Association of TCF7L2 gene polymorphisms, methylation, and gene-environment interaction with type 2 diabetes mellitus risk: A nested case-control study in the Rural Chinese Cohort Study

Background: To assess the associations of single-nucleotide polymorphisms (SNPs) and methylation of transcription factor 7-like 2 (TCF7L2) gene with type 2 diabetes mellitus (T2DM) risk and further explore the interactions among SNPs, methylation, and environmental factors involved in T2DM risk. Methods: We conducted a nested case-control study with 290 pairs of T2DM cases and matched controls. We genotyped 3 SNPs of TCF7L2 in all included participants and tested 14 CpG loci of TCF7L2 in 76 pairs of cases and controls. Conditional logistic regression models were used to estimate odds ratios (ORs) and 95% confidence intervals (95% CIs) for T2DM risk according to SNPs and methylation of TCF7L2. Multifactor dimensionality reduction (MDR) analysis was used to explore the potential TCF7L2 gene-environment interactions in T2DM risk. Results: We found no statistically significant association between the TCF7L2 polymorphisms and T2DM risk. We observed significant positive associations of methylation at CpG5 and CpG7_8 with T2DM risk. For each 1% increase in DNA methylation at CpG5 and CpG7_8, T2DM risk increased 12% (OR 1.12.95% CI 1.01-1.25) and 32% (OR 1.32, 95% CI 1.07-1.63), respectively. Additionally, MDR analyses identified significant SNP-environment interactions among rs290487, alcohol drinking, and hypertension and methylation-environment interactions among CpG5, CpG7_8 and hypertension (P <0.05). Rates and predictors of brief intervention for women veterans returning from recent wars: Examining gaps in service delivery for unhealthy alcohol use

Background: Approximately one in four women veterans accessing the Department of Veterans Affairs (VA) engage in unhealthy alcohol use. There is substantial evidence for gender-sensitive screening (AUDIT-C = 3) and brief intervention (BI) to reduce risks associated with unhealthy alcohol use in women veterans; however, VA policies and incentives remain gender-neutral (AUDIT-C = 5). Women veterans who screen positive at lower-risk-level alcohol use (AUDIT-C = 3 or 4) may screen out and therefore not receive BI. This study aimed to examine gaps in implementation of BI practice for women veterans through identifying rates of BI at different alcohol risk levels (AUDIT-C = 3-4; =5-7; =8-12), and the role of alcohol risk level and other factors in predicting receipt of BI. Methods: From administrative data (2010-2016), we drew a sample of women veterans returning from recent wars who accessed outpatient and/or inpatient care. Of 869 women veterans, 284 screened positive for unhealthy alcohol use at or above a gender-sensitive cut-point (AUDIT-C >= 3). We used chart review methods to abstract variables from the medical record and then employed logistic regression comparing women veterans who received BI at varying alcohol risk levels to those who did not. Results: While almost 60% of the alcohol positive-risk sample received BI, among the subset of women veterans who screened positive for lower-risk alcohol use (57%; AUDIT-C = 3 or 4) only 34% received BI. Nurses in primary care programs were less likely to deliver BI than other types of clinicians (e.g., physicians, psychologists, social workers) in mental health programs; further, nurses in women’s health programs were less likely to deliver BI than other types of clinicians in mixed-gender programs; Those women veterans with more medical problems were no more likely to receive BI than those with fewer medical problems. Conclusions: Given that women veterans are a rapidly growing veteran population and a VA priority, underuse of BI for women veterans screening positive at a lower-risk level and those with more medical comorbidities requires attention, as do potential gaps in service delivery of BI in primary care and women’s health programs. Comparison of determination of sugar-PMP derivatives by two different stationary phases and two HPLC detectors: C-18 vs. amide columns and DAD vs. ELSD

Analytes were separated chromatographically by two common high performance liquid chromatography (HPLC) columns and subsequently detected by two tandem detectors. Results pointed out that C-18 column had a stronger capacity to separate the reducing sugar-PMP derivatives rather than the sugar themselves. While amide column could only effectively separate the original sugars. These phenomena demonstrated that polarity of reducing sugars decreased after their PMP derivatization. Moreover, both diode array detector (DAD) and evaporative light scattering detector (ELSD) were able to detect the reducing sugar-PMP derivatives, including the PMP-derivative of the oligosaccharide hydrolysate of chitosan, though the DAD exhibits a higher sensitivity than ELSD. In conclusion, carbohydrates themselves are more likely to be efficiently separated by an amide column and detected by ELSD, while DAD combined with a C-18 column has more power to determine carbohydrates derivatives.

A microstructural analysis was performed to determine the presence of oxygen in a wire rod section of AISI 1008 steel with a surface mechanical failure produced during wire drawing. The failure zone was analyzed by comparison using three different attack reagents: alkaline sodium chromate (ASC), ASC with hydrogen peroxide and amyl alcohol, and a solution of nitric acid in ethyl alcohol. The reagents were applied in samples in the failure zone, showing different types of zones in regions with internal and superficial defects such as carbides, pores, cracks, deformation, and detachment zones, indicating the possible presence of oxygen. The areas identified were observed by means of an optical microscope and were correlated with the content of elements that were present in the region using a scanning electron microscope and a scattered X-ray energy spectrometer, which determined that the areas identified by chromate sodium correspond to regions with a high concentration of oxygen and slag-forming elements such as silicon, aluminum, and iron; this is associated with the presence of oxygen in the oxide form. It was also observed that the ASC and the modified ASC solution with hydrogen peroxide and amyl alcohol could identify areas with a high presence of oxides, while the nitric acid solution only identified the steel microstructure. Alcohol consumption and risk of stomach cancer: A meta-analysis

Stomach cancer is one of the most common cancers in the world. The relationship between alcohol consumption and the risk of stomach cancer remains unclear. Epidemiology studies investigating this relationship have shown inconsistent findings. A meta-analysis was performed to explore the association between alcohol consumption and increased stomach cancer risk. Eighty-one epidemiology studies, including 68 case-control studies and 13 cohort studies, were included in this study. A significant association was found between alcohol consumption and increased risk of stomach cancer (OR = 1.20, 95% CI 1.12-1.27). To explore the source of the significant heterogeneity (p < 0.05, I-2 = 86%), analysis was stratified by study type (case-control study and cohort study), control type (hospital-based control and population-based control), gender (male, female, and mix), race (White and Asian), region (United States, Sweden, China, Japan), subsite of stomach cancer, and type of alcohol. The stratified analyses found that region and cancer subsite are major sources of the high heterogeneity. The inconsistent results in different regions and different subsites might be related to smoking rates, Helicobacter pylori infection, obesity, and potential genetic susceptibility. The positive association between drinking and increased risk of stomach cancer is consistent in stratified analyses. The dose-response analysis showed a clear trend that a higher daily intake of alcohol is associated with a higher risk of stomach cancer. Developing value-added material from industrial waste is one of the sustainable ways of recycling solid waste produced from the leather industry. Nanofibers can be used as sound absorbers due to their properties like porosity and high surface area. In this study, collagen hydrolysate extracted from waste leather trimmings was utilized to produce multilayer hybrid sound-absorbing material. Collagen hydrolysate was electrospun along with polyvinyl alcohol (PVA) and the layer was sandwiched between polyacrylonitrile (PAN) nanofibrous layers. The hierarchical structure of the composite is more porous on outer layers than medium porous inner collagen hydrolysate- PVA layer. The hybrid material was characterized using various experimental techniques and the sound absorption was measured using two-microphone impedance tube method. From acoustic measurements, it was revealed that the composite showed improved sound absorption in the frequency range of 800-2500 Hz due to its varying pore size. Hence, the leather trimmings as a component of sound-absorbing material creates an innovative solution for discarded leather waste and they can be used in practical applications like room acoustics.

Alterations in the function of prefrontal cortex (PFC) and hippocampus have been implicated in underlying the relapse to alcohol seeking behaviors in humans and animal models of moderate to severe alcohol use disorders (AUD). Here we used chronic intermittent ethanol vapor exposure (CIE), 21d protracted abstinence following CIE (21d AB), and re-exposure to one vapor session during protracted abstinence (re-exposure) to evaluate the effects of chronic ethanol exposure on basal synaptic function, neuronal excitability and expression of key synaptic proteins that play a role in neuronal excitability in the medial PFC (mPFC) and dentate gyms (DG). CIE consistently enhanced excitability of layer 2/3 pyramidal neurons in the mPFC and granule cell neurons in the DG. In the DG, this effect persisted during 21d AB. Re-exposure did not enhance excitability, suggesting resistance to vapor-induced effects. Analysis of action potential kinetics revealed that altered afterhyperpolarization, rise time and decay time constants are associated with the altered excitability during CIE, 21d AB and reexposure. Molecular adaptations that may underlie increases in neuronal excitability under these different conditions were identified. Quantitative polymerase chain reaction of large-conductance potassium (BK) channel subunit mRNA in PFC and DG tissue homogenates did not show altered expression patterns of BK subunits. Western blotting demonstrates enhanced phosphorylation of Ca2+/calmodulin-dependent protein kinase II (CaMKII), and reduced phosphorylation of glutamate receptor GluN2A/2B subunits. These results suggest a novel relationship between activity of CaMKII and GluN receptors in the mPFC and DG, and neuronal excitability in these brain regions in the context of moderate to severe AUD.
