Kabadi, Shruti V.;Fisher, Jeffrey W.;Doerge, Daniel R.;Mehta, Darshan;Aungst, Jason;Rice, Penelope research published 《 Characterizing biopersistence potential of the metabolite 5:3 fluorotelomer carboxylic acid after repeated oral exposure to the 6:2 fluorotelomer alcohol》, the research content is summarized as follows. Our previous report on pharmacokinetic (PK) evaluation of 6:2 fluorotelomer alc. (6:2 FTOH) examined the biopersistence potential of its metabolites based on data published from single inhalation and occupational 6:2 FTOH exposure studies. We calculated internal exposure estimates of three key metabolites of 6:2 FTOH, of which 5:3 fluorotelomer carboxylic acid (5:3 acid) had the highest internal exposure and the slowest clearance. No oral repeated 6:2 FTOH exposure data were available at the time to fully characterize the biopersistence potential of the metabolite 5:3 acid. We recently received addnl. data on 6:2 FTOH and 5:3 acid, which included a 90-day toxicokinetic study report on repeated oral 6:2 FTOH exposure to rats. We reviewed the study and analyzed the reported 5:3 acid concentrations in plasma, liver, and fat using one-compartment PK modeling and calculated elimination rate constants (kel), elimination half-lives (t1/2) and times to steady state (tss) of 5:3 acid at three 6:2 FTOH doses. Our results showed that tss of 5:3 acid in plasma and evaluated tissues were approx. close to 1 yr, such that the majority of highest values were observed at the lowest 6:2 FTOH dose, indicating its association with the biopersistence of 6:2 FTOH. The results of our PK anal. are the first to characterize biopersistence potential of the 5:3 acid after repeated oral exposure to the parent compound 6:2 FTOH based on steady state PK parameters, and therefore, may have an impact on future study designs when conducting toxicity assays for such compounds

Kassotis, Christopher D.;Hoffman, Kate;Phillips, Allison L.;Zhang, Sharon;Cooper, Ellen M.;Webster, Thomas F.;Stapleton, Heather M. research published 《 Characterization of adipogenic, PPARγ, and TRβ activities in house dust extracts and their associations with organic contaminants》, the research content is summarized as follows. In this study, we sought to expand our previous research on associations between bioactivities in dust and associated organic contaminants. Dust samples were collected from central NC homes (n = 188), solvent extracted, and split into two fractions, one for anal. using three different bioassays (nuclear receptor activation/inhibition and adipocyte development) and one for mass spectrometry (targeted measurement of 124 organic contaminants, including flame retardants, polychlorinated biphenyls, perfluoroalkyl substances, pesticides, phthalates, and polycyclic aromatic hydrocarbons). Approx. 80% of dust extracts exhibited significant adipogenic activity at concentrations that are comparable to estimated exposure for children and adults (e.g. ∼20μg/well dust) via either triglyceride accumulation (65%) and/or pre-adipocyte proliferation (50%). Approx. 76% of samples antagonized thyroid receptor beta (TRβ), and 21% activated peroxisome proliferator activated receptor gamma (PPARγ). Triglyceride accumulation was significantly correlated with TRβ antagonism. Sixty-five contaminants were detected in at least 75% of samples; of these, 26 were correlated with adipogenic activity and ten with TRβ antagonism. Regression models were used to evaluate associations of individual contaminants with adipogenic and TRβ bioactivities, and many individual contaminants were significantly associated An exploratory g-computation model was used to evaluate the effect of mixtures Contaminant mixtures were pos. associated with triglyceride accumulation, and the magnitude of effect was larger than for any individually measured chem. For each quartile increase in mixture exposure, triglyceride accumulation increased by 212% (RR = 3.12 and 95% confidence interval: 1.58, 6.17). These results suggest that complex mixtures of chems. present in house dust may induce adipogenic activity in vitro at environmental concentrations and warrants further research.

Here, templateless electropolymerization is employed to produce nanotubular structures from various thieno[3,4-b]thiophene-based monomers that differ in substituent structure and size, as well as the linker connecting the thieno[3,4-b]thiophene core and substituent. The formation of densely packed vertically aligned are obtained from monomers with a pyrene substituent and when a significant amount of water (CH2Cl2 + H2O) is included in the solvent. The geometrical parameters of the nanotubes are highly dependent on the electopolymn. method. A significant amount of air is trapped within the structure of the densely packed open nanotubes obtained with Qs = 100 mC cm-2 causing an increase in water contact angle (θw) up to 82.6° (intermediate state between the Wenzel and the Cassie-Baxter state), and θw can become even more hydrophobic by further modifying the deposition method or the electrolyte.

Jackson, Thomas W.;Scheibly, Chris M.;Polera, M. E.;Belcher, Scott M. research published 《 Rapid Characterization of Human Serum Albumin Binding for Per- and Polyfluoroalkyl Substances Using Differential Scanning Fluorimetry》, the research content is summarized as follows. Per- and polyfluoroalkyl substances (PFAS) are a diverse class of synthetic chems. that accumulate in the environment. Many proteins, including the primary human serum transport protein albumin (HSA), bind PFAS. The predictive power of physiol. based pharmacokinetic modeling approaches is currently limited by a lack of exptl. data defining albumin-binding properties for most PFAS. A novel thermal denaturation assay was optimized to evaluate changes in the thermal stability of HSA in the presence of increasing concentrations of known ligands and a structurally diverse set of PFAS. Assay performance was initially evaluated for fatty acids and HSA-binding drugs ibuprofen and warfarin. Concentration-response relationships were determined and dissociation constants (Kd) for each compound were calculated using regression anal. of the dose-dependent changes in HSA melting temperature Estimated Kd values for HSA binding of octanoic acid, decanoic acid, hexadecenoic acid, ibuprofen, and warfarin agreed with established values. The binding affinities for 24 PFAS that included perfluoroalkyl carboxylic acids (C4-C12), perfluoroalkyl sulfonic acids (C4-C8), mono- and polyether perfluoroalkyl ether acids, and polyfluoroalkyl fluorotelomer substances were determined These results demonstrate the utility of this differential scanning fluorimetry assay as a rapid high-throughput approach for determining the relative protein-binding properties and identification of chem. structures involved in binding for large numbers of structurally diverse PFAS.

Jin, Hangbiao;Mao, Lingling;Xie, Jiahui;Zhao, Meirong;Bai, Xiaoxia;Wen, Jie;Shen, Tao;Wu, Pengfei research published 《 Poly- and perfluoroalkyl substance concentrations in human breast milk and their associations with postnatal infant growth》, the research content is summarized as follows. Perfluoroalkyl carboxylates (PFCAs) and perfluoroalkyl sulfonates are widespread in human breast milk. However, the occurrence of chlorinated polyfluorinated ether sulfonates (Cl-PFESAs) and fluorotelomer alcs. (FTOHs) in breast milk and their effects on postnatal growth of infants through breast milk consumption are still not well known. This study characterized the occurrence of 16 poly- and perfluoroalkyl substances (PFASs) in breast milk from 174 women in Hangzhou, China and investigated the association between lactation exposure to these PFASs through breast milk consumption and the postnatal growth of infants. Our results showed that perfluorooctanoate (mean 87 pg/mL) was the predominant PFAS in breast milk, followed by perfluorohexanoate (41 pg/mL), 6:2 Cl-PFESA (28 pg/mL), and perfluorooctane sulfonate (25 pg/mL). The occurrence and levels of Cl-PFESAs in Chinese breast milk were firstly reported in the current study. The 8:2 and 10:2 FTOH were detected in half of breast milk samples, with the mean concentration of 9.0 pg/mL and 10 pg/mL, resp. Breast milk concentrations of C8-C10 PFCAs and 6:2 Cl-PFESA were neg. correlated with infant’s length gain rate. Exposed to higher levels of 8:2 FTOH were correlated with decreased infant’s weight gain rate. Daily intakes of PFASs via the consumption of breast milk were calculated for infants. Overall, this study firstly demonstrated that lactation exposure to C8-C10 PFCAs, 8:2 FTOH, and 6:2 Cl-PFESA through breast milk consumption may affect the postnatal growth of infants.

Hu, Xinhao;Li, Zhan;Yang, Zhen;Zhu, Fang;Zhao, Weifeng;Duan, Gaigai;Li, Yiwen research published 《 Fabrication of Functional Polycatechol Nanoparticles》, the research content is summarized as follows. While low dimensional (1D and 2D) polycatechol materials were widely described for a range of biomedical and surface engineering applications, few examples were explored to focus on the construction of functional polycatechol nanoparticles. Herein we report the facile fabrication of a series of polycatechol nanoparticles via a general and robust strategy based on one-step oxidation reaction. IO3-induced catechol redox chem. could facilitate the precise size control of the resulting nanoparticles and also allow the successful transferring and amplifying of microscopic monomer function into macroscopic polycatechol material property. The ease, facileness and controllability of such one-step fabrication process could highly promote the development of polycatechol nanomaterials for various applications.

Hua, Xia;Luo, Jianbo;Zhao, Zhen;Wang, Qi;Sun, Hongwen research published 《 Neutral polyfluoroalkyl and perfluoroalkyl substances in surface water and sediment from the Haihe River and Dagu Drainage Canal deserve more attention》, the research content is summarized as follows. Neutral polyfluoroalkyl and perfluoroalkyl substances (nPFASs) were detected in the surface water and sediment from the Haihe River (HR) and Dagu Drainage Canal (DDC), Tianjin, China. N-Me perfluorooctane sulfonamide ethanol (MeFOSE) and N-Et perfluorooctane sulfonamide ethanol (EtFOSE) were the predominant nPFASs in surface water and sediment, which was different from the composition in air. The concentrations of ΣnPFASs in water from the HR (1.88-8.21 ng/L) were lower than those from the DDC (3.72-11.32 ng/L). Concentrations of ΣnPFASs were higher in the middle of the HR in the Dongli District due to industrial activity, whereas at lower reaches of the DDC, high ΣnPFAS concentrations might be due to effluent from wastewater treatment plants (WWTPs). The detection frequency in sediment (13.5%) was less than that in water (83%). The concentrations in sediment from the DDC (below limit of qualification (LOQ) to 5.58 ng/g) were higher than those from the HR (below LOQ to 2.46 ng/g). The distribution coefficient (log KD) between water and sediment was calculated, and they were highly related to the compound structures. The contribution of nPFASs to nPFASs+PFAAs was up to 52% in sediment in the DDC, suggesting the importance of nPFASs in aquatic systems.

M. research published 《 Heterotelechelic homopolymers mimicking high χ – ultralow N block copolymers with sub-2 nm domain size》, the research content is summarized as follows. Three fluorinated, hydrophobic initiators have been utilized for the synthesis of low mol. mass fluoro-poly(acrylic acid) heterotelechelic homopolymers to mimic high chi (χ)-low N diblock copolymers with ultrafine domains of sub-2 nm length scale. Polymers were obtained by a simple photoinduced copper(II)-mediated reversible-deactivation radical polymerization (Cu-RDRP) affording low mol. mass (< 3 kDa) and low dispersity (D = 1.04-1.21) homopolymers. Heating/cooling ramps were performed on bulk samples (ca. 250μm thick) to obtain thermodynamically stable nanomorpologies of lamellar (LAM) or hexagonally packed cylinders (HEX), as deduced by small-angle X-ray scattering (SAXS). Construction of the exptl. phase diagram alongside a detailed theor. model demonstrated typical rod-coil block copolymer phase behavior for these fluoro-poly(acrylic acid) homopolymers, where the fluorinated initiator-derived segment acts as a rod and the poly(acrylic acid) as a coil. This work reveals that these telechelic homopolymers mimic high χ-ultralow N diblock copolymers and enables reproducible targeting of nanomorphologies with incredibly small, tunable domain size.

Herkert, Nicholas J.;Kassotis, Christopher D.;Zhang, Sharon;Han, Yuling;Pulikkal, Vivek Francis;Sun, Mei;Ferguson, P. Lee;Stapleton, Heather M. research published 《 Characterization of Per- and Polyfluorinated Alkyl Substances Present in Commercial Anti-fog Products and Their In Vitro Adipogenic Activity》, the research content is summarized as follows. Anti-fog sprays and solutions are used on eyeglasses to minimize the condensation of water vapor, particularly while wearing a mask. Given their water-repellent properties, we sought to characterize polyfluorinated alkyl substance (PFAS) compounds in four anti-fog spray products, five anti-fog cloth products, and two com. fluorosurfactant formulations suspected to be used in preparing anti-fog products. Fluorotelomer alcs. (FTOHs) and fluorotelomer ethoxylates (FTEOs) were detected in all products and formulations. While 6:2 FTOH and the 6:2 FTEO polymeric series were predominant, one anti-fog cloth and one formulation contained 8:2, 10:2, 12:2, 14:2, and 16:2 FTOH and FTEO polymeric series. PFAS concentrations varied in samples and were detected at levels up to 25,000μg/mL in anti-fog sprays and 185,000μg (g cloth)-1 in anti-fog cloth products. The total organic fluorine (TOF) measurements of anti-fog products ranged from 190 to 20,700μg/mL in sprays and 44,200 to 131,500μg (g cloth)-1 in cloths. Quantified FTOHs and FTEOs accounted for 1-99% of TOF mass. In addition, all the four anti-fog sprays and both com. formulations exhibited significant cytotoxicity and adipogenic activity (either triglyceride accumulation and/or preadipocyte proliferation) in murine 3T3-L1 cells. Results suggest that FTEOs are a significant contributor to the adipogenic activity exhibited by the anti-fog sprays. Altogether, these results suggest that FTEOs are present in com. products at toxicol. relevant levels, and more research is needed to fully understand the health risks from using these PFAS-containing products.

In general, the hydroxyl group makes alcohols polar. Those groups can form hydrogen bonds to one another and to most other compounds. 647-42-7, formula is C8H5F13O, Owing to the presence of the polar OH alcohols are more water-soluble than simple hydrocarbons. Methanol, ethanol, and propanol are miscible in water. Butanol, with a four-carbon chain, is moderately soluble. HPLC of Formula: 647-42-7

Highly repellent surfaces offer many interesting applications as oil-repellent textiles and self-cleaning coatings, making these surfaces highly sought-after. Engineering these repellent surfaces requires the combination of a low surface energy material with special surface geometries with multiple levels of hierarchy in roughness. Yet, the current approaches for the fabrication of these surfaces have many drawbacks (complex fabrication techniques, fragile nature of the surface features with low abrasion resistances) that preventing their wider application. Here, we present a study of a new double-hierarchical Fluoropor which allows creating a bulk-porous fluorinated polymer foam with two individually tunable pore hierarchies. We combined our previous structuring approach for the micro /nanoscale (addition of an adaptable porogen mixture to the monomer) with sacrificial templating utilizing spherical polystyrene particles to obtain a second micropore hierarchy, resulting in a novel double-hierarchical material. We found that a micro-/nanoporosity with a larger pore structure in combination with a higher volumetric loading of structuring particles was beneficial to increase the repellency. We found the highest repellency for double-hierarchical Fluoropor with a bulk-porosity with 259 nm median pore diameter structured with 50 vol% of 40 μm sized microspheres. These superhydrophobic surface showed high static contact angles for ethylene glycol of 164° with a roll-off angle of 13° and a static contact angle for n-hexadecane of 145°. The material was covalently coated onto tech. substrates (glass, copper, steel) and demonstrated good self-clean abilities for inorganic as well for organic contaminants. Abrasion test with 1000 cycles showed the repellency was not affected and persisted.
