The cross-linked receptor was electrophoretically resolved into 2 oligomeric complexes of apparent mol. mass 288 kDa and 347 kDa, reflecting the association of the 53-kDa protein with a fraction of the receptor. Since no higher oligomeric complexes could be generated by crosslinking cell extracts under different conditions, it was concluded that most of the untransformed cytosolic receptor is devoid of addnl. components.

Microtubule assembly kinetics have been studied quantitatively under solution conditions supporting microtubule dynamic instability. Purified GTP-tubulin (Tu-GTP) and covalently cross-linked short microtubule seeds (EGS-seeds; Koshland et al. (1988) Nature 331, 499) were used with and without biotinylation. Under sub-critical concentration conditions ([Tu-GTP] < 5.3 microM), significant microtubule growth of limited length was observed on a proportion of the EGS-seeds by immuno-electron microscopy. A sensitive fluorescence assay for microtubule GDP production was developed for parallel assessment of GTP utilisation. This revealed a correlation between the detected microtubule growth and the production of tubulin-GDP, deriving from the shortening phase of the dynamic microtubules. This correlation was confirmed by the action of nocodazole, a specific inhibitor of microtubule assembly, that was found to abolish the GDP release. The variation of the GDP release with tubulin concentration (Jh(c) plot) was determined below the critical concentration (Cc). The GDP production observed was consistent with the elongation of the observed seeded microtubules with an apparent rate constant of 1.5 x 10(6) M-1 second-1 above a threshold of approximately 1 microM tubulin. The form of this Jh(c) plot for elongation below Cc is reproduced by the Lateral Cap model for microtubule dynamic instability adapted for seeded assembly. The behaviour of the system is contrasted with that previously studied in the absence of detectable microtubule elongation (Caplow and Shanks (1990) J. Biol. Chem. 265, 8935-8941). The approach provides a means of monitoring microtubule dynamics at concentrations inaccessible to optical microscopy, and shows that essentially the same dynamic mechanisms apply at all concentrations. Numerical simulation of the subcritical concentration regime shows dynamic growth features applicable to the initiation of microtubule growth in vivo.

Chem. crosslinking would conceivably cause structural disruption of a protein, but few cross-linkers have been fully evaluated in this aspect. Furthermore, integral membrane proteins may differ from soluble proteins in the selection of suitable cross-linkers, which has never been investigated. In this study, we systematically evaluated the impact of five conventional cross-linkers targeting Lys, Asp and Glu, and two Arg-reactive cross-linkers on the structural and functional integrity of two G protein-coupled receptors (GPCRs). Perturbation of the receptor structure and ligand-binding activity was observed, depending on the receptor and crosslinking conditions. In particular, our study demonstrated that the concentrations of PDH and KArGO need to be fine-tuned in order to minimize the structural and functional disturbance of specific GPCRs. A set of amenable cross-linkers was selected to acquire the most comprehensive cross-link maps for two GPCRs. Our in-depth crosslinking mass spectrometry (CXMS) anal. has revealed dynamic features of structural regions in GPCRs that are not observable in the crystal structures. Thus, CXMS anal. of GPCRs using the expanded toolkit would facilitate structural modeling of uncharacterized receptors and gain new insights into receptor-ligand interactions.

The upregulation of the protein myeloid cell leukemia-1 (Mcl-1) is closely associated with various human cancers, which can result in the evasion of apoptosis and a low survival rate. Therefore, developing Mcl-1 inhibitors has become a promising paradigm for cancer therapy. Interestingly, compound I (R1 = 3,5-di-Me-4-Cl-Ph, X = H, R2 = Ph, n = 1) showed not only potent activity against Mcl-1 but also considerable selectivity over Bcl-2 and Bcl-xL, which was rationalized by mol. docking and fragment-centric topog. mapping (FCTM). This paper aims to explore the clin. anal. of carbetocin combined with romethamine for preventing high-risk postpartum hemorrhage after caesarean section. 76 Cases of high-risk factor caesarean section puerpera were divided into control group and treatment group(n=38). In control group, the puerpera were injected with oxytocin 20 IU right after delivery, and i.v. infused with carbetocin 100 μg. Based on the treatment of control group, the treatment group was injected with romethamine 250 μg. The hemorrhage amount, side effect of drug and exhaust time in both groups were observed during operation, 2 h and 24 h after operation. The hemorrhage amounts during operation, 2 h and 24 h after operation in treatment group were significantly less than those of control group(all P<0.05). The exhaust time of treatment group was shorten than that of control group(P<0.05). There were no significant differences in drug side effects between two groups. Carbetocin combined with romethamine has definite effect and less side effect for preventing high-risk postpartum hemorrhage after caesarean section.

Objective: To observe the effect and safety of hemabat (H) on prevention of postpartum hemorrhage in cesarean section and after cesarean section in high risk pregnant women. Methods: Four hundred and sixty-nine pregnant women with high hemorrhagic risk factors including twin pregnancy, polyhydramnios, fetal macrosomia, placenta previa were planned cesarean section. A total of 457 pregnant women were divided into 3 groups by operation indications. There were 239 cases of fetal macrosomia, 145 cases of twin pregnancy and polyhydramnios, and 73 cases of placenta previa. Three kinds of hysterotonics were used randomly in each group. Group oxytocin (O): 20 U oxytocin injected into the uterine plus 20 U oxytocin intravascularly, 152 women; Group oxytocin + hemabate (O + H): 20 U oxytocin and 250 μg hemabat injected into the uterine, 192 women; group H: 250 μg hemabat, injected into the uterine, 125 women. The amount of bleeding during the operation and within 2-h after delivery were measured. The side effect of each group was observed Results: The amount of bleeding during cesarean section in group O was (445 ± 262) ml, in group 0 + H (332 ± 218) ml, and in group H (375 ± 265) ml. There was an extremely significant difference between group O and group O + H (P < 0.01). The amount of bleeding within 2 h after delivery in group O was (176 ± 193) ml, in group O + H was (110 ± 114) ml, and in group H was (124 ± 103) ml. There was a significant difference between groups O, O + H and H. Among the 469 women, 31 had total amount of bleeding more than 1000 mL during operation and within 2 h after delivery, 48% (15 women) were in group O, 23% in group O + H and 29% in group H. The total amount of bleeding in group O was much more than group O + H and group H in the group of fetal macrosomia (P < 0.01, P < 0.01). Similar results were found in the group of twin pregnancy and polyhydramnios (P < 0.01, P < 0.01). The total amount bleeding in group O + H was much less than group O in the group of placenta previa (P < 0.05). There were 5% (12) pregnant women whose total amount of bleeding was ≥ 1000 mL in the group of fetal macrosomia, 8% (11) in the group of twin pregnancy, 11% (8) in the group of placenta previa. No hysterectomy was done among the women. The incidence of side effects in the three groups was 2.6%, 11.5% and 7.0% resp. Vomiting was frequently seen in the latter two groups, but recovered soon without treatment. Conclusion: Hemabat can significantly reduce the amount of bleeding during the cesarean section in pregnant women with high hemorrhagic risk factors and can be used with oxytocin as firstline medicine to prevent hemorrhage during and after delivery.

OBJECTIVE: The paper explored the clin. efficacy of intraoperative injection of carboprost tromethamine to prevent postpartum hemorrhage in cesarean sections with placenta previa. Methods: The paper selected 54 cases of cesarean delivery with placenta previa who were treated in the hospital from March 2013 to March 2015 as the research subjects, and were randomly divided into observation group and control group, 27 cases in each group. The mothers in the observation group were injected with carboprost tromethamine in the uterine body after the newborn was delivered, while the women in the control group were given oral misoprostol. The paper observed and recorded the occurrence of bleeding and adverse reactions in the two groups during and after operation, within 2 h and within 24 h. Results: The intraoperative blood loss of patients in the postpartum observation group, and within 2 h and 24 h after the operation were lower than those in the control group, and the difference was statistically significant (P < 0.05); the incidence of maternal adverse reactions was significantly lower in the observation group than in the control group. The difference was statistically significant (P < 0.05). Conclusion: Intraoperative injection of carboprost tromethamine during cesarean delivery with placenta previa could effectively prevent postpartum hemorrhage, reduce the amount of intraoperative and postoperative bleeding, and improve the quality of life of the woman.
