Facile and robust fabrication methodologies of complex materials with high added value for controlled drug delivery systems are highly sought-after in the pharmaceutical industry. Here, the authors have relied on the use of supercritical fluids for the matrix synthesis as well as for the drug impregnation process. As a model system of a hydrophobic and moisture sensitive active agent, triflusal was impregnated in different aerogel and polymeric matrixes using supercritical CO2. In vitro release profiles of drug carriers consisting of SiO2 aerogels as mesoporous monoliths and as microporous nanospheres loaded with triflusal are reported and compared to a more conventional polymeric system (PMMA). Silica-based matrixes are found to prevent the hydrolization of the active ingredient more efficiently than the polymeric matrix and displayed much faster release kinetics. Moreover, the triflusal is dispersed in a mol. form inside the silica-based materials. Such features are considered of great interest to enhance the bioavailability of low solubility drugs.

A comprehensive study of 14 hybrid aerogels of different composition with applications in drug delivery has been carried out. The overall objective was to modulate the release behavior of drug-impregnated aerogels, from an almost instantaneous release to a semi-retarded delivery prolonged during several hours, through internal surface functionalization. The designed hybrid aerogels were composed of silica and gelatin and functionalized with either Ph, long (16) hydrocarbon chain or Me moiety. As model systems, three class II active agents (pKa < 5.5), ibuprofen, ketoprofen and triflusal, were chosen to impregnate the aerogels. The work relied on the use of supercritical fluid technol. for both the synthesis and functionalization of the hybrid aerogels, as well as for the impregnation with an active agent using supercritical CO2 as a solvent. For the impregnated aerogels, in vitro release profiles were recorded under gastric and intestinal pH-conditions using HPLC techniques. The release behavior observed for the three studied drugs was explained considering the measured dissolution profiles of the crystalline drugs, the aerogel composition and its functionalization. Such features are considered of great interest to tailor the bioavailability of drugs with low water solubility

Abstract: Background: Black bean is one of the important beans as the second most consumed bean globally after soybean. It has been reported as a sustainable source of anthocyanins, important natural colorants used in food and consumer products. However, industrial uses of black bean anthocyanins in pharmaceutics and cosmetics have rarely been explored. Thus, here, a black bean anthocyanin extract was prepared, quality controlled and developed into an efficient hair coloring product. Results: Black bean was macerated in hydrochloric ethanol to produce a dark-reddish 1.85 ± 0.04% dry extract with total anthocyanin content (TAC) of 1.24 ± 0.48 μg/mL cyanidin-3-glucoside. The extract was readily dissolved in propylene glycol (25%), giving a reddish to red-brownish solution at pH of 4.00-7.00. The addition of antioxidant significantly (p = 0.000) enhanced the stability of the extract in an accelerated stability test (90.34 ± 5.32 from 67.83 ± 3.45% TAC remaining). Three base hair gels containing different thickeners were developed to yield clear gels with a viscosity of 101,666.67-447,333.33 cps and a pH of 4.80-4.97. The gel base with the best hair application properties was chosen to mix with 8, 10 and 12% black bean extract to produce similar visually assessed red-brown gels (L* = 33.25-33.76, a* = 0.06-0.09, b* = – 0.45-0.38 and h* = 277.54-283.31) at 73,733.33-99,666.67 cps and a pH of 5.00-5.01. All of the black bean hair coloring gels were stable under the accelerated test. The 10% hair gel was tested for its hair staining efficacy. The black bean hair coloring gel was found to efficiently color hair, and the color was maintained through four washings. Conclusions: An alternative application approach of using black bean as a natural colorant is presented here, including its basic requirements for preparation and quality control from an industrial aspect. The efficient semi-permanent hair coloring product containing black bean is an innovative natural product and is considered as one progress towards a sustainable bio-based economy using a readily available industrial crop, black bean. A redox-neutral photocatalytic strategy to accomplish this goal in both β-O-4 and β-1 lignin models at room temperature (RT) via proton-coupled electron transfer (PCET) process without any pretreatments of substrate, by adjusting the alkalinity of base to obtain a lignin models/base PCET pair with a bond dissociation free energy close to 102 kcal/mol was developed. Without breaking down Cβ-Cγ bond and any C-O bonds, this PCET method is 100% atom economy and produces exclusive Cα-Cβ bond cleavage products, such as benzaldehydes (up to 97%) and Ph ethers (up to 96%), in high to excellent yields and selectivities. Preliminary studies indicated that the PCET strategy was also effective for the depolymerization of native lignin at RT, thus providing significantly important foundation to the depolymerization of lignin.

The byproduct (HMDS or NH3·H2O) of the first step turned out to be the promoter for the second step, which could efficiently proceed in aqueous media without the addition of other catalysts. The reactions using trifluoroethyl salicylates 2-OH-3-R6-4-R5-5-R4-6-R3C6HC(O)OCH2CF3 could be performed under mild conditions to ensure the generation of vulnerable DHXs I and II in high yields. The substrate scope is very broad regardless of the substituent type and its position on the structure. Specifically, the versatility of DHXs I (R = R1 = R3 = R4 = R5 = R6 = H; R2 = Me) was demonstrated by their conversion to xanthones I (R = 3-methylbut-2-en-1-yl; R1 = R3 = R4 = R5 = R6 = H; R2 = Me)/II (R7 = H) and other complex structures III/IV.

Recently the D1 protease located in the thylakoid membranes of higher plants was found to be a potential herbicide target and based on a lead compound [Nα-[(5-isoxazolyl)carbonyl]lysine, D1 protease inhibitor] a series of novel N5-[(phenylamino)carbonyl]-N2-[(3-phenyl-5-isoxazolyl)carbonyl]lysine derivatives were designed. The synthesis of the target compounds was achieved using as reactants 3-phenyl-5-isoxazolecarbonyl chlorides and N5-[(phenylamino)carbonyl]lysine and the products thus obtained were confirmed by IR, 1H-NMR and elemental anal. The biol. activity was examined in vivo and it was discovered that most compounds possessed moderate herbicidal activity. Meanwhile, the e activity of one compound was evaluated against spinach D1 protease (i.e., thylakoid protein precursor processing peptidase, chloroplast protein precursor-processing proteinase, photosystem D1 protein precursor carboxyl-terminal processing proteinase, proteinase CtpA) and the result showed its inhibiting effect against this enzyme.

In this Article, the authors report the structures and photoelectrochem. (PEC) performances of a family of TOCs, including pure [Ti12O8(OEt)16L8] ({Me-Ti12}) and six Cd-doped clusters formulated as [H4Cd2Ti10O8(OEt)16(L)8(H2O)2] ({Cd2Ti10}; L = salicylic acid and their derivatives). The six Cd-doped clusters are isostructural, containing the same {Cd2Ti10O8} core, but are protected by salicylic ligands modified with different functional groups. The compositions, structures, and solution stability of these clusters were studied in detail by single-crystal x-ray diffraction and electrospray ionization mass spectrometry measurements. The embedding of heterometallic Cd(II) and chem. modification of organic protective shells can effectively regulate the PEC water oxidation activity of those clusters, with {F-Cd2Ti10} having the highest turnover number of 518.55 and the highest turnover frequency of 172.85 h-1. The authors’ work highlights the potential of using TOCs that do not contain noble metals as water oxidation catalysts, and their catalytic activity can be regulated by structural modification.
