Stereospecific recognition of metabolites plays a significant role in the detection of potential disease biomarkers thereby providing new insights in diagnosis and prognosis. D-Hdroxy/amino acids are recognized as potential biomarkers in several metabolic disorders. Despite continuous advances in metabolomics technologies, the simultaneous measurement of different classes of enantiomeric metabolites in a single anal. run remains challenging. Here, we develop a novel strategy for untargeted chiral metabolomics of hydroxy/amine groups (-OH/-NH2) containing metabolites, including all hydroxy acids (HAs) and amino acids (AAs), by chiral derivatization coupled with liquid chromatog.-high resolution tandem mass spectrometry (LC-HR-MS/MS). Diacetyl-tartaric anhydride (DATAN) was used for the simultaneous derivatization of-OH/-NH2 containing metabolites as well as the resulting diastereomers, and all the derivatized metabolites were resolved in a single anal. run. Data independent MS/MS acquisition (DIA) was applied to pos. identify DATAN-labeled metabolites based on reagent specific diagnostic fragment ions. We discriminated chiral from achiral metabolites based on the reversal of elution order of D and L isomers derivatized with the enantiomeric pair (±) of DATAN in an untargeted manner. Using the developed strategy, a library of 301 standards that consisted of 214 chiral and 87 achiral metabolites were separated and detected in a single anal. run. This approach was then applied to investigate the enantioselective metabolic profile of the bone marrow (BM) and peripheral blood (PB) plasma samples from patients with acute myeloid leukemia (AML) at diagnosis and following completion of the induction phase of chemotherapeutic treatment. The sensitivity and selectivity of the developed method enabled the detection of trace levels of the D-enantiomer of HAs and AAs in primary plasma patient samples. Several of these metabolites were significantly altered in response to chemotherapy. The developed LC-HR-MS method entails a valuable step forward in chiral metabolomics.

Conventionally, hexaric acids have been produced mainly by nitric acid oxidation of aldohexose, but full-scale commercialization has not been realized; there are many problems regarding yield, safety, environmental burden, etc. In recent years, therefore, improvements in hexaric acid production by nitric acid oxidation have been made, while new production methods, including biocatalytic methods, are actively being studied. Proper selection of rootstocks in tree fruit crops such as citrus is important for successful production Despite a large number of com. available rootstocks, studies have mostly been limited to basic horticultural observations. We used untargeted gas chromatog.-time of flight-mass spectrometry (GC-TOF MS) based metabolomics to understand the biochem. influence of rootstock on 2-yr-old field-grown ‘Cara Cara’ navel orange (Citrus sinensis L. Osbeck) and ‘Hirado Buntan’ pummelo (C. grandis L. Osbeck) trees grown on four rootstock cultivars with different genetic background. Five hundred unique metabolites were quantified in all trees, of which 147 were chem. identified. In navel orange trees, 48 root metabolites differed significantly in concentrations among rootstocks, compared with 29 metabolites in pummelo trees. In navel orange trees, raffinose, conduritol beta-epoxide, allantoic acid, myo-inositol, gamma-tocopherol, and beta gentiobiose were among the compounds that contributed most to this variation. In pummelo trees, hexitol, allantoic acid, glucoheptulose, tryptophan, gamma-tocopherol, glycerol-3-galactoside, and raffinose were among the most discriminating metabolites, but only allantoic acid passed significance criteria. Rootstock was also found to influence the quantities of 226 metabolites in leaves of the navel orange scion. Conduritol-beta-epoxide and myo-inositol were among the metabolites most influenced by rootstock. In contrast, the influence of rootstock on the pummelo scion was less prominent, with only six metabolites displaying significant differences. Our findings demonstrate that rootstock variety can influence the metabolic profile of the leaves in a grafted tree, but that the extent of the effect is also influenced by the scion. The majority of root metabolites that discriminated most between rootstocks did not display the same rootstock-specific discrimination in the leaves, suggesting tissue specificity or limited movement across the graft union.

Triphala is a traditional polyherbal formula used in Indian Ayurvedic and Chinese Tibetan medicine. A wide range of biol. activities have been attributed to Triphala, but the impact of various extraction methods on efficacy has not been determined The study aimed to evaluate Triphala extracts obtained by various methods for their hepatoprotective effects and mol. mechanisms in a mouse model of carbon tetrachloride (CCl4)-induced liver injury. HPLC fingerprinting was used to characterize the chem. characteristics of Triphala extracts obtained by (a) 0.5 h ultrasonication, (b) 2 h reflux, and (c) 4 h reflux. Hepatoprotective efficacy was evaluated in a mouse model of CCl4-induced liver damage. Serum levels of alanine transaminase (ALT) and aspartate aminotransferase (AST) were measured, as well as the liver antioxidant and inflammatory markers malondialdehyde superoxide dismutase glutathione peroxidase (GSH-Px), TNF-α, and IL-6. Gene and protein expression of Nrf-2 signaling components Nrf-2, heme oxygenase (HO-1), and NADPH Quinone oxidoreductase (NQO-1) in liver tissue were evaluated by real-time PCR and western blotting. Chem. anal. showed a clear difference in content between extracts produced by ultrasonic and reflux methods. The pharmacol. anal. showed that all three Triphala extracts reduced ALT, AST, MDA, TNF-α, and IL-6 levels and increased SOD and GSH-Px. Triphala extracts also induced transcript and protein expression of Nrf-2, HO-1, and NQO-1. Triphala extract prevents CCl4-induced acute liver injury. The ultrasonic extract of Triphala was most effective, suggesting that hepatoprotection may be related to the larger tannins via activation of Nrf-2 signaling.

There exists great potential in combining heterogeneous metallic nanoparticle active sites and enzymic active sites for sequentially catalyzed chem. transformations. This study provides an illustrative system where glucose oxidase and gold nanoparticles are successfully combined into a hybrid catalytic material. This multi-site catalyst is used to convert glucose to a more valuable saccharic acid product without the need for a strong oxidant such as H2O2, hypochlorite, or nitric acid to be externally added. We synthesize a hybrid material that is able to oxidize glucose to gluconic acid and H2O2 with glucose oxidase, and gold catalyzes the further oxidation to saccharic acid. The in situ generation of H2O2 provides a proximal, dilute oxidizing agent for further oxidation It is observed that higher pH promotes H2O2 decomposition and saccharic acid generation catalyzed by the gold nanoparticles. A 33% yield of saccharic acid in 90 min is observed without the addition of nitric acid, harsh oxidant, above ambient temperatures, or high oxygen pressures. The results demonstrate a cooperative reaction cascade system that opens new possibilities for metal nanoparticle-enzyme hybrid materials that can be applied to catalytic biomass conversion in mild reaction conditions.

The unbalanced distribution of carbon flux in microbial cell factories can lead to inefficient production and poor cell growth. Uncoupling cell growth and chem. synthesis can therefore improve microbial cell factory efficiency. Such uncoupling, which requires precise manipulation of carbon fluxes, can be achieved by up-regulating or down-regulating the expression of enzymes of various pathways. In this study, a dynamic turn-off switch (dTFS) and a dynamic turn-on switch (dTNS) were constructed using growth phase-dependent promoters and degrons. By combining the dTFS and dTNS, a bifunctional mol. switch that could orthogonally regulate two target proteins was introduced. This bifunctional mol. switch was used to uncouple cell growth from shikimic acid and D-glucaric acid synthesis, resulting in the production of 14.33 g/L shikimic acid and the highest reported productivity of D-glucaric acid (0.0325 g/L/h) in Escherichia coli MG1655. This proved that the bifunctional mol. switch could rewire carbon fluxes by controlling target protein abundance.

A novel Gram-neg., aerobic, rod-shaped, non-spore-forming bacterial strain, RYU5T, was isolated from a stool sample of an inpatient at a hospital in Okinawa, Japan. The optimal growth temperature of RYU5T was 30°C. Phylogenetic anal. based on the sequences of housekeeping genes, including the 16S rRNA, rpoB, rpoD and gyrB genes, showed that RYU5T was a member of the Pseudomonas putida group and was located close to Pseudomonas monteilii and P. putida. Whole-genome comparisons, using average nucleotide identity and digital DNA-DNA hybridization, confirmed that strain RYU5T should be classified as a novel species of Pseudomonas. Phenotypic characterization tests showed that utilization of d-mannose, d-serine, l-arabinose and d-fructose could distinguish this strain from other related species of the genus Pseudomonas. Based on genetic and phenotypic evidence, strain RYU5T should be classified as a novel species, for which the name Pseudomonas asiatica sp. nov. is proposed. The type strain is RYU5T (= DSM 107182T, = JCM 32716T), with a DNA G + C content of 62.25 mol%.

The present work showcases the versatility of nanogold systems supported on Zn-doped ceria when applied in two important environmental processes, the total CO oxidation, and the liquid phase oxidation of glucose to gluconic acid. In the CO oxidation the suitability of these materials is clearly demonstrated achieving full conversions even at sub-ambient conditions. Regarding the glucose oxidation our materials display high conversion values (always over 50%) and very importantly full or almost full selectivity toward gluconic acid-an added value platform chem. in the context of biomass upgrading routes. The key factors controlling the successful performance on both reactions are carefully discussed and compared to previous studies in literature. To our knowledge this is one of the very few works in catalysis by gold combining liquid and gas phase reactions and represents a step forward in the flexible behavior of nano gold catalysts.

Cosmopolitan bacteria are those that are found practically everywhere in the world. One of them is Bacillus subtilis, which can travel around the world through dust storms rising from various deserts. Upon landing, bacterial survival is determined by the ability to adjust to the heterogonous environments and bacteria isolated from extremely different environments, such as desert and riverbank soil, are expected to be less related due to the environmental pressure of each region. However, little is known about the influence of soil and habitat on B. subtilis evolution. Here, we show that desert and riverbank B. subtilis strains differ in genetic relatedness and physiol. traits, such as biofilm morphol. and utilization of carbon sources. Desert strains showed more diversity at the genetic level and were able to utilize more carbon sources than riverbank strains which were highly genetically conserved. Biofilm morphologies of desert and riverbank strains generally segregated and both groups formed different morphol. clusters despite the astonishing diversity observed among riverbank strains. We also show that relatedness of B. subtilis strains does not decrease with distance inside the same habitat, which, together with diversity data implies that the difference in environmental selection pressures plays a fundamental role in the evolution of this species.

The flower extracts of Saraca asoca were evaluated in their three different phenol. stages of flowering [bud (S1), mature (S2) and senescent (S3)] in terms of chem. composition and antioxidant activity. The GC/MS based fingerprinting led to identification of 85 metabolites, including 9 amino acids, 20 organic acids, 7 fatty acids, 20 sugar and sugar acids, 8 sugar alcs., 13 phenols and phenolic acids and 8 others compounds The three flowering stages showed prominent changes in their metabolite profile during the process of maturation of the flowering stages from bud to mature to senescence stages determined via GC/MS based metabolomics and chemometric approaches. The amounts and composition of metabolites in each stage showed statistically significant differences, which were reflected in their antioxidant capacities. The three phenol. stages showed antioxidant activities in a dose dependent manner, but the senescent stage showed highest superoxide radical scavenging activity (IC50 = 65.17 ± 2.647 mg/mL) and metal chelating effect (6.65 ± 0.331 mg/mL) in agreement with their high content of phenolic acids. These differences were strongly reproduced in the chemometric analyses (PCA, PLS-DA and s-PLS-DA), identifying the most distinctive features of the three flowering stages. This study might be beneficial to select the most potent flowering stage for incorporation in functional food.
