Cornstalk was subjected to methanolysis in the presence of NaOH at 220-320 °C to afford soluble portions (SPs) 1-5 (SP1-SP5) and an inextractable portion (IEP). The maximum total yield (ca. 51%) of SPs was acquired at 300 °C with the same mass of NaOH and cornstalk. Under the same conditions, SP1 has the highest yield, followed by SP5 and SP2. The relatively volatile and less polar species in the resulting SPs and IEP were identified with a gas chromatograph/mass spectrometer (GC/MS). The polar species in SP1, SP2, and SP5 were further analyzed with a neg.-ion electrospray ionization Fourier transform ion cyclotron resonance mass spectrometer (FTICRMS). The anal. with GC/MS shows that phenolic compounds and alcs. are the dominant group components in SP1 and SP2, resp., while the predominant compounds in esterified SP3-SP5 and IEP are esters. After that, the solubilities of these 3 compounds were tested and compared in supercritical CO2 (scCO2) within the temperature range of (313-353) K and in the pressure range of (9.3-19.0) MPa. The influence of molar mass and end group on the solubility of compound was also studied. In addition, the exptl. solubility data were correlated according to Bartle, Chrastil and JCF model, and good consistencies were obtained. The average absolute relative deviation (AARD) of the measured values were (5.83, 8.98, 8.77)% when using Bartle’s method, (10.95, 7.53, 7.55)% for Chrastil’s method, and (6.89, 4.10, 4.19)% for the JCF method, resp. It is obvious that the JCF model was proven to be the best model because it had more adjustable parameters of temperature and pressure. Finally, the partial molar volumes V2 for each compound in the supercritical phase were estimated in accordance with Kumar and Johnston’s theory. In most muscle biopsies from humans with myopathies, sarcolemma-localized neuronal nitric oxide synthase (nNOS) is either reduced or not detected, particularly in dystrophin-deficient Duchenne muscular dystrophy (DMD). Abnormal NO signaling at the sarcolemmal level is integrally involved in the pathogenesis and accounts, at least in part, for the muscle weakness of DMD. Dystrophic muscle fibers exhibit an increased susceptibility to contraction-induced membrane damage. Muscle relaxants function to prevent muscle wasting by decreasing nerve impulses and reducing calcium influx that regulates tensing or tightening of muscle fibers. We have recently developed a new class of nitric esters that combines the pharmacol. functions of NO and muscle relaxation. Here, we report the synthesis and properties of the nitric ester (MMPN) of 2-methyl-2-n-propyl-1,3-propanediol (MPP) and its effect in mdx dystrophic mice, a murine model of DMD. MMPN produced significant improvements in biochem., pathol., and functional phenotypes in the mouse model. The endurance of exercise was extended by 47% in time to exhaustion and 84% in running distance. Serum CK level was decreased by 30%. Addnl., MMPN decreased intracellular free calcium concentration without causing skeletal muscle weakness. No hepatic or renal toxicities were observed during the study. Our investigations unveil a potential new treatment for muscular diseases.
