Development of bio oil and bio asphalt by hydrothermal liquefaction using lignocellulose

Bio asphalt is an organic polymer derived from biomass resources, which has the potential to partially or completely replace petroleum asphalt. This study used lignocellulose to prepare bio oil by hydrothermal liquefaction method. A mixed solvent of ethanol and ethylene glycol was introduced to increase the oil yield and decrease the reaction temperature. The oil yield reached the highest when solvent mixing ratio of 1:1, liquid-solid ratio of 6:1, catalyst dosage of 3%, and reaction temperature of 250 degrees C respectively. The bio oil and 50# asphalt were mixed in proportion sheared for 30 min (1000 rpm) under the condition of 135 degrees C to produce bio asphalt. The chemical structures and rheological properties of bio oil and bio asphalt were investigated and compared with petroleum asphalt. Fourier transform infrared spectroscopy (FTIR) and Nuclear Magnetic Resonance analysis indicate that the bio oil expressed a lower H/C ratio, higher O content and more light components including liquid compounds comparing to the petroleum asphalt. The content of the polar group including O-H and C=O of bio oil was higher than that of petroleum asphalt. Gas Chromatography-Mass Spectrometry (GC-MS) was used to prove the presence of olefins and alcohols in bio oil. Thermogravimetric analysis showed that the bio oil expressed better thermal stability than that of petroleum asphalt, which expressed a potential of using as a pavement material. The dynamic shear rheometer (DSR) results showed that the complex shear modulus, G* decreased as the content of bio oil. The Glass transition temperature Tg indicated that the bio asphalt with 10% bio oil content showed similar performance with that of 70# asphalt. (C) 2020 Published by Elsevier Ltd.

Validity and measurement invariance of the Addictions Neuroclinical Assessment incentive salience domain among treatment-seekers with alcohol use disorder

Incentive salience, or the attribution of motivational value to stimuli, is a biopsychological process that is disrupted in alcohol use disorder (AUD). The Addictions Neuroclinical Assessment (ANA) is a framework to characterize heterogeneity in addiction and establish a common assessment battery for research and clinical use. The ANA framework hypothesizes three constructs that correspond to processes in the etiology, course, and treatment of addiction: incentive salience, negative emotionality, and executive function. The current study extends prior findings on the ANA by validating the incentive salience construct among participants (n = 563) in a multisite prospective study of individuals entering treatment for AUD. We used confirmatory factor analysis to test a one-factor model of incentive salience. Indicators included items assessing perception of urges to drink from the Alcohol Dependence Scale, Impaired Control Scale, and Marlatt Relapse Interview. Results indicated the one-factor model fit the data well (chi(2) (12) = 19.42, p = .08; RMSEA = 0.034 [90% CI: 0.000, 0.060], CFI = 0.992) and was measurement invariant across sex. Incentive salience was associated with drinking patterns (e.g., drinks per day, r = 0.447 [95% CI: 0.379, 0.514]); reasons for drinking (urges/temptation r = 0.529 [95% CI: 0.460, 0.599]); testing personal control, r = 0.384 (95% CI: 0.308, 0.461); social pressure, r = 0.549 (95% CI: 0.481, 0.617); and family history of AUD, r = 0.134. The incentive salience factor demonstrated greater predictive validity for drinking outcomes compared to alternative preexisting scales. Overall, this study provides support for the construct validity and measurement invariance of the ANA incentive salience construct in a sample of individuals seeking AUD treatment.

Tissue-specific transcriptome recovery on withdrawal from chronic alcohol exposure in zebrafish

Alcohol consumption can lead to a wide range of systemic disorders brought about by transcriptional changes. Recent studies have documented altered behavior and physiology in zebrafish exposed to alcohol. In this work, we have identified the changes in the zebrafish transcriptome in response to chronic alcohol exposure. We have further followed the extent of transcriptional recovery upon withdrawal from alcohol and found evidence of tissue-specific responses. Our results indicate a greater extent of recovery of the brain transcriptome compared to the liver. We identify two distinct classes of genes in response to withdrawal from alcohol exposure – those that recover their pre-alcohol expression profile versus those that retain altered expression even after the fish are removed from the alcohol environment. Finally, we have examined gender-specific responses to alcohol exposure in zebrafish and find evidence for distinct alcohol tolerance levels. Upon chronic alcohol exposure, a higher percentage of genes show perturbation in expression profile in males compared to females. Female fish also recover better with more genes regaining the control expression level upon withdrawal from alcohol. Overall, our work identifies genes and pathways perturbed by exposure to alcohol, and demonstrates the extent of gender- and tissue-specific transcriptional changes associated with chronic alcoholism and withdrawal. Transcriptional regulation of alcohol induced liver fibrosis in a translational porcine hepatocellular carcinoma model

Hepatocellular carcinoma (HCC) is the 5th most common and 2nd deadliest cancer worldwide. HCC risk factors include alcohol induced liver cirrhosis, which prompts hepatic inflammation, cell necrosis, and fibrosis deposition. As 25% of HCC cases are associated with alcohol induced liver disease, understanding the effects of the cirrhotic liver microenvironment on HCC tumor biology and therapeutic responses are critical. This study utilized the Oncopig Cancer Model-a transgenic pig model that recapitulates human HCC through induced expression of KRAS(G12D) and TP53(R167H )driver mutations-to investigate the molecular mechanisms underlying alcohol induced liver disease. Oncopigs (n = 5) underwent fibrosis induction via infusion of ethanol and ethiodized oil (1:3 v/v dosed at 0.75 mL/kg) into the hepatic arterial circulation. Eight-weeks post induction, liver tissue samples from fibrotic and age-matched control (n = 5) Oncopigs were collected for histological evaluation and transcriptional profiling. Increased hepatic inflammation and fibrosis was observed in fibrotic Oncopigs via pathological assessment. Transcriptional profiling (RNA-seq) resulted in the identification of 4387 differentially expressed genes between Oncopig fibrotic and control livers. GO term enrichment analysis identified pathway alterations associated with cirrhosis progression in humans, including cell proliferation, angiogenesis, extracellular matrix deposition, and oxidation-reduction. Key alterations include activation of hepatic stellate cells, increased matrix metalloproteinase production, and altered expression of ABC and SLC transporter genes involved in transport of anticancer drugs.These results demonstrate Oncopig liver fibrosis recapitulates transcriptional hallmarks of human cirrhosis, making the Oncopig an ideal model for studying the effects of the cirrhotic liver microenvironment on HCC tumor biology and therapeutic response. Social interaction anxiety and perceived coping efficacy: Mechanisms of the association between minority stress and drinking consequences among sexual minority women

Sexual minority women (SMW; individuals who identify as women and as lesbian, bisexual, or with another sexual minority identity) are at increased risk for problematic alcohol use compared to their heterosexual counterparts. This increased risk has been attributed to minority stress. However, longitudinal research examining associations between minority stress and alcohol use outcomes is extremely limited and examinations of these associations at the daily level are nearly non-existent. Further, few longitudinal studies have examined mechanisms through which minority stress may impact alcohol use. We utilized data from a 14-day daily diary study of 98 SMW to examine daily-level associations between experiences of minority stress, alcohol consumption and consequences, and two proposed mediators of these associations (perceived coping efficacy, social interaction anxiety). Results indicated that on days when participants experienced minority stress events, they experienced lower coping efficacy, higher social interaction anxiety, and more drinking consequences than usual. Minority stress was not associated with same-day alcohol consumption. Perceived coping efficacy and social interaction anxiety mediated the same-day association between minority stress and drinking consequences. No prospective associations were significant, suggesting that studies with multiple assessments per day may be necessary to detect immediate effects of minority stress. Findings highlight the potential impact of daily experiences of minority stress on alcohol consequences and provide evidence that two general psychological processes may be mechanisms through which minority stress impacts alcohol consequences. These results provide evidence of a need for interventions that teach SMW skills for coping with minority stress and its psychological consequences.

Comparative Transcriptome Analysis of Hybrid Population Provides Insights Into Starch Content in Sweet Potato (Ipomoea batatas L.) Storage Root

Sweet potato is an important starchy crop, which is mainly utilized as food, animal feed, and fermented to produce fuel-grade alcohol. However, the genetic mechanism regulating starch content in sweet potato is not yet fully understood. In this study, transcriptome analysis was performed in sweet potato cultivars Luoxushu 8 (high starch), Zhengshu 20 (low starch), and 12 hybrid F-1 lines (six high-starch lines and six low-starch lines), which detected a total of 85,493 genes. The high and low-starch content groups were compared and found that 47 and 64 genes were up-regulated and down-regulated, respectively. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that seven differentially expressed genes were significantly enriched in the ATP-binding cassette (ABC) transporter pathway. Among them, five genes (annotated as ABCB1) that might involve in the auxin transport were significantly down-regulated in high-starch group. Meanwhile, the storage root sizes of Zhengshu 20 were higher as compared with Luoxushu 8. Simultaneously, four of the five differentially expressed genes enriched in the starch and sucrose metabolism pathway were up-regulated in high-starch group. These results provided the basis for the study of starch content in sweet potato storage root.

Conversion of ethanol to 1,3-butadiene over Ag-ZrO2/SiO2 catalysts: The role of surface interfaces

A series of Ag-ZrO2/SiO2 catalysts with different metal-support interfaces were synthesized in an effort to elucidate the roles of specific interfaces in controlling the ethanol to 1,3-butadiene conversion and selectivity. According to the results of detailed characterizations (e.g. CO/pyridine-DRIFTS, XPS, TEM, NH3-TPD, and H-1 MAS NMR), it was found that the Ag-O-Si interfaces significantly enhanced the dehydrogenation of ethanol while the presence of ZrO2 improved the interaction between Ag and ZrO2 /SiO2, creating more Ae active sites. The high dispersion of ZrO2 on SiO2 generated abundant Zr-O-Si interfaces with medium and weak Lewis acidity, promoting the condensation of acetaldehyde to crotonaldehyde. These Zr-O-Si interfaces in close interaction with Ae species played a critical role in the enhanced H transfer during the MPV reduction of crotonaldehyde to crotyl alcohol. The synergies among the interfaces resulted in retarded ethanol dehydration reactivity, balanced ethanol dehydrogenation and condensation reactions, and a subsequent high 1,3-butadiene yield. Skull Fracture Patterns and Morphologies Among Fatal Motorcycle Traffic Accident Victims in Vietnam

We aim to describe morphological structures of skull fractures and relevant factors in motorcycle accident victims in Vietnam. This work represents a retrospective cross-sectional study based on forensic reports of fatal motorcycle accident victims. Between January 2013 and August 2019, a total of 226 fatal motorcycle accident patients with skull fracture diagnoses were enrolled. Linear and depressed fractures were the common patterns (46.0% and 37.2% of cases, respectively), whereas stellate (11.5%) fractures were rare. Fractures of the temporal bone (68.6% of cases) and basilar skull (60.6%) were the most common, whereas fractures of the parietal bone were the least common (9.7%). Two or more patterns of skull fracture were recorded in 25.5% of cases, and 2 or more fracture locations were recorded in 76.6% of cases. Fractures of the parietal bone were associated with victims not wearing a helmet. In addition, fractures of the basilar skull were more likely to occur among victims with alcohol consumption. We found the common pattern of skull fractures was linear and depressed fractures, and the common location of skull fractures was temporal and basilar bone. Further studies that include larger sample sizes and collect more information should be conducted to better understand relationships between skull fractures and related factors.

Regular Elucidating the pH influence on pulsed electric fields-induced self-assembly of chitosan-zein-poly(vinyl alcohol)-polyethylene glycol nanostructured composites

Hypothesis: The high incompatibility of bio-based materials such as protein and polysaccharides require a series of modifications to develop stable microstructures effectively. By modifying the density and charge of surface residues, pulsed electric fields processing can improve inter/intramolecular interactions, compatibility, and microstructure of bio-based nanostructured composites. Experiment: In this work, the impact of pulsed electric fields at a specific energy of 60-700 kJ/kg (electric field strength = 1.6 kV/cm) on self-assembly of zein-chitosan-poly(vinyl alcohol)-polyethylene glycol composite dispersion was investigated at pH 4.0, 5.7, and 6.8. Findings: Superior complex coacervated matrices were assembled at pH 4.0 and 5.7 before and after pulsed electric fields treatment at a specific energy of 390-410 kJ/kg. The compact and homogenous behaviour was attributable to pulsed electric fields-induced alteration of functional group interactions in a pH-dependent manner. Irrespective of the pH, very high electric field intensity caused excessive system perturbation leading to severe fragmentation and poor development of coacervates. The crucial insights from this study reveal that the self-assembly behaviour and integration of biopolymer-based sys-tems possessing different local charges can be enhanced by optimising pulsed electric fields processing parameters and the properties of the colloidal systems such as the pH. Selective CO2 Electrochemical Reduction Enabled by a Tricomponent Copolymer Modifier on a Copper Surface

Electrochemical CO2 reduction over Cu could provide value-added multicarbon hydrocarbons and alcohols. Despite recent breakthroughs, it remains a significant challenge to design a catalytic system with high product selectivity. Here we demonstrate that a high selectivity of ethylene (55%) and C2+ products (77%) could be achieved by a highly modular tricomponent copolymer modified Cu electrode, rivaling the best performance using other modified polycrystalline Cu foil catalysts. Such a copolymer can be conveniently prepared by a ring-opening metathesis polymerization, thereby offering a new degree of freedom for tuning the selectivity. Control experiments indicate all three components are essential for the selectivity enhancement. A surface characterization showed that the incorporation of a phenylpyridinium component increased the film robustness against delamination. It was also shown that its superior performance is not due to a morphology change of the Cu underneath. Molecular dynamics (MD) simulations indicate that a combination of increased local CO2 concentration, increased porosity for gas diffusion, and the local electric field effect together contribute to the increased ethylene and C2+ product selectivity.
