Freitas, David S.;Rocha, Diana;Castro, Tarsila G.;Noro, Jennifer;Castro, Vania I. B.;Teixeira, Marta A.;Reis, Rui L.;Cavaco-Paulo, Artur;Silva, Carla research published 《 Green Extraction of Cork Bioactive Compounds Using Natural Deep Eutectic Mixtures》, the research content is summarized as follows. Quercus suber cork generates bioactive components, such as phenolic acids, terpenoids, and tannins, with excellent biol. properties, including antioxidant, anti-inflammatory, and antiaging activities. Aiming to design environmentally benign processes to eliminate, or reduce, the use of toxic chems., we propose the green extraction of bioactives from cork using natural deep eutectic solvents (NADES). Several deep eutectic mixtures were developed, through the mixture of natural compounds, namely, lactic acid, glycerol, ethylene glycol, sodium citrate, and sodium lactate, chosen according to their origin, toxicity, biocompatibility, polarity, and pH. The results revealed higher extraction yields when using NADES instead of harsh solvents like dioxane, with the extraction process governed by several phys.-chem. parameters, including pH, polarity, viscosity, and d., and also by the method of extraction Acidic NADES composed of lactic acid and glycerol, or sodium citrate, extracted a greater amount of aromatic compounds, terpenoids, and fatty acids and their derivatives More basic eutectic mixtures, composed of sodium lactate and a polyol (ethylene glycol or glycerol), extracted predominantly low mol. weight polar compounds The extracts range encompassed by the developed NADES, together with the associated nontoxicity, low price, and ease of preparation, establish these solvents as a green approach to extract high added-value compounds from cork.

Ehlers, Stephanie;Szczerbowski, Daiane;Harig, Tim;Stell, Matthew;Hotling, Susan;Darragh, Kathy;Jiggins, Chris D.;Schulz, Stefan research published 《 Identification and Composition of Clasper Scent Gland Components of the Butterfly Heliconius erato and Its Relation to Mimicry》, the research content is summarized as follows. The butterfly Heliconius erato occurs in various mimetic morphs. The male clasper scent gland releases an anti-aphrodisiac pheromone and addnl. contains a complex mixture of up to 350 components, varying between individuals. In 114 samples of five different mimicry groups and their hybrids 750 different compounds were detected by gas chromatog./mass spectrometry (GC/MS). Many unknown components occurred, which were identified using their mass spectra, gas chromatog./IR spectroscopy (GC/IR)-analyses, derivatization, and synthesis. Key compounds proved to be various esters of 3-oxohexan-1-ol and (Z)-3-hexen-1-ol with (S)-2,3-dihydrofarnesoic acid, accompanied by a large variety of other esters with longer terpene acids, fatty acids, and various alcs. In addition, linear terpenes with up to seven uniformly connected isoprene units occur, e. g. farnesylfarnesol. A large number of the compounds have not been reported before from nature. Discriminant analyses of principal components of the gland contents showed that the iridescent mimicry group differs strongly from the other, mostly also separated, mimicry groups. Comparison with data from other species indicated that Heliconius recruits different biosynthetic pathways in a species-specific manner for semiochem. formation.

Single cultivar Olivastro di Bucchianico extra virgin olive oil is obtained from olives cultivated in a narrow area of the Abruzzo region, Italy. This cultivar is mostly present in the municipality of Bucchianico and in some neighboring municipalities in the province of Chieti. There is very little research in the literature describing the morphol. and chem. characteristics of this cultivar. A morphol. characterization of the plant and the fruit was carried out. In addition, we characterized the chem., phys.-chem. and sensory properties of the extra virgin olive oil. The following analyses were conducted: free acidity, peroxide value, UV spectrophotometric indexes, contents in fatty acid Et esters, waxes, tocopherols, fatty acids, triglycerides, sterols, alcs., phenolic substances, volatile compounds and sensory profile. The anal. of the volatile compounds was performed using a headspace-gas chromatog.-ion mobility spectrometry (HS-GC-IMS) instrument connected to a nitrogen generator for carrier/drift gas production The results of the chem. analyses showed good levels of nutraceutical components in the oils, which were found to be organoleptically well balanced with medium values of fruity, bitter and pungent. The HS-GC-IMS method based on the anal. of 15 volatile mols. might be a useful tool for a chemometric discrimination of the varietal origin for the oils under investigation. 2021 Society of Chem. Dong, Hua;Chen, Shan;Zhu, Jianxun;Gao, Ke;Zha, Wenlong;Lin, Pengcheng;Zi, Jiachen research published 《 Enhance production of diterpenoids in yeast by overexpression of the fused enzyme of ERG20 and its mutant mERG20》, the research content is summarized as follows. Yeast has been widely used for large-scale production of terpenoids. In yeast, modifications of terpenoid biosynthetic pathways have been intensively studied. tHMG1 (encoding the catalytic domain of 3-hydroxy-3-methylglutaryl CoA reductase of yeast) and UPC2-1 (the G888D mutant of UPC2 encoding a transcription factor) were integrated into yeast chromosome, and ERG9 (the squalene synthase gene of yeast) was knocked down to yield the chassis strain DH02. A F96C mutation in ERG20 (farnesyl diphosphate synthase of yeast) was conducted to obtain mERG20 which can function as a geranylgeranyl diphosphate synthase (GGPS). Then, three fused genes, including BTS1 (the yeast innate GGPS)-ERG20, ERG20-mERG20 and mERG20-ERG20, were constructed, and expressed either by the pESC-based plasmids in DH02, or by being integrated into DH02 chromosome. The highest geranylgeraniol (GGOH) content was observed in the extracts of DH12 integrated with ERG20-mERG20, corresponding to 3.2 and 2.3 folds of those of the strains integrated with BTS1 and mERG20, resp. Finally, three genes encoding nor-copalyl diphosphate synthase (nor-CPS), ent-CPS and syn-CPS were integrated into the chromosome of DH12, resp., to construct yeasts for producing corresponding copalyl diphosphates (CPPs). Thus, a yeast-based platform was built for characterizing all types of diterpene synthases using GGPP or various CPPs as their substrates.

Phytochem. anal. of methanolic extract of A. subulatum dry fruits (MEAS) confirmed the presence of different bioactive secondary metabolites. The MEAS scavenged reactive free radicals and inhibited lipid peroxidation in vitro. To confirm the antioxidant efficiency of MEAS, in vivo experiment was carried out in which MTX was administered to induce oxidative stress. Co-administration of MEAS reduced MTX-induced hepatic, renal, and pulmonary toxicities via significantly (p < .01) enhancing antioxidant status and reducing oxidative stress. The MTX treatment significantly (p < .01) increased liver and kidney toxicity markers and increased proinflammatory cytokine (TNF-α, IL-1β, and IL-6) levels. However, co-administration of MEAS significantly (p < .01) reduced their levels, and tissue histopathol. confirmed the protective effect of MEAS in maintaining normal tissue architecture following MTX treatment. Protective effect of MEAS is accredited to the antioxidant and anti-inflammatory properties exhibited by bioactive compounds in MEAS.

Du, Chunyan;Li, Yanan;Fan, Junting;Tan, Rui;Jiang, Hezhong research published 《 Chemical composition, antioxidant and antimicrobial activities of essential oil from the leaves of Lindera fragrans Oliv》, the research content is summarized as follows. The chem. composition of the essential oil obtained by hydrodistillation from the leaves of Lindera fragrans Oliv. was determined by gas chromatog. (GC) and gas chromatog. coupled with mass spectrometry (GC-MS). Sixty two compounds accounting 76.45% of the essential oil were identified. The main constituents found to be spathulenol (27.63%), ledol (6.81%), β-caryophyllene (4.01%), (+)-cis-limonene oxide (3.69%), α-cadinol (3.24%). The disk diffusion method on antimicrobial activities revealed that it has remarkable inhibition effect against Escherichia coli (CP009072.1), Staphylococcus aureus (CP009361.1), Pseudomonas aeruginosa (CP015117.1) and Candida albicans (FJ159643.1). Antioxidant capacity of the essential oil was evaluated by 2,2′ -diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and β- carotene bleaching assay, and it did not show effective antioxidant activity.

Cui, Qingyu;Ye, Liqin;Guo, Chang-e;Li, Weidong research published 《 Assessing Volatiles in Three Chinese Dwarf Cherry Cultivars during Veraison and Maturation Using Headspace-Solid Phase Microextraction with Gas Chromatography-Mass Spectrometry》, the research content is summarized as follows. Chinese dwarf cherry is a native shrub in northwest China with a rich and unique fruit aroma. This study aims to determine the changes in volatile profiles during the maturation period, which provides a theor. basis for the optimal harvest times and the breeding of aroma-rich varieties. The variation in the production of 164 volatile compounds from three Chinese dwarf cherry cultivars, namely, “Jing’ou 1”, “Jing’ou 2”, and “Jing’ou 3”, were investigated by headspace-solid phase microextraction (HS-SPME)-GC-MS. These volatiles mainly constituted alcs., carbonyls, esters, terpenoids, and hydrocarbons. Their maturation process could be divided into three stages, namely prophase, metaphase, and anaphase. Prophase contained an abundance of hydrocarbons and carbonyls, primarily benzaldehyde being dominant among all volatiles. During metaphase, volatiles remained at a low level of abundance and diversity. Anaphase coincided with full maturation and was associated with esters and terpenoids; in particular, “Jing’ou 3” presented more compound diversity and a high level of acetate esters. The periods including the week prior to veraison and the week during maturation were particularly critical in volatile formation in Chinese dwarf cherries. This study reveals that the low level or lack of hexanal might be one of the distinctive characteristics separating Chinese dwarf cherries from other Cerasus or Rosaceae fruits.
