Cannabis and crash risk among older drivers

The purpose of this research was to reanalyze data collected from the National Highway Traffic Safety Administration’s Drug and Alcohol Crash Risk Study to investigate whether driving under the influence of cannabis (THC-positive) was associated with elevated crash risk for younger and older drivers. The data came from a case-control relative risk study collected from Virginia Beach, VA, over a 20-month period. Data collectors gathered driver information from the scene of vehicle crashes and, in some cases, from hospitals. Non-crash controls were sampled from the same locations, days, and times as crashes. Key data items included driver demographics and oral fluid and blood samples, which were assayed for licit and illicit drugs. We found no overall association between cannabis use and risk of crash involvement. However, when age and age(2) were allowed to interact with THC, significant interaction effects emerged. THC was associated with increased risk of crash involvement for older drivers. Difference between THC-positive and sober drivers emerged as significant at age 64. The research underscores the value of examining drugged driving in the context of driver age. Age-related declines in neurocognitive and psychomotor functioning were not measured but might be important in explaining the results.

Factors associated with the progression of gastric intestinal metaplasia: a multicenter, prospective cohort study

Background and study aims Gastric cancer (GC) is usually preceded by premalignant gastric lesions (GPLs) such as gastric intestinal metaplasia (GIM). Information on risk factors associated with neoplastic progression of GIM are scarce. This study aimed to identify predictors for progression of GIM in areas with low GC incidence. Patients and methods The Progression and Regression of Precancerous Gastric Lesions (PROREGAL) study includes patients with GPL. Patients underwent at least two upper endoscopies with random biopsy sampling. Progression of GIM means an increase in severity according to OLGIM (operative link on gastric intestinal metaplasia) during follow-up (FU). Family history and lifestyle factors were determined through questionnaires. Serum Helicobacter pylori infection, pepsinogens (PG), gastrin-17 and GC-associated single nucleotide polymorphisms (SNPs) were determined. Cox regression was performed for risk analysis and a chi-squared test for analysis of single nucleotide polymorphisms. Results Three hundred and eight patients (median age at inclusion 61 years, interquartile range (IQR: 17; male 48.4%; median FU 48 months, IQR: 24) were included. During FU, 116 patients (37.7%) showed progression of IM and six patients (1.9%) developed high-grade dysplasia or GC. The minor allele (C) on TLR4 (rs11536889) was inversely associated with progression of GIM (OR 0.6; 95%CI 0.4-1.0). Family history (HR 1.5; 95%CI 0.9-2.4) and smoking (HR 1.6; 95%CI 0.9-2.7) showed trends towards progression of GIM. Alcohol use, body mass index, history of H. pyloriinfection, and serological markers were not associated with progression. Conclusions Family history and smoking appear to be related to an increased risk of GIM progression in low GC incidence countries. TLR4(rs11536889) showed a significant inverse association, suggesting that genetic information may play a role in GIM progression.

Computational design of highly stable and soluble alcohol dehydrogenase for NADPH regeneration

Nicotinamide adenine dinucleotide phosphate (NADPH), as a well-known cofactor, is widely used in the most of enzymatic redox reactions, playing an important role in industrial catalysis. However, the absence of a comparable method for efficient NADP(+) to NADPH cofactor regeneration radically impairs efficient green chemical synthesis. Alcohol dehydrogenase (ADH) enzymes, allowing the in situ regeneration of the redox cofactor NADPH with high specific activity and easy by-product separation process, are provided with great industrial application potential and research attention. Accordingly, herein a NADP(+)-specific ADH from Clostridium beijerinckii was selected to be engineered for cofactor recycle, using an automated algorithm named Protein Repair One-stop Shop (PROSS). The mutant CbADH-6M (S24P/G182A/G196A/H222D/S250E/S254R) exhibited a favorable soluble and highly active expression with an activity of 46.3 U/mL, which was 16 times higher than the wild type (2.9 U/mL), and a more stable protein conformation with an enhanced thermal stability: Delta T-1/2(60min) = + 3.6 degrees C (temperature of 50% inactivation after incubation for 60 min). Furthermore, the activity of CbADH-6M was up-graded to 2401.8 U/mL by high cell density fermentation strategy using recombinant Escherichia coli, demonstrating its industrial potential. Finally, the superb efficiency for NADPH regeneration of the mutant enzyme was testified in the synthesis of some fine chiral aromatic alcohols coupling with another ADH from Lactobacillus kefir (LkADH).

A new measure of health motivation influencing food choices and its association with food intakes and nutritional biomarkers in European adolescents

Objective: To develop a scale to assess health motivation influencing food choices and to explore its performance in the associations with food intakes and nutritional biomarkers. Design: Psychometric study using cross-sectional self-report questionnaires and nutritional biomarkers. Setting: Multi-centre investigation conducted in ten European cities. Participants: 2954 adolescents who were included in the HELENA study and completed the Food Choices and Preferences (FCP) questionnaire. Results: Nineteen out of 124 items of the FCP questionnaire were in the same dimension. Sixteen presented adequate parameters for the Scale of evaluatiOn of Food choIcEs (SOFIE). The scores were positively associated with the intakes of cereals, dairy products, meats and eggs, and fish, as well as with blood concentrations of vitamin C, beta-carotene, n-3 fatty acids, cobalamin, holo-transcobalamin and folate; scores were negatively associated with the intake of alcohol. Conclusions: SOFIE can improve the assessment of motivation influencing food choices based on items with the best performance and is proposed as a new measure to health-related studies.

Health behaviour and risk behaviour of children and adolescents with and without disabilities and health-related impairment: Results of the representative German child and adolescent’s health survey (KiGGS Welle 2)

Background Although around 10% of children and adolescents in Germany live with disabilities and health-related restrictions, their health and risk behaviours have not yet been examined by disability and health-related restrictions. Research objective This study aimed at examining differences in health and risk behaviour of 3-17 year olds with disabilities and/or health-related restrictions and those without in Germany. Materials and methods Based on data from the German KiGGS wave 2 (2014-2017): of the Robert Koch-Institute, data from parents representing 3-10 year olds and from 11-17 year olds with/without disabilities/health-related restrictions were analysed. As dependent variables, physical activity, nutritional and oral health behaviour (n = 11,110) as well as alcohol and tobacco consumption (n = 3060) were used in uni-, bi- and multivariate analyses. Results Children and adolescents with disabilities/health-related restrictions showed more frequent physical inactivity, a higher rate of consumption of beverages containing sugar, a lower tooth brushing frequency and a lower alcohol consumption than their peers. Even after controlling for sociodemographic and -economic background characteristics, these associations for alcohol consumption, the consumption of beverages containing sugar and physical inactivity maintained. There were no differences in tobacco consumption among young people with and without disabilities/health-related restrictions. Conclusions Findings suggest that actions to strengthen health-promoting behaviour of children and adolescents with disabilities/health-related restrictions in the areas of physical inactivity and the consumption of beverages containing sugar are required. By applying behavioral and structural prevention, health behaviour should be target-group-specifically promoted in kindergarten and elementary school.

Hypothesis: The high incompatibility of bio-based materials such as protein and polysaccharides require a series of modifications to develop stable microstructures effectively. By modifying the density and charge of surface residues, pulsed electric fields processing can improve inter/intramolecular interactions, compatibility, and microstructure of bio-based nanostructured composites. Experiment: In this work, the impact of pulsed electric fields at a specific energy of 60-700 kJ/kg (electric field strength = 1.6 kV/cm) on self-assembly of zein-chitosan-poly(vinyl alcohol)-polyethylene glycol composite dispersion was investigated at pH 4.0, 5.7, and 6.8. Findings: Superior complex coacervated matrices were assembled at pH 4.0 and 5.7 before and after pulsed electric fields treatment at a specific energy of 390-410 kJ/kg. The compact and homogenous behaviour was attributable to pulsed electric fields-induced alteration of functional group interactions in a pH-dependent manner. Irrespective of the pH, very high electric field intensity caused excessive system perturbation leading to severe fragmentation and poor development of coacervates. The crucial insights from this study reveal that the self-assembly behaviour and integration of biopolymer-based sys-tems possessing different local charges can be enhanced by optimising pulsed electric fields processing parameters and the properties of the colloidal systems such as the pH. Highly flexible, self-healable and conductive poly(vinyl alcohol)/Ti3C2Tx MXene film and it’s application in capacitive deionization

Self-healing materials show great application potential in the field of flexible energy storage and wearable devices due to their special properties. Here, a self-healing material, application in Capacitive deionization for the first time is reported, which could autonomously self-healing damaged interfaces. We fabricated that hydrogel (MNH) composites incorporating MXene (Ti3C2Tx) by dynamic poly(vinylalcohol)-based network cross-linked by diol-borate ester bonding. The material fast restores its configuration, mechanical properties, and capacitive performances during after healing and shift different shape in the actual application environment transformation. The MNH electrode has good mechanical properties, self-healing efficiency is as high as 92%, the electrochemical healing properties is of 95.8%. The feasibility of its application in capacitive deionization was proved. The electrode performed a salt adsorption capacity of about 51 mg/g at a current density of 10 mA.g(-1) and a voltage window of 1 V and showed excellent cyclic stability. Our work expands the application scenarios of self-healing materials and provides a promising approach for the development of capacitive deionizing electrodes with good self-healing properties and long life.

As corroborated by isotopic labeling studies, a dual catalytic process is operative in which alkyne-to-allene isomerization is followed by allene-carbonyl reductive coupling via hydrogen auto-transfer. Crossover of ruthenium hydrides emanating from these two discrete catalytic events is observed. The utility of this method is illustrated by conversion of selected reaction products to the corresponding phenethylamines and the first total syntheses of the neolignan natural products (-)-crataegusanoids A-D.

Dihydromyricetin improves mitochondrial outcomes in the liver of alcohol-fed mice via the AMPK/Sirt-1/PGC-1 alpha signaling axis

Alcoholic liver disease (ALD), due to the multifactorial damage associated with alcohol (ethanol) consumption and metabolism, is one of the most prevalent liver diseases in the United States. The liver is the primary site of ethanol metabolism and is subsequently injured due to the production of reactive oxygen species (ROS), acetaldehyde, and metabolic stress. Building evidence suggests that dihydromyricetin (DHM), a bioactive flavonoid isolated from Hovenia dulcis, provides hepatoprotection by enhancing ethanol metabolism in the liver by maintaining hepatocellular bioenergetics, reductions of oxidative stress, and activating lipid oxidation pathways. The present study investigates the utility of DHM on hepatic mitochondrial biogenesis via activation of the AMP-activated protein kinase (AMPK)/Sirtuin (Sirt)-1/PPARG coactivator 1 (PGC)-1 alpha signaling pathway. We utilized a forced drinking ad libitum study that chronically fed 30% ethanol to male C57BL/61 mice over 8 weeks and induced ALD pathology. We found that chronic ethanol feeding resulted in the suppression of AMPK activation and cytoplasmic Sirt-1 and mitochondrial Sirt-3 expression, effects that were reversed with daily DHM administration (5 mg/kg; intraperitoneally [i.p.]). Chronic ethanol feeding also resulted in hepatic hyperacetylation of PGC-1 alpha, which was improved with DHM administration and its mediated increase of Sirt-1 activity. Furthermore, ethanol-fed mice were found to have increased expression of mitochondrial transcription factor A (TFAM), reduced mitochondrial content as assessed by mitochondrial DNA to nuclear DNA ratios, and significantly lower levels of hepatic ATP. In contrast, DHM administration significantly increased TFAM expression, hepatic ATP concentrations, and induced mitochondrial expression of respiratory complex III and V. In total, this work demonstrates a novel mechanism of DHM that improves hepatic bioenergetics, metabolic signaling, and mitochondrial viability, thus adding to the evidence supporting the use of DHM for treatment of ALD and other metabolic disorders. Folate, alcohol, ADH1B and ALDH2 and colorectal cancer risk

Objective: There is limited evidence on the interaction by alcohol dehydrogenase 2 (ADH1B) (rs1229984) and aldehyde dehydrogenase 2 (ALDH2) (rs671) regarding the associations of alcohol and a methyl diet (low folate and high alcohol intake) with cancer risk, partly because of rare polymorphisms in Western populations. Design: In a case-control study, we estimated the ORs and 95 % CIs to evaluate the associations of ADH1B and ALDH2 genotypes with colorectal cancer (CRC) and the joint association between methyl diets and ADH1B and ALDH2 polymorphisms with CRC risk using logistic regression models. Setting: A hospital-based case-control study. Participants: In total, 1001 CRC cases and 899 cancer-free controls admitted to two university hospitals. Results: We found that alcohol intake increased the risk of CRC; OR (95 % CI) was 2 center dot 02 (1 center dot 41, 2 center dot 87) for >= 60 g/d drinkers compared with non-drinkers (P (trend) < 0 center dot 001). The associations for two polymorphisms with CRC were not statistically significant. However, we found a potential interaction of ALDH2 with methyl diets and CRC. We observed a 9 center dot 08-fold (95 % CI 1 center dot 93, 42 center dot 60) higher risk of CRC for low-methyl diets compared with high-methyl diets among individuals with an A allele of ALDH2, but the association was not apparent among those with ALDH2 GG (P (interaction) = 0 center dot 02). Conclusions: Our data support the evidence that gene-methyl diet interactions may be involved in CRC risk in East Asian populations, showing that a low-methyl diet increased the risk of CRC among individuals with an A allele of ALDH2.
