Inflammatory cytokine interleukin-6 (IL-6) has emerged as a potential therapeutic target. Our studies demonstrated that metabolism of flavanol rich cocoa preparation (FRP) led to the accumulation of select phenolic acids that may contribute to its anti-inflammatory activity. Using a repeated social defeat stress (RSDS) model of depression, we showed that oral administration of FRP attenuates susceptibility to RSDS-mediated depression, supporting the further development of FRP as a novel therapeutic for the treatment of stress disorders and anxiety in humans.

Raspberry ketone (RK) is a major flavor compound in red raspberries, and it has been marketed as a popular weight-loss dietary supplement with high potential in accumulating in fatty tissues. However, challenges in extracting and characterizing RK and its associated phenolic compounds in fatty tissues persist due to the complex matrix effect. In this work, we reported a high-throughput sample preparation method for RK and 25 related phenolic compounds in white adipose tissues using an improved micro-scale QuEChERS (quick, efficient, cheap, easy, rugged and safe) approach with enhanced matrix removal (EMR)-lipid cleanup in 96-well plates, followed by UHPLC-QqQ-MS/MS anal. The absolute recovery was 73-105% at the extraction step, and achieved 71-96% at the EMR cleanup step. The EMR cleanup removed around 66% of total lipids in the acetonitrile extract as profiled by UHPLC-QTOF-MS/MS. The innovative introduction of a reversed-phase C18 sorbent into the extract significantly improved the analytes′ recovery during SpeedVac drying. The final accuracy achieved 80-120% for most analytes. Overall, this newly developed and validated method could serve as a powerful tool for analyzing RK and related phenolic compounds in fatty tissues.

Tuo-Min-Ding-Chuan decoction (TMDCT) is a Traditional Chinese Medicine (TCM) formula consisting of twelve herbs that can relieve the symptoms and treat allergic asthma. Yet, the underlying mechanism of action is still unclear. In this study, we investigated the effect of TMDCT in regulating Treg/Th17 cells immune balance and explored potential metabolic and gut biomarkers associated with Treg and Th17 cells in eosinophilic asthma mice treated by TMDCT. We found that TMDCT increases Treg cells percentage and decreases Th17 cells percentage in the ovalbumin (OVA) -induced eosinophilic asthma mice model. Furthermore, Imidazoleacetic acid, DL-glutamine, L-pyroglutamic acid, 2-deoxy-D-glucose were preliminary identified as biomarkers in plasma metabolites treated by TMDCT, meanwhile genus Desulfovibrio, genus Butyricimonas and genus Prevotella 9 were preliminary identified as gut microbiota biomarkers after TMDCT treatment. These results provide an exptl. foundation for the treatment of allergic asthma with Chinese herbal compounds

The fruit of Rosa Roxburghii Tratt (RRT) is edible and has been considered as a medicinal fruit with a lipid-lowering effect, but the mechanism is still unclear. In this study, we integrated the results of 16S rRNA gene sequence and fecal metabolome to explore the potential mechanisms of RRTJ treatment in hyperlipidemia. The result suggests the useful effects of RRT juice (RRTJ) in hyperlipidemic rats. The 12 wk of RRT juice (RRTJ) treatment decreased serum lipids, fat accumulation, body weight and liver weight RRTJ treatment reversed the HFD-induced gut dysbiosis by lowering Dorea, Coprobacillus abundance and enriching Bifidobacterium, Roseburia abundance. Also, we conducted a targeted metabolic profile anal. of 160 metabolites in feces and 19 feces metabolites have significant changes. Correlation analyzes suggest that the reductions in serum lipids by RRTJ are associated with the gut microbial community (Dorea, Coprobacillus and Roseburia, Bifidobacterium) and their associated metabolites (palmitoleic acid, succinic acid, propionic acid, Glyceric acid). In conclusion, the 12 wk of treatment with RRT juice (RRTJ) can regulate dyslipidemia by improving intestinal microbiota disorders and related metabolites disorders.

Two of the most important Mexican plant-foods are tomato (Solanum lycopersicum L.) and husk tomato (Physalis ixocarpa Brot.). The study shows new knowledge of the in vitro intestinal digestion and fermentation of T and HT final compounds with biol. potential which should be evaluated in further studies.

The elevated intestinal oxygen in certain unhealthy conditions (e.g., mucosa injury) enhances the expansion of aerobic/facultative anaerobic bacteria (mainly Proteobacteria) in gut microbiota (GM) and is strongly linked to various diseases. The alteration of GM, influenced by oxygen, may affect the bioavailability of dietary polyphenols. In vitro digestion, dialysis and fermentation of phenolic blueberry extract (BE) were performed here using the GM of mice under different oxygen conditions. Oxygen delayed the degradation of the main phenolic components, including quercetin, kaempferol and their rutinose-conjugates, in BE during in vitro fermentation In addition, the metabolites of BE were also influenced by oxygen. It is still uncertain whether the consumption of Lachnum sp. polysaccharides (LEP) alleviates colorectal cancer (CRC) through the gut microbiota. In this study, our efforts are focused on the influence of LEP on CRC, intestinal barrier and inflammation, and fecal microbiota and the metabolites, in azoxymethane (AOM)/dextran sodium sulfate (DSS)-induced CRC mice. Results showed that LEP inhibited CRC mouse colon shortening and weight loss, decreased tumor incidence, restored intestinal barrier integrity, and reduced excessive inflammation. LEP consumption significantly altered microbiota overall structure and community, with reduced pernicious bacteria (such as Parabacteroides, Escherichia_Shigella, Desulfovibrio and Helicobacter), and increased beneficial bacterium (such as Alistipes, Alloprevotella and Ruminiclostridium). Fecal-metabolome profile indicated that a total of 43 metabolites were clearly changed, with 10 down-regulated and 33 up-regulated metabolites. In addition, short-chain fatty acids (SCFAs), including acetic acid, propionic acid and n-butyric acid, were significantly increased after LEP administration. Moreover, a strong correlation between the fluctuant gut microbiota and metabolites was found. These findings provided not only deeper insights into the responsibility of LEP for CRC alleviation, and but also the potential of LEP as a promising candidate for CRC prevention and treatment.

Stress urinary incontinence (SUI), which is defined as an involuntary loss of urine upon phys. exertion coughing, sneezing or laughing, has a significant neg. impact on the quality of life of many women. Multi-center, large-scale and randomized clin. trials have illustrated that non-invasive electroacupuncture is an effective treatment for SUI, but its therapeutic mechanism in treating SUI remains unknown. Here, gas chromatog.-mass spectrometry based serum metabolomics was performed to reveal metabolic profiles and diagnostic biomarkers from recruitment of 25 patients and 25 healthy women before and after electroacupuncture. We identified 10 differentially abundant metabolites, including butantriol, 3,4-dihydroxybutanoic acid, succinic acid, 1-deoxypentitol, psicose, citric acid, 3-hydroxybutyric acid, hydracrylic acid, 3-hydroxyphenylacetic acid and D-mannitol, from patients between before and after electroacupuncture. The electroacupuncture therapy altered propanoate metabolism, butanoate metabolism and the tricarboxylic acid cycle. A panel of 8 biomarkers (butantriol, 3,4-dihydroxybutanoic acid, succinic acid, 1-deoxypentitol, psicose, citric acid, 3-hydroxybutyric acid and hydracrylic acid) was evaluated to determine the effect of electroacupuncture on SUI and differentiated well between before and after treatment. The area under the receiver operating characteristic curve was 0.962. The sensitivity, specificity and coincidence rate were 92%, 92% and 96%, resp., at a 95% confidence interval ranging from 0.9053 to 1. Furthermore, the levels of these diagnostic biomarkers were not significantly altered in healthy subjects after sham electroacupuncture. It was indicated that an 8-biomarker panel might be constructed for the therapeutic evaluation of electroacupuncture treatment for SUI.

Key message: Sweet cherry tree branches have important food and medicinal value. The ultrasound and microwave-assisted extraction method is more efficient with higher yield than conventional extraction methods (heat-reflux, Soxhlet, etc.). Plant byproducts are known as sources of natural bioactive compounds The objective of this study was to rationally use sweet cherry tree branches (SCTB) discarded during pruning. Ultrasonication and microwaves are considered green techniques, and an ultrasound and microwave-assisted extraction (UMAE) method was established to obtain extracts from SCTB by response surface methodol. A math. model was established using the Box-Behnken design, and the effects of various factors and their interactions were analyzed as well. Taking the yield (weight/weight) as the objective, the optimal process conditions for UMAE of SCTB were 56 mL·g-1 liquid-solid ratio, 34 min extraction time, and 40-50 mesh particle size. The yield of SCTB extracts was 5.02%, which was close to the theor. prediction. The optimized extraction process can obtain a higher yield than that of conventional extraction methods. The chem. composition of the extracts was identified by HPLC-MS/MS, and 400 metabolites, including carboxylic acids and derivatives (29%), fatty acyls (19%), organooxygen compounds (15%), flavonoids (12%), benzene and substituted derivatives (12%), phenols (8%), and imidazopyrimidines (5%), were annotated and classified. L-arginine and mannitol were the main chem. components of the SCTB extracts, suggesting their potential uses in the food and medical industries.
