Achyranthes bidentata Blume (Amaranthaceae) (ABR) and semen vaccariae (SV) are used commonly in the clin. treatment of erectile dysfunction in males with diabetes mellitus (DMED) to strengthen the kidney and promote blood circulation, and often achieve good curative effects. Explore mechanistic details of ABR + SV treatment against DMED. Prediction of key targets by network pharmacol. A rat model of DM was established by streptozotocin injection (55 mg/kg). Apomorphine (100μg/kg) was injected into rats to screen the DMED model. Group C (n = 6) and group M (n = 6) were gavaged with deionized water; group T (n = 6) was given Achyranthis bidentatae radix-semen vaccariae granule suspension (2.5 g/kg). It lasted 8 wk. Real-time reverse transcription-quant. polymerase chain reaction (RT-qPCR) and western blotting (WB) were used to measure the expression of tissue-related proteins and mRNA. The predicted key targets are albumin (ALB), caspase-3 (CASP3), vascular endothelial growth factor A (VEGFA), angiotensin-converting enzyme (ACE), and endothelial nitric oxide synthase (eNOS). Compared with the M group (0.52 ± 0.04; 0.50 ± 0.03; 0.49 ± 0.02; 0.23 ± 0.03), CASP3, VEGFA, and ACE protein expression reduced in the T group (0.39 ± 0.06; 0.34 ± 0.03; 0.39 ± 0.03), and eNOS protein expression increased (0.34 ± 0.03). ABR + SV can improve erectile function in DMED rats. This study provides a potential mechanism for the treatment of DMED with ABR + SV and can benefit from more patients.

The antimalarial drug quinine (QUN) has poor aqueous solubility and belongs to Biopharmaceutical Classification System (BCS) Class-II. We report 12 novel mol. salts of QUN with α,ω-aliphatic dicarboxylic acids and aromatic coformers. The high basicity of QUN and ΔpKa of ~5 make the complexes ionic, and most of them are hydrates. The solid forms showed enhanced aqueous solubility compared to the pristine QUN. The single-crystal and powder X-ray diffraction, thermal, and microscopy data provide structural, compositional, and stability profiles of the salts. The calculated Full Interaction Maps (FIMs) provide statistical insights into the salt formation and high probability of hydration in QUN. Though with prospective torsional freedom, QUN in most complexes adopts a unique conformation; this indicates that the structure class has a higher statistical probability and belongs to a relatively deep potential energy trough in the vast crystal landscape.

Diseases caused by Vibrio harveyi in marine organisms have been generally reported in aquaculture, but there has been no reported V. harveyi infection in fish cultivated in freshwater. Herein, we isolated 3 strains of V. harveyi from diseased American eels (Anguilla rostrata) cultivated in freshwater and subsequently confirmed the virulence of V. harveyi and the extracellular product (ECP). The three isolates were identified as V. harveyi based on phenotypic features and homol. of 16S rDNA and 3 house-keeping genes (HKGs). Exptl. infection revealed that V. harveyi and ECP were highly pathogenic to American eels, and the median LDs (LD50) were 1.67 x 103 cfu/g and 7.29μg/g body weight, resp. Moreover, our antibiotic susceptibility test showed that V. harveyi was susceptible to 16 of 20 antibiotics using the VITEK system of Merieux. Furthermore, liver and kidney samples were collected for paraffin sectioning, stained with haematoxylin-eosin (H & E) and Periodic Acid-Schiff (PAS). H & E staining results revealed hepatocyte atrophy and necrosis, hepatic intercellular hemorrhage and intrahepatic venous thrombosis, while the kidney was characterized by renal tubular epithelial cell swelling and renal interstitial haematopoietic cell loss. Interestingly, glycogen deposition was more obvious in the kidney compared with the liver after PAS staining. Cultivated eel infected by V. harveyii was reported for the first time, and the results of this study provide a valuable reference for further prevention and control of fish diseases caused by V. harveyi.

This study describes the biochem. and phylogenetic characteristics of a Gram-neg. strain, SNU WT1T, isolated from rainbow trout kidney. The 16S rRNA gene sequencing indicated that strain SNU WT1T was highly similar to Pseudomonas wadenswilerensis CCOS 864T and closely related to other Pseudomonas putida-related strains. Multilocus sequence anal. of concatenated partial gyrB, rpoB and rpoD sequences revealed that strain SNU WT1T was distinct from P. putida-related strains and formed a sep. clade. The average nucleotide identity and Genome-to-Genome Distance Calculator values were 90.19 and 41.7%with its closest relativeP. wadenswilerensis CCOS 864T ; however, it was phenotypically distinct from CCOS 864T with respect to arginine dihydrolase, glucose fermentation, aesculin hydrolysis and N-acetyl-glucosamine assimilation. The major polar lipid of the strain was phosphatidylethanolamine and the major quinone was Q-9. The genome of strain SNU WT1T had 5 685 196 bp with a G+C content of 61.83 mol%. We describe a novel species of genus Pseudomonas, for which the name Pseudomonas tructae has been proposed, with SNU WT1T (= KCTC 72265 = JCM 33436) as the type strain.

One challenge associated with the development of such materials is their fabrication method, which requires to be low-cost, scalable, and environmental-friendly. Herein, we developed a full “”green”” route to fabricate machine-washable conductive textiles by coating textiles with a novel crosslinked and conductive polymer composite coating, using single-walled carbon nanotubes (SWNTs) and bio-mass derived glucaric acid/chitosan (GA-chitosan) organic salt aqueous solution with dip-coating or spray-coating. The crosslinked SWNTs/GA-chitosan polyamide coatings exhibit a high elec. conductivity of up to 7.4 x 102 S/m and high water/organic solvents resistance. The conductive textiles can achieve an exceptional Joule heating performance driven by moderate voltage and exhibit a high electromagnetic interference shielding efficiency of approx. 30 dB at X-band under optimized formulation. The high adhesive energy between the polymer composite coatings and textile substrates enables the ultra-high durability and stability of textiles, confirmed by mech. deformation, rubbing, and washing tests. This simple and organic solvent-free processing method provides an environmentally friendly, cost-effective fabrication approach, holding great promise for large-scale production of multifunctional conductive wearable textiles for EMI shielding and/or personal heating applications.

Six Gram neg., motile bacteria were isolated from rainbow trout (Oncorhynchus mykiss). The 16S rRNA sequence similarity values grouped them in the Pseudomonas mandelii (strains P49, P50T, 154aT and P154b), Pseudomonas fluorescens (strain P115T) and Pseudomonas koreensis (strain P155T) phylogenetic subgroups in the genus Pseudomonas. The DNA G + C content ranged from 58.5 to 60 mol%. The strains were characterized phenotypically using API 20NE and Biolog GENIII tests, and chemotaxonomically by their whole-cell MALDI-TOF MS protein profiles and fatty acid contents. Multi-locus sequence anal. with four housekeeping gene sequences (rpoD, rpoB, gyrB and 16S rRNA) together with genome comparisons by average nucleotide identity and genome-to-genome distance calculations were performed. Results showed that the similarity values of these strains to known species type strains were lower than the thresholds established for species in the genus Pseudomonas. Based on these data, we concluded that strains P49, P50T, P115T, P154aT, P154b and P155T belonged to four novel species. The names proposed are: Pseudomonas piscium sp. nov. for strains P49 and P50T with P50T (= CECT 30175T = CCUG 74871T) as the type strain; Pseudomonas pisciculturae sp. nov. for strain P115T (CECT 30173T = CCUG 74873T); Pseudomonas mucoides sp. nov. for strains P154aT and P154b with P154aT (= CECT 30177T = CCUG 74874T) as the type strain; and Pseudomonas neuropathica sp. nov. for strain P155T (= CECT 30178T = CCUG 74875T).

Of the eight A/N-INV genes identified in the tomato genome, NI6 mRNA is present in all organs, but its expression was higher in sink tissues (mainly roots and fruits). A NI6-GFP fusion protein localized to the cytosol of mesophyll cells. Tomato NI6-silenced plants showed impaired growth phenotype, delayed flowering and a dramatic reduction in fruit set. Global gene expression and metabolite profile analyses of these plants revealed that NI6 is not only essential for sugar metabolism, but also plays a signaling role in stress adaptation. We also identified major hubs, whose expression patterns were greatly affected by NI6 silencing; these hubs were within the signaling cascade that coordinates carbohydrate metabolism with growth and development in tomato.
