Amines are incorporated into various membranes to improve their CO2 separation performance. With amine-containing polymeric membranes, gas transport properties are often enhanced under humidity, where CO2 migrates through the membranes in the form of bicarbonate ions. Piperazine (Pz) and its derivatives are known to catalyze the conversion of CO2 to bicarbonate ions and have been used in liquid amine scrubbing technol. Piperazines were immobilized in poly(vinyl alc.) (PVA), and the resulting polymeric membranes showed high CO2 separation performance over H2 and CH4. The gas transport properties were dependent on the chem. structure of the amines. In particular, 3-(1-piperazinyl)-1,2-propanediol (PzPD)-containing polymeric membranes gave excellent CO2 separation performance, and the CO2 permeability and CO2 selectivity over CH4 were 1060 Barrer and 370, resp., at 50°C and 90% relative humidity with a transmembrane CO2 pressure of 11 kPa. The interaction between PzPD and CO2 was quant. studied by inverse-gate decoupling 13C NMR spectroscopy. CO2 interacted with the secondary amino group on the Pz ring to form a carbamate, which was readily hydrolyzed to produce bicarbonate ions. The hydroxyl group on the C2 carbon of PzPD facilitated the interaction between CO2 and the amine through hydrogen bonding, resulting in enhanced diffusivity of CO2 in the membranes.

Although mollugin, the main ingredient of the oriental medicinal herb Rubia cordifolia, has considerable anti-inflammatory effects, it has poor aqueous solubility as well as poor metabolic and plasma stability. To overcome these shortfalls, various mollugin derivatives have been synthesized and evaluated for their ability to inhibit U937 monocyte cell adhesion to HT-29 colonic epithelial cells in TNF-α or IL-6-induced models of colon inflammation. The 2-(4-morpholinyl)-Et ester of CF3-substituted mollugin (compound 15c) showed good water solubility, improved metabolic and plasma stability, and greater inhibitory activity than mesalazine in both the TNF-α and IL-6-induced colonic epithelial cell adhesion assays, suggesting that 15c is a potential anti-inflammatory agent.

Recently, emerging evidence demonstrates that the PI3K signaling transduction pathway is linked to the pathol. of IPF. In this work, we rationally designed a new series of 4-methylquinazoline derivatives as highly potent PI3K inhibitors that significantly suppress the phosphorylation of the main PI3K downstream effectors and displays marked antiproliferative activity in mouse MLg2908 lung fibroblasts. In a bleomycin-induced mouse pulmonary fibrosis model, 5d from the series improved mouse lung function and slowed the progression of pulmonary fibrosis. Overall, this work promises a therapeutic potential for PI3K inhibitors to treat IPF.

This study unveils the in vitro and in vivo antibiofilm potential of 2,6-Di-tert-butyl-4-methylphenol (DTBMP) from Chroococcus turgidus against Vibrio spp. In the preliminary study, cell free culture supernatant (CFCS) of C. turgidus inhibited the violacein production in biomarker strain Chromobacterium violaceum and its mutant strain CV026 in a dose dependent manner. The effective biofilm inhibitory concentration (BIC) of pure compound DTBMP from C. turgidus was identified as 250μg/mL concentration in tested Vibrio species. Furthermore, DTBMP proved to effectively inhibit the bioluminescence production in V. harveyi and other biofilm related virulence traits such as exopolysaccharides (EPS) production, hydrophobicity index, swimming and swarming motility at its BIC concentration in three major pathogenic vibrios: V. harveyi, V. parahaemolyticus and V. vulnificus. The antibiofilm potential of DTBMP was validated through light, confocal laser scanning and scanning electron microscopic analyses. In addition, the non-bactericidal effect of DTBMP was determined through growth curve and 2,3-bis (2-methyloxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide (XTT) assay. Real-time PCR studies revealed the down-regulation of master quorum sensing (QS) regulator genes of V. harveyi such as luxR, luxS, luxP, luxQ and luxO on treatment with DTBMP. In vivo results confirmed that DTBMP augmented the survival rate of Litopenaeus vannamei larvae up to 75, 88 and 66% upon infection with V. harveyi, V. parahaemolyticus and V. vulnificus, resp. The results of this study ascertain the promising effects of DTBMP as an antibiofilm agent, which could be pos. explored to treat biofilm-associated vibrios infections in aquaculture.

None of them showed significant human recombinant MAO inhibition, but compounds I [R = 4-(4-isopropylpiperazin-1-yl)butyl, 5-(4-isopropylpiperazin-1-yl)pentyl] and compound II [R1 = 4-(4-isopropylpiperazin-1-yl)butyl] displayed promising human recombinant acetylcholinesterase (hrAChE) and butyrylcholinesterase (hrBuChE) inhibition. In particular, compound I [R = 4-(4-isopropylpiperazin-1-yl)butyl] was found to be a potent and quite selective non-competitive inhibitor of hrAChE (IC50 = 0.29μM), with Ki value in nanomolar range (79 nM). Pertinent docking anal. confirmed this result, suggesting that this ligand was an interesting hit for further investigation.

In the present study, a new class of triazino-caffeine derivatives with good antimicrobial activities is reported. Eight new triazino-caffeine derivatives were synthesized via Mitsunobu reaction, followed by intramol. cyclization. Their chem. structures were established on the basis of elemental anal., IR, 1H NMR, mass spectral data as well as through an alternate synthetic route. The compounds were also screened for their antimicrobial activities, among them, compounds I (R = Me, H) showed good inhibitory potentials, indicating the importance of the morpholine moiety in enhancing antimicrobial activity. Therefore, it is concluded that the triazino-caffeine derivatives developed, show encouraging potentials in their medicinal applications.

The replacement of one chem. motif with another that is broadly similar is a common method in medicinal chem. to modulate the phys. and biol. properties of a mol. (i.e., bioisosterism). In recent years, bioisosteres such as cubane and bicyclo[1.1.1]pentane (BCP) have been used as highly effective Ph mimics. Herein, we show the successful incorporation of a range of Ph bioisosteres during the open-source optimization of an antimalarial series. Cubane (19) and closo-carborane (23) analogs exhibited improved in vitro potency against Plasmodium falciparum compared to the parent Ph compound; however, these changes resulted in a reduction in metabolic stability; unusually, enzyme-mediated oxidation was found to take place on the cubane core. A BCP analog (22) was found to be equipotent to its parent Ph compound and showed significantly improved metabolic properties. While these results demonstrate the utility of these atypical bioisosteres when used in a medicinal chem. program, the search to find a suitable bioisostere may well require the preparation of many candidates, in our case, 32 compounds

This study describes a new method for simultaneous identification of uropathogens in the case of polybacterial urinary tract infections. The method utilizes recently developed preparative isoelec. focusing (IEF) in cellulose-based separation medium with a subsequent anal. by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). Preparative IEF was successfully used for both purification and separation of bacteria, Escherichia coli (pI 4.6) and Staphylococcus aureus (pI 3.4), in urine samples. The focused zones of bacteria, localized by the positions of focused colored pI markers, were easily collected from the separation media after the IEF anal. and then unambiguously identified by MALDI-TOF MS. The proposed method enables the identification of bacteria in urine specimens when the concentration of individual bacteria is ≥104 cells mL-1. Another benefit is the viability of bacteria extracted from the collected fractions after preparative IEF.

Shown herein is that Co(OAc)2 and Mn(OAc)3·2H2O act as powerful catalysts for oxidative p-quinone functionalization with a collection of O, N, and S nucleophiles, using oxygen as the terminal oxidant. Preliminary mechanistic observations and the first synthesis of the cytotoxic natural product strongylophorine-26, I, is presented.
