolerance. Also, NHS modification of HRP results in a 3-fold extension of the range of linear response of the peroxide sensor in both aqueous and nonaqueous media. When used as a Me isothiocyanate sensor in the organic phase, the amperometric responses of the biosensors were doubled on changing from native HRP to NHS-HRP electrodes.
