The purpose of the present work was to mask the taste of Ambroxol hydrochloride and to formulate its patient-friendly dosage form. Complexation technique using KYRON -T-114 an ion-exchange resin was used to mask the bitter taste and the drug loading onto the ion-exchange resin was optimized for ratio of drug to resin, effect of pH and effect on soaking time. The resinate was evaluated for taste masking and characterized by FT-IR and DSC. Then the taste-masked drug was formulated into a fast dissolving tablet (FDT). The developed tablets were evaluated for hardness, friability, drug content, weight variation, content uniformity, friability, water absorption ratio, in-vitro disintegration time and in-vitro drug release studies. IR and DSC spectra revealed that there were no possible interactions between the drug and polymer. Maximum loading was obtained at drug- resin ratio 1:2, pH 7, and soaking time 30 min. A successful taste masking of resinate was confirmed by time intensity method. The values of pre-compression parameters evaluated and were within prescribed limits and indicated good free flowing properties. The data obtained of post-compression parameters found superior over conventional formulation. The F3 batch with disintegration time 30.4±0.64 s and dissolution 98.4±2.69 was selected as optimized formulation, This was compared with conventional marketed formulation and was found superior. The obtained results revealed that Ambroxol hydrochloride has been successfully taste masked and formulated into an FDT as a suitable alternative to the conventional tablets.

Chromatog. was carried out on UHPLC (WATERS)⊕MMETRY C18 4.6mm × 1000mm, 3.5μm, (Agilent – Zorbax Eclipse Plus C18 – Rapid Resolution) with an isocratic mobile phase with pH 3.0 composed of buffer, methanol and Acetonitrile (60:20:20) with a flow rate of 0.8mL/min. The detection was carried out with column temperature at 25°C using a UV detector at 276nm. Validation parameters like linearity, specificity, precision, accuracy, limit of detection (LOD), limit of quantification (LOQ), system suitability, Solutions stability and robustness were considered as affirmed in the ICH guidelines. Retention times for LEV, GUA & AMB were 1.07 min, 1.99 min & 3.55 min resp. The assay of syrups with the relative standard deviation found to be less than 2%. The parameters values were found, and the method was found to be satisfactory. This validated UHPLC method is cost-effective, receptive and precise than other chromatog. methods.

AH loading in erythrocytes using preswell dilution technique with glutaraldehyde (GA) as a crosslinking agent was evaluated and validated. Drug-loaded erythrocyte was characterized in terms of in vitro drug release followed by osmotic fragility study which showed amplified drug entrapment efficiency (DEE) and Hb content values as well. In vivo lung fibrosis study, rats were sensitized to egg albumin by i.p. (i.p.) injection and then inhalation in a whole body inhalation chamber. A sign of inflammation, airway sub-mucosal fibrosis, hypertrophy, and hyperplasia was observed A series of in vivo studies were carried out to describe the effect of AH-loaded RSE including measurement of cytokines in Bronchoalveolar Lavage (BAL) fluid and histopathol. study. AH showed a stepwise reduced level of cytokines in BAL at a different time interval after being injected of AH-loaded RSE. Furthermore, in vivo lung distribution experiments were performed for optimized formulation, and degree of distribution of the drugs inside the targeted organ was found to be satisfactory.

Thus, for example, quaterthiophene was obtained in 6 min by reaction of 2-bromo-2,2′-bithiophene with bis(pinacolato)diboron (isolated yield 65%), whereas quinquethiophene was obtained in 11 min by reaction of dibromoterthiophene with thienylboronic acid (isolated yield 74%). The synthesis of new chiral 2,2′-bithiophenes is reported. The detailed anal. of the byproducts of some reactions allowed us to elucidate a few aspects of reaction mechanisms. While the use of microwaves proved to be very convenient for the coupling between conventional thienyl moieties, the same was not true for the coupling of thienyl rings to thienyl-S,S-dioxide moieties. Indeed, in this case, the targeted product was obtained in low yields because of the competitive, accelerated, Diels-Alder reaction that affords a variety of condensation products.
