Polydatin, a natural analog of resveratrol, has many biol. activities. The better bioavailability of polydatin than resveratrol makes it an ideal candidate for therapy. Polydatin has protective effects against various diseases (cardiovascular, neurol., inflammatory, etc.) including cancer. However, its mechanism of action has not been fully established. Therefore, the present study was initiated to explore the mechanism/s associated with chemotherapeutic effects of polydatin in in vitro using lung cancer A549 cells. Polydatin treatment reduced the proliferation of A549 cells by inducing DNA damage and cell cycle arrest in a concentration-dependent manner. The inhibition of cell proliferation was induced by dual mechanism of senescence and apoptosis. Proteins involved in various pathways were studied using western blotting and immunocytochem. Interestingly, senescent and apoptotic cells induced a differential bystander response (proliferative/toxic) in naive A549 cells. Our results show that polydatin can induce both senescence and apoptosis in A549 cells in a concentration-dependent manner and the differential bystander effects induced by polydatin are regulated by mTOR pathway. This study aimed to investigate the beneficial role of Millettia ferruginea extract (MF) in preventing cisplatin (Cisp) induced nephrotoxicity in rats. A total of 55 metabolites were identified using LC-MS anal. The in vivo results indicated that MF pretreatment for 4 wk (20 mg/kg b.w.) remarkably attenuated the altered renal biomarkers by decreasing the levels of plasma creatinine, urea, and uric acid when compared to the Cisp-group. The nephroprotective capacity of MF was further strengthened by histopathol. observations, where Cisp + MF treated rats showed lower number of inflammatory cells and tubular degenerative changes than the Cisp-group. The harmful effects of cisplatin on renal oxidative stress indicators (MDA, SOD, CAT, and GPx), were restored by the treatment of MF. In addition, the reduction of inflammatory markers (IL-6 and TNF-α), associated with alleviating DNA fragmentation, highlighted the preventive effect of MF in kidney tissue. Addnl., MF components presented lower binding energies when docked into the active site of TNF-α and IL-6. The present findings concluded that M. ferruginea extract exhibited nephroprotective potential, which may be attributed to its antioxidant and anti-inflammatory properties. The present study employs exogenous acetaldehyde at six levels of treatment (7.86 ± 0.10-259.02 ± 4.95 mg/L) before the bottle aging of Merlot wines to encourage phenolic modification. Secondary injury is the main cause of high mortality and poor prognosis of TBI, which has recently been suggested to be related to ferroptosis. Polydatin, a monocrystalline compound extracted from the rhizome of Polygonum, has been shown to exert potential neuroprotective effects. However, its role and mechanism in the secondary injury of TBI has not been elucidated. In this study, the inhibition of Polydatin on ferroptosis was observed both in the Hb treated Neuro2A cells in vitro and in TBI mouse model in vivo, characterized by reversion of accumulation or deposition of free Fe2+, increased content of MDA, decreased activity of key REDOX enzyme GPx4, cell death and tissues loss. Although Polydatin corrected the increased mRNA levels of ferroptosis signaling mols. GPX4, SLC7A11, PTGS2, and ATP5G3 after TBI, TBI and Polydatin treatment had no significant effect on their protein expression. Notably, Polydatin could completely reverse the decrease of GPx4 activity after TBI in vivo and in vitro, and the effect was stronger than that of the classical ferroptosis inhibitor FER-1 in vitro. Further, Polydatin has been shown to reduce the severity of acute neurol. impairment and significantly improve subacute motor dysfunction in TBI mice. Our findings provided translational insight into neuroprotection with Polydatin in TBI by inhibiting ferroptosis mainly depending on the maintenance of GPx4 activity. Treated diabetic rats also showed improvement in motor/sensory nerve conduction velocities and nerve blood flow. Although the grape surface appears to be normal under high temperature during the selling period, the nutritional quality of grape might be greatly affected. Phenolics content and composition are key indicators reflective of grape′s nutritional quality. Herein, the paper investigated effects of short-term high temperature (40 °C; 2 h) on grape phenolics from five table grape varieties to evaluate grape quality. Twenty-three phenolics were identified and quantified through UHPLC-ESI-qTOF-MS2 and UHPLC-QQQ-MS2, resp. The results observed that short-term high temperature significantly reduced flavonoids in European and American varieties. In Eurasian varieties, high temperature induced the accumulation of stilbenes and flavonols compounds while lowered other compounds Myricetin 4′-methylether-3-O-rhamnoside, a differential metabolite responding to high temperature in all table grapes was screened. Combining with the anal. of mol. level, high temperature down-regulated the phenolics biosynthesis-related genes involved in phenolics metabolic pathways (flavonols, stilbenes, anthocyanins and flavan-3-ols metabolic pathways) in grapes except Muscat de Hambourg grapes, such as the expression of VvUFGT, VvANR, VvLAR, VvFLS, VvSTS, VvF3′5′H and VvAOMT. The current research aims to screen and characterize phenolic compounds and estimate their antioxidant potential in rejected mangoes.
