Linalool is a natural product present in fruits and aromatic plants with biol. activities. Researchers have reported that the inhalation of linalool exerts anti-inflammatory activities. In this study, we examined the therapeutic effects of linalool on airway inflammation and mucus overproduction in mice with allergic asthma. Oral administration of linalool significantly inhibited the levels of eosinophil numbers, Th2 cytokines and IgE (IgE) caused by ovalbumin (OVA) exposure. Linalool exerted preventive effects against the influx of inflammatory cells and mucus hypersecretion in the lung tissues. Linalool also dose-dependently decreased the levels of inducible nitric oxide synthase (iNOS) expression and protein kinase B (AKT) activation in the lung tissues. Linalool effectively downregulated the activation of mitogen-activated protein kinases (MAPKs) and nuclear factor-kappaB caused by OVA exposure. Furthermore, linalool exerted inhibitory effect on OVA-induced airway hyperresponsiveness (AHR). In the in vitro study, the increased secretion of MCP-1 was attenuated with linalool treatment in lipopolysaccharide (LPS)-stimulated H292 airway epithelial cells. In conclusion, linalool effectively exerts a protective role in OVA-induced airway inflammation and mucus hypersecretion, and its protective effects are closely related to the downregulation of inflammatory mediators and MAPKs/NF-kappaB signaling.

Iberian cured ham from purebred pigs fattened on grazing acorns is a highly appreciated product. There are several anal. methods to avoid its labeling fraud; however, these require opening the ham. The aim of this work is testing a non-destructive sampling method which does not spoil the phys. integrity of the ham. It consists of a puncture with a needle imitating a traditional olfactory system. After removing the needle impregnated with fat, its volatiles are analyzed with a Gas Chromatog. (GC) coupled to Ion Mobility Spectrometry (IMS). The potential of this methodol. was studied analyzing 156 Iberian hams from pigs under two different feeding regimes (acorns vs. feed) and from different breed (Iberian vs. Duroc crossed). Intensity of GC-IMS plot features was extracted and chemometric differentiation models were obtained; one for feeding regime and another for breed, providing validated classification rates of 91.7% and 100%, resp. In addition, 29 features used for construction of both models were tentatively identified using chem. standards The suitability of the method for quality control anal. was characterized by means of a precision study. As a conclusion, GC-IMS becomes a useful tool to guarantee dry-cured Iberian ham authenticity and detect labeling fraud.

Cymbopogon citratus essential oil has been shown to have antibacterial, antifungal, antiprotozoal, anti-carcinogenic, anti-inflammatory and antioxidant activities amongst others. This study was conducted to determine skin irritation, acute and subacute toxicity of C. citratus essential oil in mice and rabbits. The essential oil was analyzed using GC-MS. The essential oil at dose of 2000 mg/kg body weight was administered to mice for 21 consecutive days. The mice mortality, behavioral change, injury and other signs of illness were recorded once daily. Biochem. parameters were evaluated. Liver and kidney were taken after sacrifice for gross findings and histol. analyses. 10% ointment formulation of C. citratus oils was applied on the rabbit skin to determine skin irritation effects. The result revealed, the presence of citral (71.297%), myrcene (19.034%), 4, 5-epoxycarene (2.780%), linalool (1.713%), (S)-cis-verbenol (1.110%), linalool (1.713%), (S)-cis-verbenol (1.110%) and undecan-2-one (1.001%) in the C. citratus essential oil. There was no significant difference (p > 0.05) in the body weights, gross abnormalities of the organs and biochem. parameters compared to the control. No histopathol. changes were detected in the organs tested. 10% ointment formulation of C. citratus oils did not cause skin irritation. Anal. of results leads to the conclusion that Ethiopian C. citratus essential oil may be considered as relatively safe and non-toxic.

This study was done to determine the antifungal activities of essential oil (EO) vapors of 97 plants against Penicillium corylophilum and to test combinations of EO vapors for synergistic antifungal effects. Among 97 com. available EOs extracted from plant parts, garlic, cinnamon bark, may chang (mountain pepper), citronella, thyme thymol, oregano, spearmint, and thyme linalool EO vapors exhibited relatively strong antifungal activities. The minimal inhibitory concentrations of these EO vapors were 0.0390-0.6250 μL/mL. A combination of cinnamon bark, citronella, and may chang EO vapors, as well as a combination of cinnamon bark and citronella EO vapors, showed synergistic inhibitory activities to P. corylophilum on a laboratory medium. A combination of cinnamon bark, citronella, and may chang EO vapors had synergistic activity in inhibiting growth of P. corylophilium on beef jerky. Observations reported here provide basic information valuable when developing strategies to inhibit the growth of P. corylophilum and possibly other moderately xerophilic molds on intermediate-moisture foods.

A review. The incidence of fungal infections has been steadily increasing in recent years. Systemic mycoses are characterized by the highest mortality. At the same time, the frequency of infections caused by drug-resistant strains and new pathogens e.g., Candida auris increases. An alternative to medicines may be essential oils, which can have a broad antimicrobial spectrum. Rich in the essential oils are plants from the Lamiaceae family. In this review are presented antifungal activities of essential oils from 72 Lamiaceae plants. More than half of these have good activity (min. inhibitory concentrations (MICs) < 1000μg/mL) against fungi. The best activity (MICs < 100) have essential oils from some species of the genera Clinopodium, Lavandula, Mentha, Thymbra, and Thymus. In some cases were observed significant discrepancies between different studies. In the review are also shown the most important compounds of described essential oils. To the chem. components most commonly found as the main ingredients include β-caryophyllene (41 plants), linalool (27 plants), limonene (26), β-pinene (25), 1,8-cineole (22), carvacrol (21), α-pinene (21), p-cymene (20), γ-terpinene (20), and thymol (20). Mitochondrial dysfunction and inflammation contribute to the initiation and development of several brain pathol. conditions, including Alzheimer’s disease and cerebral ischemia. Linalool is an aromatic plant-derived monoterpene alc. with reported anti-inflammatory, and anti-oxidant properties. We investigated the role of linalool on glutamate-induced mitochondrial oxidative stress in immortalized neuronal HT-22 cells. Glutamate induced oxidative stress in neuronal cells, as detected by real-time cell impedance measurements, MTT assay, and anal. of Annexin V/PI. Administration of linalool 100μM reduced cell death mediated by glutamate. Staining of glutamate-stimulated mitochondria by MitoTracker revealed improved morphol. in the presence of linalool. Furthermore, we demonstrated a potential neuroprotective effect of linalool in conditions of oxidative stress by a reduction of mitochondrial ROS and mitochondrial calcium levels, and by preserving mitochondrial membrane potential. Experiments using both high-resolution respirometry and Seahorse Extracellular flux analyzer showed that linalool was able to promote an increase in uncoupled respiration that could contribute to its neuroprotective capacity. Linalool protection was validated using organotypic hippocampal slices as ex vivo model with NMDA as a stimulus to induce excitotoxity cell damage. These results demonstrate that linalool is protective in an in vitro model of glutamate-induced oxidative stress and in an ex-vivo model for excitotoxity, proposing linalool as a potential therapeutic agent against neurodegenerative brain diseases where oxidative stress contributes to the pathol. of the disease.

A novel synergistic formulation (TML) based on the combination of thymol (T), Me cinnamate (M), and linalool (L) has been prepared using the mixture design assay. Nanoencapsulation of developed formulation TML (Ne-TML) was prepared and characterised by SEM, XRD and FTIR. The Ne-TML was assessed for its antifungal and anti-aflatoxin B1 potential in vitro and in the food systems (Pennisetum glaucum L.), and also examined its effects on organoleptic properties. The Ne-TML cause complete inhibition of growth and AFB1 production at 0.3 μl/mL and 0.2 μl/mL. In-situ results revealed that Ne-TML exhibited maximum protection from fungal (75.40%) and aflatoxin B1 contamination (100%) at 0.3 μl/mL during six months of storage. The speculated antifungal mode of action of Ne-TML was related to the decrease in ergosterol content, membrane ions leakage, impairment in carbon-source utilization, mitochondrial functioning, anti-oxidative defense system (SOD, CAT, and GR) and Ver-1 gene of aflatoxin B1 biosynthesis.

Alzheimer’s disease (AD) is a neurodegenerative disorder leading to cognitive deficits and cognitive decline. Since no cure or preventing therapy is currently available to counteract AD, natural-derived compounds are investigated to find new potential neuroprotective agents for its treatment. In the present study, we tested the neuroprotective effect of lavender and coriander essential oils (EOs) and their main active constituent linalool, against the neurotoxicity elicited by Aβ1-42 oligomers, a key mol. factor in the neurodegeneration of AD. Importantly, our findings on neuronally differentiated PC12 cells exposed to Aβ1-42 oligomers are in accordance with previous in vivo studies reporting the neuroprotective potential of lavender and coriander EOs and linalool. We found that lavender and coriander EOs at the concentration of 10μg/mL as well as linalool at the same concentration were able to improve viability and to reduce nuclear morphol. abnormalities in cells treated with Aβ1-42 oligomers for 24 h. Lavender and coriander EOs and linalool also showed to counteract the increase of intracellular reactive oxygen species production and the activation of the pro-apoptotic enzyme caspase-3 induced by Aβ1-42 oligomers. Our findings provide further evidence that these EOs and their main constituent linalool could be natural agents of therapeutic interest against Aβ1-42-induced neurotoxicity.

To better understand wine aroma persistence, the nasal cavity of nine volunteers was monitored by Proton Transfer Reaction-Time of Flight-Mass Spectrometry (PTR-ToF-MS) after they rinsed their mouths with three rose wines (one control and the same wine supplemented with two tannin extracts) during four minutes. Wines were aromatized with a mixture of five target aroma compounds Results showed that wine aroma persistence was highly compound-dependent: while esters disappeared very fast, other compounds such as linalool remained in the oral cavity for longer times after wine expectoration. A low effect of tannins (at 50 mg/L) on nasal cavity parameters was observed, with the exception for the compound Et decanoate that was significantly higher released in the presence of tannins. Strong interindividual differences on aroma persistence were also found.
