The feasibility of manufacturing yoghurts with three prebiotics, i.e., fructooligosaccharides (FOS), galactooligosaccharides (GOS) or lactulose added at two concentrations (2 and 4%, w/v) was investigated. Physicochem. and microbial characterization was conducted on the effect of prebiotics on acidification, viability of Streptococcus thermophilus and Lactobacillus delbrueckii ssp. bulgaricus, lactose and prebiotic consumption and production of organic acids during fermentation (up to 6 h) and cold storage (for 28 d). GOS and FOS remained unaltered throughout fermentation and cold storage, while the viability of starter culture was similar to that observed in the control yoghurt. Yoghurts manufactured with 4% lactulose showed a significant decrease of lactulose content associated with a lower decrease of lactose relative to the control. This effect was associated with a significant increase (2.2 log10 cfu mL-1) in L. delbrueckii ssp. bulgaricus and a moderate decrease (1.0 log10 cfu mL-1) in S. thermophilus at the end of cold storage.

The microwave-assisted extraction of pectin from sweet lemon peel (SLP) was optimized using Box-Behnken design. The highest pectin yield (25.31%) was observed under optimal condition (microwave power of 700 W, irradiation time of 3 min and pH of 1.5). The physicochem., structural and some bioactivity of the SLP pectin isolated at optimum condition was evaluated. The SLP pectin was rich in galacturonic acid and galactose (87.2 mol%), high in mol. weight (615.836 kDa) and low in degree of esterification (1.2-35.1%). Furthermore, the SLP pectin was composed of 55.7% linear region (homogalacturonan) and 42.2% hairy region (rhamnogalacturonan-I). Also, the FT-IR and H-NMR results confirm the major presence of low methylated galacturonic acid rich structure in the isolated samples. In addition, SLP pectin showed good emulsifying and antioxidant properties. A pseudoplastic flow behavior was observed for SLP pectin solution at higher concentrations (1% w/v <). These results represent an inexpensive source for pectin extraction with high pectin yield and good properties. The aim of the present study was to evaluate the effect of water-soluble polysaccharides from Cuminum cyminum to induce inflammatory response in immune cells and understand their underlying mechanisms. Weight average mol. weight (Mw) of polysaccharides varied between 191.4-512.2 × 103 g/mol. Polysaccharides induced RAW264.7 cells to release nitric oxide and express TNF-α, IL-1β, IL-6 and IL-12 inflammatory cytokines. Polysaccharides activated NK-92 cells to produce TNF-α, IFN-γ, perforin, granzyme B, NKG2D and FasL. Activations of RAW264.7 and NK-92 cells were through NF-κB and MAPKs signal pathways indicated by the presence of phosphorylated NF-κB, ERK, JNK and p38 proteins. The polysaccharide structure was mainly constituted of â†?)-Galp-(1â†? â†?)-Galp-(1â†? â†?)-Arap-(1â†?and â†?)-Arap-(1â†?glycosidic linkages. Overall results suggested that polysaccharides from C. cyminum possessing lower MW and greater expanded conformation more effectively stimulate RAW264.7 and NK-92 cells and thus could be considered for further studies on their biomedical applications.

Exerting control over the glycan moieties of antibody therapeutics is highly desirable from a product safety and batch-to-batch consistency perspective. Strategies to improve antibody productivity may compromise quality, while interventions for improving glycoform distribution can adversely affect cell growth and productivity. Process design therefore needs to consider the trade-off between preserving cellular health and productivity while enhancing antibody quality. In this work, we present a modeling platform that quantifies the impact of glycosylation precursor feeding – specifically that of galactose and uridine – on cellular growth, metabolism as well as antibody productivity and glycoform distribution. The platform has been parameterized using an initial training data set yielding an accuracy of ±5% with respect to glycoform distribution. It was then used to design an optimized feeding strategy that enhances the final concentration of galactosylated antibody in the supernatant by over 90% compared with the control without compromising the integral of viable cell d. or final antibody titer. This work supports the implementation of Quality by Design towards higher-performing bioprocesses.

The structural characteristics and biol. activity of polysaccharides were influenced by different extraction methods. In this study, polysaccharides from mulberry fruits (Murus alba L., which were pre-treated with superfine grinding process) (MFP) were exacted using hot-water extraction (HWE), enzyme-assisted hot water extraction (EAHE), ultrasonic-assisted hot water extraction (UAHE), and high-speed shear homogenization-assisted hot water extraction (HSEHE). The extraction yield, structure, rheol. properties and antioxidant activities of MFPs were investigated. MFP extracted using the HSEHE method have the highest extraction yields than other extraction methods. The smaller particle size of mulberry powder was found to improve the extraction yields. The MFPs were obtained by the combination between different extraction methods and superfine grinding pretreatment (through 100 mesh sieve) (MFP-HWE100, MFP-EAHE100, MFP-UAHE100, MFP-HSEHE100) showed the same levels of monosaccharide compositions and glycosyl linkages, However, these methods can produce MFP with different monosaccharide proportions, branching degree, different mol. weight, particle size and microstructure. MFP-HSEHE100 achieved the lowest mol. weight and particle size, which exhibited better thixotropy and antioxidant activities than other MFPs. This study identified that HSEHE was the most suitable extraction method for MFP.

The aim of this study was to investigate the effect of sulfated modification on the in vitro antioxidant and hypoglycemic activities of Sargassum pallidum polysaccharides (SPP). Three sulfated derivatives (S-SPP1-4, S-SPP1-6 and S-SPP1-8) were prepared using chlorosulfonic acid-pyridine method under different reaction conditions. Physicochem. characterization indicated that sulfated modification had successfully occurred. The degrees of substitution (DS) of S-SPP1-4, S-SPP1-6 and S-SPP1-8 were determined to be ranging from 0.85 to 1.19. Sulfated modification resulted in some changes in chem. component, mol. weight and monosaccharide composition of SPP. Among these sulfated polysaccharides, S-SPP1-4 exhibited the best DPPH and ABTS radical scavenging activities, while S-SPP1-8 exhibited the best α-glucosidase inhibitory activity and promoting-effect on glucose consumption in insulin resistant (IR)-HepG2 cells. Furthermore, all the sulfated derivatives exhibited better hypoglycemic ability than native SPP. These results suggest that appropriately sulfated modification could enhance the antioxidant and hypoglycemic activities of S. pallidum polysaccharides, which might be used as an alternative derivative of natural antioxidant and hypoglycemic agent.

The influences of different extraction methods on the yields, physicochem. properties, antioxidant activities and antiproliferative abilities of polysaccharides from Morinda citrifolia L. (Noni) were investigated and compared. Three extraction methods were applied to extract Noni polysaccharides (NP), which include hot water extraction (HWE), pulsed elec. field-assisted extraction (PEFAE) and ultrasonic-assisted extraction (UAE). The extraction yields, sugar contents, uronic acid contents, mol. weights, monosaccharide proportions, antioxidant activities and antiproliferative abilities of three NP samples were significantly different. However, their preliminary structural characteristics and monosaccharide types were similar. More specifically, UAE-NP had the highest extraction yield, the smallest mol. weight and the best antioxidant activities. Especially, UAE-NP showed excellent antiproliferative abilities. Compared with HWE-NP and PEFAE-NP, the IC50 value of UAE-NP on HepG2 cells reduced by 45.45% and 33.14%, resp. The highest antioxidant activities of the UAE-NP might be attributed to its smaller mol. weight, lower Gal content and higher Fuc content. The strongest antiproliferative effects of the UAE-NP might be related to the low mol. weight Overall, UAE could be a potential technique for the extraction of high-quality NP due to its high yield, high efficiency and notable bioactivities.

Schisandra sphenanthera and Schisandra chinensis are widely consumed either as food or for medicinal purposes. Nevertheless, no detailed comparative assessments of their physicochem. properties and biol. activity have been reported. In this paper, using hot-water extraction, alc. precipitation, and deproteinization, we obtained polysaccharidic extracts from Schisandra sphenanthera and Schisandra chinensis (denoted as SSP and SCP, resp.) and investigated their antioxidant and immunol. activities. The extracts were different from each other with regard to sugar, protein, and uronic acid contents. Both extracts were mainly composed of arabinose, glucose, and galactose, but their contents varied greatly; SSP had more galacturonic acid. Compared with SCP, SSP had stronger free radical scavenging ability, protective effects on biomols., cellular antioxidant activity, owing to its higher protein (35.35 ± 1.73%) and uronic acid (12.81 ± 1.15%) contents. With respect to cell viability, neutral red phagocytosis, NO production, and acid phosphatase activity, SCP had stronger effects than SSP; this was largely due to its high levels of mannose, galactose, arabinose, and glucose. These results provide evidence to support the use Schisandra-derived polysaccharides for several purposes, including clin., agricultural, and industrial applications.

Microwave-assisted aqueous two-phase extraction (MAATPE) was applied to simultaneous extraction of diverse polysaccharides from Lentinus edodes with aqueous two-phase extraction system (ATPS) of ethanol/ammonium sulfate as the biphasic extractant. Key factors in MAATPE process were optimized by response surface methodol., and the optimum conditions were concluded as follows: the ATPS composition of ethanol concentration 26.0% (weight/weight) and (NH4)2SO4 concentration 19.58% (weight/weight), extraction temperature 78.7°C, extraction time 19.55 min and solid-to-solvent ratio 1:50. The extraction yields of the polysaccharides from the top phase (PTP) and the bottom phase (PBP) were 2.12 ± 0.21% and 11.16 ± 0.28%, resp. By means of FT-IR, HPGPC and HPLC anal., PTP and PBP were different from each other in chem. structure, their average mol. weight (MW) were resp. 119,533 Da and 3906 Da. Also, the mechanism of MAATPE process was explored by composition anal. and SEM. Both polysaccharides exhibited stronger antioxidant activities, EC50 values of scavenging DPPH, hydroxyl and superoxide radicals for PBP achieved 580 ± 3, 83.1 ± 3.6 and 91.1 ± 0.5 ug/mL as against 560 ± 4, 76.3 ± 2.2 and 89.3 ± 2.4 ug/mL of PTP, resp. Compared with conventional methods using a mono-phase solvent, MAATPE can not only greatly improve extraction efficiency, but also selectively extract diverse polysaccharides.

The antioxidant activities of polysaccharide from balsam pear were studied. It was determined by gel permeation chromatog. that the mol. weight distribution of purified polysaccharide was concentrated near 33582 Da. It indicated that the monosaccharide components were rhamnose, galacturonic acid, galactose, xylose and arabinose. Fourier transform IR spectroscopy, NMR and Congo red experiments showed that there are C1, C2, C3, C5 links, and stable β-triple helix conformation in aqueous solution It was found that polysaccharide had good scavenging effect on free radicals. So, balsam pear polysaccharide should be a potential antioxidant.
