An efficient and mild one-pot convergent synthesis protocol has been developed for benzo[b]oxazolo[3,4-d][1,4]oxazin-1-one derivatives through the Mitsunobu reaction and sequential cyclization. Various tricyclic fused benzoxazinyl-oxazolidinones (20 examples) were obtained in good to excellent yields and high enantioselectivities with facile operation. Furthermore, four stereoisomers were afforded resp. in high ee values (>97.8%) via using different chiral 2,3-epoxy-4-trityloxybutanol. This methodol. has been applied to the synthesis of key intermediates of drug candidates.

The objective of this paper was to establish the method system for identification of 2-keto-L-gulonic acid (2-KLG) in fermentation broth, and to distinguish 2-KLG from its byproducts (5-DKG and 2-DKG). The standard substance and fermentation product samples were analyzed by thin layer chromatog.(TLC), HPLC and m.p. determination, then the results were compared to confirm the consistency of the three methods in identification of 2-KLG. The TLC results indicated that the Rf value, color and size of chromatogram were concordant between fermentation product and the standard substance of 2-KLG. HPLC results showed that the fermentation product exhibited concordant retention time (8 min), m.p.(170 °C or so) and pH value with standard substance of 2-KLG, but different from 2-DKG and 5-DKG. This method system could identify the existence of 2-KLG in unknown fermentation products, with great significance in screening of new strains of 2-KLG from further fermentation of D-glucose.

The study aims to investigate the antioxidant effect of glutathione on promoting 2-KGA in Vc fermentation system using Ketogulonicigenium vulgare 25B-1 and Bacillus endophyticus ST-1 as the co-culturing microbes. Methods and Results : The activities of antioxidant-related enzymes and qPCR were used to study the antioxidant effect of glutathione addition in Vc fermentation system. The addition of GSH and GSH/GSSG increased 2-KGA production and decreased fermentation time, and the highest 2-KGA production increased by 40·63% and the lowest fermentation time shortened to 60 h when the addition of optimal concentration ratio of GSH/GSSG was 50 : 1. Moreover, the increased production of 2-KGA was accompanied by up-regulated the activities of total antioxidant capacity (T-AOC), total superoxide dismutase (T-SOD), catalase (CAT) and over-expressed oxidative stress-related genes sod,gst,gr,zwf,gp, which resulted in scavenging reactive oxygen species to reduce oxidative stress in Vc fermentation system. Conclusions : Glutathione showed a significant effect on increasing 2-KGA production and decreasing fermentation time in Vc fermentation system. GSH/GSSG could maintain a dynamic balance with two forms of glutathione and the optimal concentration ratio of GSH/GSSG was 50 : 1. Significance and Impact of the Study : Glutathione is proved to be effective to relieve oxidative stress. The promotion effects of GSSG and GSH on 2-KGA production could help to further explore the optimization of co-culture fermentation process for Vc industrial production

(OPLS-DA), it was found that there were 25 categories and 145 differential metabolites with significant changes (VIP>1 and p<0.05) in beverages before and after fermentation, mainly including 34 organic acids and their derivatives, 21 flavonoids, 20 sugars and their derivatives, 16 alkaloids, 10 nucleotides and their derivatives 10 amino acids and their derivatives The bioactive substances with significantly increased content after fermentation include 36668 times of 1-hydroxy-2-naphthoic acid, 1000 times of dihydroresveratrol, 800 times of 2-hydroxyvaleric acid, 517 times of daidzein, 488 times of vitexin, 238 times of apigenin, 148 times of isoerucic acid, 130 times of genistein, etc. The anal. of KEGG metabolic pathway showed that there were 19 metabolic pathways with significant differences. It is speculated that grain substrate may have certain effects on the synthesis and accumulation of secondary metabolism of L. paracasei L1 and L. casei YQ336. This study will lay a theor. foundation for the fermentation of plant matrix food by lactic acid bacteria and the metabolic mechanism of plant matrix.

Aminoalcs. have been addressed as activating buffers for alk. phosphatase. However, there is no record on the buffer activation regarding organophosphorus hydrolase (OPH). Here we reported the activating effects of aminoalcs. on OPH-catalyzed hydrolysis of diisopropylfluorophosphate (DFP), an analog mol. of G-type warfare agents. The kinetic parametors kcat, Vmax and kcat/Km in the OPH reaction were remarkably increased in the buffers (pH 8.0, 25 °C) containing aminoalcs. with C2 between nitrogen (N) and oxygen (O) in their structures, including triethanolamine (TEA), diethanolamine, monoethanolamine, 1-amino-2-propanol, 2-amino-2-methyl-1-propanol, and triisopropanolamine. In contrast, much lower or no rate-enhancing effects were observed in the adding of amines, alcs., amine/alc. mixtures, or 3-amino-1-propanol (C3 between N and O). The 300 mM TEA further increased DFP-degrading activities of OPH mutants F132Y and L140Y, the previously reported OPH mutants with desirable activities towards DFP. However, the treatment of ethylenediaminetetraacetate (EDTA) markedly abolished the TEA-induced activation of OPH. The product fluoride effectively inhibited OPH-catalyzed hydrolysis of DFP by a linear mixed inhibition (inhibition constant Ki ∼ 3.21 mM), which was partially released by TEA adding at initial or later reaction stage. The obtained results indicate the activation of OPH by aminoalc. buffers could be attributed to the reduction of fluoride inhibition, which would be beneficial to the hydrolase-based detoxification of organophosphofluoridate.

Com. production of 2-keto-L-gulonic acid (2-KLG), the immediate precursor of L-ascorbic acid, is by Ketogulonicigenium vulgare in co-culture with Bacillus megaterium. We used flux balance anal. (FBA) to study a genome-scale metabolic model (GSMM) of K. vulgare, iWZ663, and found that K. vulgare is deficient in nutrient biosynthetic pathways. Individually omitting L-glycine, L-cysteine, L-methionine, L-tryptophan, adenine, thymine, thiamine and pantothenate from complete chem. defined medium (CDM), caused biomass formation of K. vulgare to decrease to 1%, 21%, 16%, 1%, 26%, 57%, 73% and 24%, resp. Based on these results and FBA, a minimal chem. defined medium (MCDM) was developed that supported monoculture of K. vulgare (0.28 OD600) and 2-KLG production (3.59 g/L), which were similar to those in complete CDM or corn steep liquor powder (CSLP) medium. This study demonstrated the potential of using GSMM and FBA to characterize nutrient requirements, optimize CDM, and study interactions in co-culture.

Using a repeated social defeat stress (RSDS) model of depression, we showed that oral administration of FRP attenuates susceptibility to RSDS-mediated depression, supporting the further development of FRP as a novel therapeutic for the treatment of stress disorders and anxiety in humans.

Raspberry ketone (RK) is a major flavor compound in red raspberries, and it has been marketed as a popular weight-loss dietary supplement with high potential in accumulating in fatty tissues. However, challenges in extracting and characterizing RK and its associated phenolic compounds in fatty tissues persist due to the complex matrix effect. In this work, we reported a high-throughput sample preparation method for RK and 25 related phenolic compounds in white adipose tissues using an improved micro-scale QuEChERS (quick, efficient, cheap, easy, rugged and safe) approach with enhanced matrix removal (EMR)-lipid cleanup in 96-well plates, followed by UHPLC-QqQ-MS/MS anal. The absolute recovery was 73-105% at the extraction step, and achieved 71-96% at the EMR cleanup step. The EMR cleanup removed around 66% of total lipids in the acetonitrile extract as profiled by UHPLC-QTOF-MS/MS. The innovative introduction of a reversed-phase C18 sorbent into the extract significantly improved the analytes′ recovery during SpeedVac drying. The final accuracy achieved 80-120% for most analytes. Overall, this newly developed and validated method could serve as a powerful tool for analyzing RK and related phenolic compounds in fatty tissues.
