Pectin oligosaccharides (POSs) have prebiotic and antiglycation activities in vitro, but the specific structure-activity relationship is unclear. In this study, POSs were obtained by enzymic and ultrasound-assisted enzymic degradation of pectin polysaccharide (PPS), resp. Based on the chem. characterization, the antiglycation in vitro and prebiotic activities of POSs were compared and the structure-activity relationship was studied. The results showed that the antiglycation activity of POSs in vitro was proportional to the galacturonic acid content and GalA:Rha molar ratios except for the low mol. weight POSs (LM-POSs), and inversely proportional to its branching degree, such as Ara:Rha and Gal:Rha molar ratios. In addition, it was also found that the prebiotic activity of POSs was pos. correlated with Ara:Rha and Gal:Rha molar ratios in mol. composition and the neutral sugar content, especially galactose and arabinose. The degree of esterification (DE) was less important for both antiglycation and prebiotic activity of POSs. These results provided an important theor. basis for POSs application in food.

A novel polysaccharide DG2 was obtained from the Dendrobium officinale endophytic fungus Fusarium solani DO7. The structural characterization and immunol. activity of DG2 were investigated. Results showed that DG2 with the mol. weight of 152.8 kDa was composed of Galp, Manp, Glcp and Araf at a molar ratio of 4.3:1.9:2.8: 1.0. The main linkage types of DG2 were (1 → )-β-D-Glc, (1 → 4)-β-D-Glc, (1 → 3,6)-β-D-Gal and (1 → 6)-β-D-Gal, which demonstrated DG2 was a novel galactoglucan and different from other fungal and botanic polysaccharide. Furthermore, DG2 did not contain deoxynivalenol, and showed no direct toxicity to human embryonic kidney cells and RAW 264.7 cells. Meanwhile, DG2 could increase the TNF-α, IL-6 and iNOs mRNA expression levels of RAW264.7 cells, and further promote the production of TNF-α, IL-6 and NO. These results suggested DG2 could enhance macrophage-mediated immune responses and have the potential to be an attractive functional food supplement candidate for the hypoimmunity therapies.

As a healthy food and traditional Chinese medicine, sagittaria sagittifolia L. has been used for a long history. Nevertheless, reports on the bioactivity and chem. characterization of S. sagittifolia L. polysaccharides are still very rare. In this paper, ultrasound-assisted method (UAE) was used to extract S. sagittifolia L. polysaccharides, after alc. precipitation and column chromatog. isolation, the structural characteristics, antioxidant and immunol. activities of the purified polysaccharide (SPU60-W) were preliminarily investigated. The results indicated SPU60-W (16.62 kDa) was a pyranoid polysaccharide containing α-glycosidic bond composed of mannose, xylose, and glucose with a molar ratio of 2.69:2.04:95.27. It consisted of slender wormlike strands, which may involve some degree of aggregation of helixes, as well as a small proportion of irregular spherical structures. Furthermore, antioxidant activity anal. showed that SPU60-W possess excellent hydroxyl and ABTS radical scavenging activity comparable to vitamin C (Vc), and moderate DPPH radical scavenging activity. Immunity tests suggested that SPU60-W significantly promoted the proliferation, phagocytosis and NO production of mouse macrophage RAW264.7. According to this study, SPU60-W might be utilized as a potent antioxidant and immunomodulator in food and medicinal industry.

The polysaccharide fraction of Bletilla ochracea (BOP) was isolated from its tubers, and purified by DEAE-52 Cellulose and Sephadex G-200 column chromatog. The structural features of BOP were analyzed by GC-MS, HPLP, FT-IR, methylation and NMR. The average mol. weight of BOP was approx. 4.9 × 105 Da, and the monosaccharide composition was 7.88:2.12 mix of mannose and glucose. Although BOP had no effect on the proliferation rate of CT26 colon carcinoma cells, it significantly inhibited tumor xenograft growth in vivo by stimulating CD4+ T cell expansion in the spleen of the tumor-bearing mice. Taken together, BOP is a potent immunomodulatory agent that can be considered for anti-tumor therapy.

Tissue necrosis is a form of cell death common in advanced and aggressive solid tumors, and is associated with areas of intratumoral chronic ischemia. The histopathol. of necrotic regions appear as a scaffold of cellular membrane remnants, reflective of the hypoxia and cell degradation events associated with this cellular death pathway. Changes in the glycosylation of cell surface proteins is another common feature of cancer progression. Using a recently developed mass spectrometry imaging approach to evaluate N-linked glycan distributions in human formalin-fixed clin. cancer tissues, differences in the glycan structures of regions of tumor, stroma and necrosis were evaluated. While the structural glycan classes detected in the tumor and stromal regions are typically classified as high mannose or branched glycans, the glycans found in necrotic regions displayed limited branching, contained sialic acid modifications and lack fucose modifications. While this phenomenon was initially classified in breast cancer tissues, it has been also seen in cervical, thyroid and liver cancer samples. These changes in glycosylation within the necrotic regions could provide further mechanistic insight to necrotic changes in cancer tissue and provide new research directions for identifying prognostic markers of necrosis.

The impacts of three extraction techniques, including hot water extraction (HWE), pressurized water extraction (PWE), and microwave assisted extraction (MAE), on the physicochem. characteristics, antioxidant activities, in vitro binding properties, and in vitro inhibitory activities on α-amylase and α-glucosidase of okra polysaccharides (OPPs) were investigated and compared. The extraction yields, constituent monosaccharides, and FT-IR spectra of OPP-W, OPP-P, and OPP-M extracted by HWE, PWE, and MAE, resp., were similar. However, their mol. weights, intrinsic viscosities, uronic acids, and degree of esterification were different. Furthermore, results showed that OPP-W, OPP-P, and OPP-M exhibited remarkable antioxidant activities, binding capacities, and inhibitory activities on α-amylase and α-glucosidase. Indeed, the antioxidant activities of OPP-W were significantly lower than those of OPP-M and OPP-P, which might be attributed to the low mol. weights and high contents of unmethylated galacturonic acid of OPP-P and OPP-M. However, the binding capacities and inhibitory activities on α-amylase and α-glucosidase of OPP-W and OPP-P were similar, but significantly higher than those of OPP-M, which might be attributed to the low mol. weights of OPP-M. Results suggested that the PWE method could be a good potential technique for the extraction of OPPs with high bioactivities for industrial applications.

A review. Urinary tract infections (UTls) are very common disorders that affect adult women. Indeed, 50% of all women suffer from UTIs at least one time in their lifetime; 20-40% of them experience recurrent episodes. The majority of UTIs seems to be due to uropathogenic Escherichia coli that invades urothelial cells and forms quiescent bacterial reservoirs. Recurrences of UTls are often treated with non-prescribed antibiotics by the patients, with increased issues connected to antibiotics resistance. D-mannose, a monosaccharide that is absorbed but not metabolized by the human body, has been proposed as an alternative approach for managing UTIs since it can inhibit the bacterial adhesion to the urothelium. This manuscript discusses the mechanisms through which D-mannose acts to highlight the regulatory aspects relevant for determining the administrative category of healthcare products placed on the market. The existing literature permits to conclude that the anti- adhesive effect of D-mannose cannot be considered as a pharmacol. effect and, therefore, D-mannose-based products should be classified as medical devices composed of substances.

The edible mushroom G. frondosa has been used as a kind of functional food for the prevention and therapy of various diseases in Asian countries. In the present work, a novel acid-soluble polysaccharide (GFAP) was successfully isolated from G. frondosa under room temperature and hydrochloric acid solution treatment. Results of chem. composition anal., UV and HPGPC spectra showed that GFAP mainly contained 94.28% of carbohydrate with the average mol. weight of about 644.9 kDa. GC, FT-IR, NMR and methylation anal. further indicated that GFAP was a neutral sugar mainly composed of (1 → 3)-β-D-Glcp and (1 → 3)-α-D-Manp. The in vivo antitumor experiments demonstrated that GFAP could effectively protect thymuses and spleens of tumor-bearing mice and inhibit the growth of H22 solid tumors with the inhibitory rate of 36.72%. Besides, GFAP could significantly improve the activities of NK cells, macrophages, CD19+ B cells and CD4+ T cells, leading to the apoptosis of H22 cells via G0/G1 phase arrested. Our data demonstrated that GFAP holds great application prospect to be a safe and effective antitumor adjuvant in the future.

SEM anal. displayed some morphol. difference existing in polysaccharides before and after degradation More importantly, the antioxidant activities in vitro of AAP after degradation were significantly increased (p < 0.05) and AAPUD12h displayed the strongest antioxidant activity. These results indicate that ultrasonic degradation may be a suitable way to improve the antioxidant activity of natural polysaccharides. In this study, an active component UP1-1 was isolated from Chinese Huangshan Umbilicaria esculenta via hot water extraction and purified by anion-exchange and gel-filtration chromatog. UP1-1 mainly composed of galactose, mannose and glucose in a molar ratio of 0.8:1.0:4.6 with an average mol. weight of 281 kDa. Methylation anal. of UP1-1 revealed the major glycosidic bonds comprised 1,6-linked Glcp, 1,4-linked Glcp, t-linked Glcp, 1,3,6-linked Manp, 1,3-linked Galp, t-linked Galp at the ratio of 2.28:0.38:0.32:0.63:0.25:0.29. Structural anal. results revealed that the backbone of UP1-1 consisted of (6)-β-D-Glcp-(1→ ,→6)-β-D-Manp-(1→, → 4)-β-D-Glcp-(1 → residues with side chains of →3)-β-D-Galp-1→, β-D-Galp-1→ and β-D-Glcp-1→ branches located at O-3 position of → 6-β-D-Manp-1→. Immunostimulatory activity tests showed that UP1-1 could promote the phagocytic activity and NO production of RAW 264.7 cells in a dose-dependent manner. UP1-1 could significantly improve the proliferation effect of RAW 264.7 cells at the concentration of 50 μg/mL. Thus, UP1-1 exerted good immunostimulatory activity, suggesting that UP1-1 has a great potential application in pharmacol. industry.
