Potato peel was used for the extraction of three types of polysaccharides (PW, PAL, and PAC) using water, alk., and acid treatments, resp. The structure of the PP polysaccharides was examined by means of Fourier transform IR spectroscopy (FT-IR) anal. and Gas chromatog.-mass spectrometry (GC-MS). The results suggest that the extracted polysaccharides constituted essentially three functional groups: C=O, C-H, and O-H. The polysaccharides were comprised of low proportions of proteins, 17-23% uronic acids, and approx. 70% carbohydrates. PAL, PW, and PAC with mol. weights of 2.25 × 103, 2.18 × 103, and 1.92 × 103 kDa, resp., were composed of rhamnose, xylose, mannose, arabinose, glucose, and galactose. Functional properties (solubility, oil holding capacity, foaming, and emulsion properties) of these polysaccharides were evaluated. Among three, PAL showed the highest fat-binding capacity which was 7.50 g/g with the solubility of 95.06%. The three polysaccharides possessed appreciable in vitro anti-oxidant (scavenging DPPH and ABTS+ radicals, chelating ferrous ions, and reducing power) potential. PAL exhibited the strongest reducing power, scavenging activity on DPPH radicals and chelating capability on ferrous ions. PP polysaccharides can be used as promising natural antioxidants in food, pharmaceutical, and cosmetic preparations

We assessed the effect on lower urinary tract symptoms (LUTS) of a supplement containing cranberry, D-mannose and anti-inflammatory mols. in postmenopausal women undergoing surgery for cystocele. Forty postmenopausal women were randomized 1:1 to an active group receiving the nutritional supplement twice a day for 2 wk starting from surgery, or to a control group receiving surgery only. Primary outcomes were the effectiveness in the postoperative LUTS and urinary tract infections (UTI). LUTS were investigated by a validated questionnaire (ICIQ-FLUTS) at baseline and at week 4. Secondary outcomes were the safety and tolerability of the supplement and other perioperative outcomes. No significant differences were found in perioperative outcomes and in incidence of UTI. After surgery, women treated with the supplement experienced significantly better scores on the filling domain of the questionnaire. A non-significant decrease in voiding scores was also found. No adverse events were detected. The use of an oral supplement containing cranberry, D-mannose and anti-inflammatory mols. decreases the perception of LUTS in postmenopausal women after anterior colporraphy. Our data suggest that perioperative use of nutritional supplements may be useful in the management of postoperative LUTS.

Extraction of pectin from pomelo peel was performed using water, acid and alk. solutions with a microwave power of 1100 W for 2 min, and yield and structural/chem. characteristics of the extracted pectins were investigated. Extraction of pectin with water gave the lowest yield (6.5%, dry weight basis). The water-extracted pectin contained the highest amount of neutral sugar (29.5%), while equivalent mol. weight (MWeq) and degree of methylation (DM) were 43 kDa and 29.7%, resp. Extraction of pectin with HCl solutions at concentrations of 25, 50, 100 and 200 mM resulted in a significant increase of pectin yields (12.1-20.5%) and pectins with high amount of galacturonic acid (83.3-85.6%). The MWeq and DM of the acid-extracted pectins ranged from 171 to 368 kDa and 53.9-82.5%, resp. Compared to the acid, extraction with NaOH solutions at the same concentrations provided higher yields (13.9-24.2%), smaller size (142-187 kDa) and lower DM (<1.7%) pectins, while amount of galacturonic acid was comparable. The highest yield of extraction was found at a concentration of 50 mM for both HCl and NaOH solutions Extraction of pomelo peel with 50 mM NaOH solution by reduction of the microwave power to 550 W and extending the extraction time to 15 min resulted in an increase of pectin yield (29.2%) and pectin with smaller size (76 kDa). Through hot water extraction, protein removal and chromatog. purification, DWPP-Is was found to be the major polysaccharide present in the stem of D. wardianum. The Mn and Mw of DWPP-Is were 29.0 kDa and 98.6 kDa, resp. Furthermore, mannose and glucose were found to be the most abundant monosaccharides in DWPP-Is. Their backbones consist of (1 → 4)-β-D-Glcp and O-acetylated (1 → 4)-β-D-Manp, which are similar to the structures of other anti-tumor Dendrobium polysaccharides. The inhibition rate of DWPP-Is treatment on SPC-A-1 cells (2 mg/mL, 72 h) reached 56.0%. Intragastric administration of DWPP-Is on A549 tumor-bearing KM mice (10 mg/mL, 0.2 mL) exhibited similar inhibition ratios to that of erlotinib hydrochloride (2 mg/mL). Moreover, the highest inhibition was observed in P-CK treatment combined with DWPP-Is, reaching an inhibition rate of 23.4%. These results suggest that DWPP-Is has the potential to be a functional agent for lung cancer prevention.

Two heteropolysaccharides, BHP-1 and BHP-2, were separated and purified from the edible bulbs of Lilium davidii var. unicolor Cotton by anion-exchange and gel-permeation chromatog. The primary structural characteristics of BHP-1 and BHP-2 were investigated by Congo red test, X-ray diffractometry (XRD) anal., IR, GC and NMR. The results showed that the weight-average mol. weight (MW) and number-average mol. weight (Mn) of BHP-1 and BHP-2 were nearly 1.93 × 105 g/moL, 8.86 × 104 g/moL and 3.52 × 104 g/moL, 2.95 × 104 g/moL, resp. Glucose and mannose comprised BHP-1 in a relative molar ratio of 5.9:2.0, and BHP-2 was consisted of glucose, galactose, mannose and arabinose with approx. molar ratios of 8.3:1.5:1.0:1.1. Their backbone mainly contained α-(1 → 4)-linked D-glucopyranosyl. In addition, the C-6 of the glucose and C-2 of mannose residue of BHP-1 and BHP-2 were substituted. The two polysaccharides were semi-crystalline substance with multi-branching structure and triple helix conformation. Bioassays showed that BHP-1 and BHP-2 had effective antioxidant activities evaluated by DPPH, scavenging superoxide radical and OH radical in a dose-dependent manner. The two heteropolysaccharides also displayed distinct and selective antibacterial activity against B. cereus, P. putida, M. luteus and K. pneumonia with inhibition zones of 13 mm, 16 mm, 17 mm, 23 mm and 12 mm, 17 mm, 19 mm, 20 mm, resp. These findings provided a reference for potential applications of the polysaccharides from L. davidii as natural antioxidants and antibacterial agents in food and pharmaceuticals.

Numerous broadly neutralizing antibodies (bnAbs) have been identified that target the glycans of the HIV-1 envelope spike. Neutralization breadth is notable given that glycan processing can be substantially influenced by the presence or absence of neighboring glycans. Here, using a stabilized recombinant envelope trimer, we investigate the degree to which mutations in the glycan network surrounding an epitope impact the fine glycan processing of antibody targets. Using cryo-electron microscopy and site-specific glycan anal., we reveal the importance of glycans in the formation of the 2G12 bnAb epitope and show that the epitope is only subtly impacted by variations in the glycan network. In contrast, we show that the PG9 and PG16 glycan-based epitopes at the trimer apex are dependent on the presence of the highly conserved surrounding glycans. Glycan networks underpin the conservation of bnAb epitopes and are an important parameter in immunogen design.

The structural characterization and antioxidant activity of four Auricularia polysaccharides (A. cornea (ACP), A. auricula (AAP), A. polytricha (APP) and M.fungus (MFP)) were studied in this paper. The results shown: polysaccharides of four Auricularia were mainly composed of mannose and galactose, all polysaccharides contained uronic acid and pyran ring structure with spectroscopy and NMR anal. There was a significant difference in the total antioxidant capacity and APP was significantly higher than the other polysaccharides. The ability of APP to scavenge DPPH radicals and hydroxyl radicals was significantly higher than that of other polysaccharides, resp. The mol. weight was significantly pos. correlated with DPPH radicals, superoxide anion radicals and hydroxyl radicals. Total antioxidant capacity was significantly neg. correlated with fucose and galactose. The result indicated that fucose and galactose jointly determine total antioxidant capacity. The polysaccharide from four Auricularia had good oxidation resistance and could be used as natural antioxidants.

Longan pulp polysaccharides were extracted with hot water (LP-H), superfine grinding (LP-S) and superfine grinding-assisted enzymic treatments (LP-SE). The yields, physicochem. properties and prebiotic activities of polysaccharides were investigated. Compared with LP-H and LP-S, the yield, sugar content, solubility, arabinose and mannose percentage of LP-SE increased while its apparent viscosity, particle size, Mw and glucose percentage declined. Three LPs contained similar glycosidic linkage of →3)-α-L-Araf-(1→, →3,6)-β-D-Galp-(1→ and α-L-Rhap(l→, while they each contained specific glycosidic linkage of →4)-β-D-Glcp(l→, →4)-β-D-Galp-(1→ and →5)-α-L-Araf-(1→ in LP-H, LP-S and LP-SE, resp. Moreover, LP-SE exhibited stronger stimulation than LP-H and LP-S on the proliferation of Lactobacillus plantarum, Lactobacillus bulgaricus, Lactobacillus fermentum and Leuconostoc mesenteroides. The results indicated three extraction methods had some effect on chem. composition and structure of polysaccharide. LP-SE extracted by superfine grinding-assisted enzymic treatment exhibited the highest prebiotic activities, which have a great potential in applying in functional food and medical industry.

The anal. hierarchy process (AHP) combined with response surface methodol. (RSM) was used to optimize the purification conditions of crude polysaccharides from Carex meyeriana Kunth (CMKP) by macroporous resin. The optimal purification conditions as follows: elution volume of 2.74 BV, flow rate of 1.88 BV/h, and sample concentration of 2.10 mg/mL. Under the optimal conditions, the purification comprehensive score was 63.59 ± 1.56% (n = 3), which was agreed closely to the predicted value (64.02%). Two fractions of polysaccharides (CMKP-1 and CMKP-2) were obtained using a DEAE-52 column. Fourier Transform IR (FTIR) spectroscopy spectra revealed the general characteristic absorption peaks of CMKP-1 and CMKP-2. Congo-red test indicated that CMKP-1 and CMKP-2 have triple helix conformation. CMKP-1 and CMKP-2 were both composed of Man, Rib, Rha, GluA, GalA, Glu, Gal, Xyl, Ara and Fuc in a molar ratio of 8.93:1:4.62:1.41:1.78:10.7:24.69:4.51:15.92:1.27 and 7.85:1:5.05:1.99:3.15:12.47:15.41:1.54:6.91:3.15, resp. The major mol. weight of CMKP-1 and CMKP-2 was 22.30 kDa and 48.10 kDa, resp. Moreover, CMKP-1 and CMKP-2 had immunostimulatory effects on RAW264.7 cells in a certain concentration range. It could be used as potential immunomodulatory agents in medicine or functional food fields.
