Tuo-Min-Ding-Chuan decoction (TMDCT) is a Traditional Chinese Medicine (TCM) formula consisting of twelve herbs that can relieve the symptoms and treat allergic asthma. Yet, the underlying mechanism of action is still unclear. In this study, we investigated the effect of TMDCT in regulating Treg/Th17 cells immune balance and explored potential metabolic and gut biomarkers associated with Treg and Th17 cells in eosinophilic asthma mice treated by TMDCT. We found that TMDCT increases Treg cells percentage and decreases Th17 cells percentage in the ovalbumin (OVA) -induced eosinophilic asthma mice model. Furthermore, Imidazoleacetic acid, DL-glutamine, L-pyroglutamic acid, 2-deoxy-D-glucose were preliminary identified as biomarkers in plasma metabolites treated by TMDCT, meanwhile genus Desulfovibrio, genus Butyricimonas and genus Prevotella 9 were preliminary identified as gut microbiota biomarkers after TMDCT treatment. These results provide an exptl. foundation for the treatment of allergic asthma with Chinese herbal compounds

A de novo hit-to-lead effort involving the redesign of benzimidazole-containing antagonists of the CXCR4 receptor resulted in the discovery of a novel series of 1,2,3,4-tetrahydroisoquinoline (TIQ) analogs. In general, this series of compounds show good potencies (3-650 nM) in assays involving CXCR4 function, including both inhibition of attachment of X4 HIV-1IIIB virus in MAGI-CCR5/CXCR4 cells and inhibition of calcium release in Chem-1 cells. Series profiling permitted the identification of TIQ-(R)-stereoisomer I as a potent and selective CXCR4 antagonist lead candidate with a promising in vitro profile. The drug-like properties of I were determined in ADME in vitro studies, revealing low metabolic liability potential. Further in vivo evaluations included pharmacokinetic experiments in rats and mice, where I was shown to have oral bioavailability (F = 63%) and resulted in the mobilization of white blood cells (WBCs) in a dose-dependent manner.

Nicorandil (NCD) is a chem. unstable drug and sensitive to humidity, heat, and mech. stress in manufacturing processes. In this work, the cocrystals of NCD with organic acids were designed and synthesized to enhance the stability of NCD. Cocrystals of NCD with 1-hydroxy-2-naphthoic acid, salicylic acid, 3-hydroxybenzoic acid, and 2,5-dihydroxybenzoic acid were obtained. Hydrothermal stability study showed that cocrystals performed with remarkable stability advantages compared with pure NCD. On the basis of mechanism research, the stability improvements of NCD mols. can be attributed to the realignments of NCD mols. in cocrystals. After the formation of cocrystals, the novel packing styles of NCD break the self-catalyzed decomposition process in pure NCD structure. Moreover, intrinsic dissolution rate characterizations showed that the formation of cocrystals could also optimize the dissolution behavior of NCD to realize the objective of sustained release. The novel cocrystals stabilize nicorandil at mol. levels through lengthening the distances between N1 and C8 of adjacent mols.

In this study, the volatility of three typical wine aromas in model wine was investigated by HS-SPME-GC-MS, NMR, and sensory evaluation as influenced by different concentrations and structural properties of phenolics. Results showed that three phenolic fractions (phenolic acids, monomeric/oligomeric and polymeric procyanidins) exhibited different matrix effects on floral, fruity, and aged aromas perception. Physico-chem. and sensory analyses together indicated that all fractions reduced the perceived intensity of fruity and aged aroma attributes, and displayed stronger retention effects on fruity aromas at higher mDP and concentrations Monomeric/oligomeric and polymeric procyanidins promoted highly hydrophobic floral aromas release, whereas inhibiting the volatility of low hydrophobic fruity aromas. NMR confirmed that the reduction in the volatility of rose oxide, Et butanoate and whiskey lactone was attributed to interactions with epicatechin. This study aims to provide new thoughts and theor. support for wine aroma regulation during winemaking by reconstructing the phenolic composition in wine.

Proper chem. treatment of recycled carbon fiber composites (RCFC) has been demonstrated to potentially add value to them, by improving the performance of cementitious materials incorporating them. This laboratory investigation mainly explored the use of saturated (simulated concrete) pore solution (SPS) to treat RCFC that contains cured epoxy resin, before its use in cementitious mortar. A total of seven mortar mixtures were fabricated, incorporating untreated RCFC, RCFC treated by SPS for different time periods followed by triisopropanolamine (TIPA), or RCFC treated by montmorillonite nanoclay emulsion (mNCE, at the nanoclay dosage of 0.75% by weight of cementitious binder). The water-to-binder (w/b) ratio of all mortar mixtures was 0.50 and the RCFC was admixed at 1% by volume The compressive strength and splitting tensile strength of RCFC reinforced mortar were evaluated. The chem. characterization (by FTIR), thermogravimetric anal., and microscopic investigation (by SEM) helped explain the observed engineering performance data. The residue epoxy resin on the surface of the RCFC was removed by the immersion in SPS, but a long-time treatment may result in damage of the fiber surface. The chem. treatment of RCFC showed both mech. and chem. benefits to its adhesion with the cementitious mortar. Admixing mNCE achieved outstanding strengths of the mortar similar to those of mortars treated by SPS for 0.5 h and then TIPA.

Correlation analyzes suggest that the reductions in serum lipids by RRTJ are associated with the gut microbial community (Dorea, Coprobacillus and Roseburia, Bifidobacterium) and their associated metabolites (palmitoleic acid, succinic acid, propionic acid, Glyceric acid). This paper aims to investigate the effect of triisopropanolamine (TIPA) on compressive strength and hydration of cement-fly ash paste. The samples with various dosages of TIPA were prepared with 30% fly ash (FA) and 70% cement (water/binder ratio by weight = 0.38), and cured under the standard condition. The compressive strength, pore structure, hydration process, and hydration products were investigated. The results show that TIPA can obviously increase the compressive strength of cement-FA system at the age of 7 d and 60 d, and the reasons are involved in pore structure and hydration of cement-FA system. Pore structure was characterized with mercury intrusion porosimetry, and the results show that TIPA can reduce total porosity but increase the amount of pore with size more than 50 nm, implying the air-entraining effect with neg. effect on compressive strength. The result suggests that TIPA and defoaming agent should be used together to minimize the neg. effect in real concrete. Furthermore, anal. of hydration products shows that TIPA can accelerate the hydration of both cement and FA, and this can also be illustrated from solid-state NMR. It is noticed that TIPA can hasten the conversion of AFt to AFm, which can be indicated from hydration heat. Addnl., the acceleration of pozzolanic reaction of FA is because TIPA can accelerate the dissolution of aluminate, silicate, and ferric into liquid paste which was demonstrated from morphol. characterization and the change of ions in pore solution Such results would be expected to provide experience for the use of alkanolamine in promoting the performance of cement-based materials.

Cloud loss of orange juice could be effectively inhibited by centrifugal treatment, but it can induce flavor changes, which become a new challenge for the industry. This work aims to investigate the effect of centrifugation on flavor changes in orange juice and explore its possible mechanism. Taste- and aroma-related attributes were analyzed, and pectin was characterized. Indicated that pH (4.00), total soluble solid (9.67°Brix), titratable acidity (0.42%), sucrose (44%), fructose (29%), and glucose (27%) were less affected by centrifugation (P > 0.05). However, aroma compounds significantly changed (P < 0.05), where terpenes and alcs. tended to be distributed in pulp and serum after centrifugation, resp. Pearson correlation anal. showed that aroma compound distribution induced by centrifugation was highly related to chelator-solubilized pectin fraction and sodium carbonate-solubilized pectin fraction (|R| > 0.9). In general, centrifugation clearly changed aroma of orange juice, which was mainly affected by pectin characteristics.
