OBJECTIVE: We evaluated the efficacy of electroacupuncture combined with tropisetron in treating carboprost tromethamine-induced nausea and vomiting during cesarean section under lumbar anesthesia. METHODS: A total of 264 patients aged 22-40 years were enrolled, who received carboprost tromethamine and suffered nausea and vomiting during cesarean section under lumbar anesthesia. The patients were divided randomly into the control group, electroacupuncture group, tropisetron group, and electroacupuncture + tropisetron group. RESULTS: Compared to the control group, the nausea and vomiting scores decreased at T3 in both the electroacupuncture and electroacupuncture + tropisetron groups, and decreased at T4 in the electroacupuncture group, tropisetron group, and electroacupuncture + tropisetron group; the motilin, gastrin, and 5-hydroxytryptamine (5-HT) levels decreased at T5 in the other 3 groups. Compared to the electroacupuncture + tropisetron group, the nausea and vomiting scores increased at T3 in the control and tropisetron groups, and increased at T4 in the other 3 groups; the motilin, gastrin, and 5-HT levels increased at T5. CONCLUSIONS: Our study suggested that electroacupuncture combined with tropisetron could effectively relieve carboprost tromethamine-induced nausea and vomiting during cesarean section under lumbar anesthesia. The effect was better than its single application, and the reduced 5-HT, motilin, and gastrin levels might be involved in the underlying mechanism.

The preventive effect on postpartum bleeding in Cesarean section by carboprost tromethamine injection combined with calcium gluconate was investigated. A total of 192 maternal patients cases with the risk factors of postpartum hemorrhage and a clear indication of Cesarean section from Jan.2010 to Jan.2013 in Traditional Chinese Medicine Hospital of Beijing Shunyi District were randomly divided into oxytocin group and combination therapy group with 96 cases in each: the oxytocin group was given oxytocin 10-20 U and carboprost 10 mg, on the basic of which, the other group was added carboprost tromethamine injection 25 μg. The amount of bleeding within 24 h and adverse reactions were observed in the two groups. The postoperative hemorrhage during operation and within 24 h after operation of the combination therapy group were significantly less than oxytocin group [(216.9±54.4) mL vs (360.6±52.8) mL, (128.0±29.1) mL vs (221.9±42.7) mL, P<0.05]. And adverse reactions in the combination therapy group were significantly less than the oxytocin group [10.4% (10/96) vs 31.3% (30/96), P<0.05]. Carboprost tromethamine injection combined with calcium gluconate could effectively prevent intraoperative postpartum hemorrhage of Cesarean section, reduce adverse reaction, thus was worth of promotion in clin.

Mitochondrial dysfunction is associated with the occurrence of a variety of neurodegenerative diseases, especially Alzheimer disease (AD). As a mitochondrial quality control process, mitophagy is greatly inhibited in AD; increasing evidence shows that the induction of mitophagy is an effective therapeutic intervention strategy. However, the lack of more safe, effective, and clear mechanisms for mitophagy inducers has limited the clin. application. In recent studies, we have identified a small mol. compound, UMI-77, that can safely and effectively induce mitophagy. UMI-77 is an established BH3-mimetic for MCL1 and was developed to induce apoptosis in cancer cells. We found that UMI-77 can bind MCL1 and enhance its function as a mitophagy receptor protein, thus enhancing its interaction with LC3A to induce mitophagy. UMI-77 effectively improves the cognitive decline seen in an AD mouse model. Our findings shed light on the novel mechanisms of mitophagy, reveal that MCL1 is a mitophagy receptor that can be targeted to induce mitophagy, and identify MCL1 as a drug target for therapeutic intervention in AD.

Group B had high hemorrhage than Group A 2 h postoperatively and 24 h postoperatively with statistical significance (all P < 0.05). Group A had postoperative hemorrhage > 500 mL 8 cases (20%), uterine filling and surgery 3 cases (7.5%) and nausea and vomiting 17 cases (42.5%). Group B had postoperative hemorrhage 500 mL 17 cases (42.5%), uterine filling and surgery 6 cases (15), and nausea and vomiting 32 cases (80%) with statistical significance (all P < 0.05). Conclusion: The Hemabate combined with perioperative i.v. infusion of Na-K-Mg-Ca-glucose injection for high-risk cesarean section could reduce hemorrhage, Hemabate dose, and nausea and vomiting.

Polyenemacrolides such as amphotericin B (AmB) are thought to assemble together and form an ion channel across plasma membranes. Their antimicrobial activity has been attributed to this assemblage, whose stability and activity are dependent on sterol constituents of lipid bilayer membranes. The structure of this channel-like assemblage formed in biomembranes has been the target of extensive investigations for a long time. As the first step toward this goal, we prepared several AmB dimers with various linkers and tested their channel-forming activity. Among these, AmB dimers that bore an aminoalkyl-dicarboxylate tether covalently linked between amino groups of AmB showed potent hemolytic activity. Furthermore, K+ influx actions monitored by measuring the pH of the liposome lumen by 31P NMR revealed that the dimers formed a mol. assemblage similar to that of AmB in phospholipid membrane. Judging from changes in 31P NMR spectra, the dimers appeared to induce “all-or-none”-type ion flux across the liposome membrane in the presence of ergosterol, which suggests that the ion channel formed by ergosterol/dimer is similar to that of AmB. On the basis of this data, we are now trying to elucidate the structure of the ion-channel complex by making the labeled conjugates of AmB for NMR measurements.

Results The amounts of endometrorrhagia in exptl. group and control group were (71.43±13.96) ml and (50.56±10.78) ml, resp. (P<0.05); the amounts of colporrhagia in exptl. group and control group were (35.31±6.32) ml and (44.27±8.31) ml, resp. (P<0.05); the incidence rates of adverse reactions in exptl. group and control group were 16.12% and 11.66%, resp. (P>0.05). Conclusion It is recommended to treat uterine inertia patients by i.m. injection to prevent and treat postpartum hemorrhage; cervical injection should be selected to prevent postpartum hemorrhage among the patients with other causes.

Objective: The paper observed the effect of oxycodone on the adverse reactions of carprost tromethamine during cesarean section and its effect on inflammatory cytokines. Methods: Fifty parturients undergoing cesarean section with elective spinal anesthesia, aged 25-35 years old, ASA grade I or II, were divided into oxycodone group (group A) and control group (B) by random number table method group). Group A was given oxycodone 0.07 mg/kg after carboprost tromethamine was given to the parturients, and group B was given symptomatic treatment after carboprost tromethamine adverse reactions occurred in the parturients. The two groups of parturients applied the same formula for postoperative analgesia, and both groups were diluted with normal saline. The paper observed the adverse reactions after the administration of carprost tromethamine and observed the changes in heart rate (HR), mean arterial pressure (MAP), and pulse oxygen saturation (SpO2) at 2, 5, and 10 min, and observed the changes before and after the administration of oxycodone. Maternal peripheral blood interleukin-2 (IL-2) and VAS scores at relevant time points after surgery were performed at 6, 12, 24, and 48 h after surgery. Results: Comparing group A and group B, the adverse reactions of carboprost tromethamine were significantly reduced, the level of IL-2 in peripheral venous blood was significantly increased at 6, 12, 24, and 48 h after surgery (P < 0.05), VAS score Significantly reduced (P < 0.05). Compared with group B, there were no statistically significant differences in MAP, HR, and SpO2 at 2 and 10 min after carboprost tromethamine (P > 0.05); but 5 min after carprost tromethamine was administered when compared with group B, MAP and HR were significantly reduced, the difference was statistically significant (P < 0.05), and the SpO2 difference was not statistically significant (P > 0.05). Conclusion: Oxycodone 0.07 mg/kg applied in cesarean section could significantly reduce the adverse reactions of carprost tromethamine, provide good postoperative analgesia, and increase the secretion of IL-2 after cesarean section. The vital signs of women who were given proper oxycodone during cesarean section were more stable.

Adipocytes exposed to cAMP for 24 h exhibited a 25% decrease in cellular protein and marked decreases in enzyme activity (88%) and PC mRNA abundance (98%) compared with untreated adipocyte controls. After 48 h of exposure to cAMP, PC activity and PC mRNA diminished to levels approaching their detection limits. When exposed to medium containing cAMP plus insulin, adipocyte enzyme activity and PC mRNA declined more slowly during the first 24 h exposure (about 20% decrease) but after 48 h fell to values comparable with those of adipocytes exposed to cAMP alone. Despite these decreases in enzyme activity, the PC protein content of adipocytes treated with cAMP alone or cAMP plus insulin are nearly identical with that of control adipocytes. The inactivation of PC in cAMP-treated adipocytes does not involve loss of the prosthetic groups from the holoenzyme. Crosslinking experiments suggest that the spatial arrangement of protomers in inactive PC may differ from that in the active tetrameric enzyme. Data presented suggest that, in addition to inducing inactivation, cAMP may also regulate adipocyte PC by decreasing transcription of the PC gene and/or enhancing the rate of degradation of PC mRNA.

The clin. efficacy of midazolam and dezocine for hemabate (carboprost tromethamine injection) induced side effects in the patients undergoing caesarean section was explored. Forty patients used hemabate during saesarean section were randomly divided into two groups, each with twenty cases. Before hemabate intrauterine injection, the patients in the control group were given normal saline 3 mL i.v., while those in the exptl. group were given midazolam 2 mg and dezocine 5 mg i.v. Respiratory function, circulatory function and side effects were observed before and after hemabate medication. The changes of MAP, HR, RR and SpO2 in the exptl. group were fewer than those in the control group at any time point (P<0.05). The incidence of side effects in exptl. group was lower than that in the control group (P<0.05). Midazolam and dezocine could be used effectively and safely for bemabate-induced side effects during caesarean section.

The aim of this paper is to observe the effects of carboprost tromethamine on the amount of bleeding in cesarean section of high risk parturients at different time points. The clin. data of 180 high risk parturients with postpartum hemorrhage tendency in cesarean section were retrospectively analyzed and divided into treatment group and control group according to the duration of carboprost tromethamine treatment. Treatment group was given intrauterine injection of carboprost tromethamine 250 μg and i.v. dripping of oxytocin 10 u after the delivery of fetus; control group was given intrauterine injection of carboprost tromethamine 250 μg if the amount of bleeding increased because uterine inertia and uterine massage was invalid, following intrauterine injection of oxytocin 10 u i.v. after the delivery of fetus. Both groups were given carboprost tromethamine continuously, depending on the situation. The amounts of bleeding and adverse drug reactions were recorded in 2 groups. The incidence of postpartum hemorrhage in treatment group was 17.7%, which was significantly lower than in control group(28.7%); the amounts of bleeding in treatment group 2 h and 24 h after operation were significantly lower than in control group; there was statistical significance(P<0.05). Adverse drug reaction of 2 groups was slight and recovered spontaneously. The application of carboprost tromethamine in advance could effectively decrease postpartum hemorrhage in high risk parturients with postpartum hemorrhage tendency, and it’s safety.
