The present study highlights the importance of understanding the structural changes of micelles induced by drug loading on their physico-chem. properties. A block copolymer with attached fructose, which interacts with GLUT5 receptor, was used and conjugated with a low and a high amount of platinum drugs. Against expectations, the low-loading micelle, despite having a less defined morphol. and larger nanoparticle size according to TEM, displays higher cellular uptake and higher toxicity. This behavior can only be understood when elucidating addnl. information on the structure of micelles. Extensive solid-state NMR measurements were therefore employed to reveal that the drug loading affected swelling and mobility of core and shell of the micelle. The results obtained from solid-state NMR spectroscopy could explain all the observations on this system. In summary, solid-state NMR spectroscopy is an excellent tool to understand the effects of drug loading on the behavior of micelles.

This study aims to determine the qual. and quant. contents of Turkish propolis collected from various regions of Turkey using high-performance liquid chromatog. with the diode-array detector (HPLC-DAD) and gas chromatog.-mass spectrometry (GC-MS) in water and ethanolic extracts In HPLC-DAD analyses, it was determined that water extract of Turkish propolis contains phenolic acids such as caffeic acid (204.00 μg/mL), trans-cinnamic, chlorogenic, and caffeoylquinic acids, responsible for its antioxidant activity; whereas, the ethanolic extract of Turkish propolis contains chrysin (641.33 μg/mL), caffeic acid phenethyl ester (630.67 μg/mL), pinocembrin (572.67 μg/mL), galangin (534.11 μg/mL), naringenin (372.39 μg/mL), and also kaempferol, trans-cinnamic acid, caffeic acid, myricetin, and quercetin. GC-MS analyses showed that ethanolic extract of propolis contains caffeic acid by Rtx-1 column and the water extract of propolis contains quinic acid and ferulic acid by Rtx-5ms column. Various sugar derivatives were detected by both columns in water and ethanolic extracts of Turkish propolis. HPLC-DAD can be considered as a more effective method than GC-MS for the chem. characterization of propolis. Water extract of Turkish propolis can be a good source of raw materials for various sectors, as it is both cheap and has less health risk than ethanolic extract, and is suitable for human use.

Lactulose is considered as a prebiotic because it promotes the intestinal proliferation of Lactobacillus acidophilus which is added to various milk products. Moreover, lactulose is used in pharmaceuticals as a gentle laxative and to treat hyperammonemia. This study was aimed at the total synthesis of two Lactulose-derived oligosaccharides: one is 3-O-β-D-galactopyranosyl-D-fructose, D-fructose and β-D-galactose bounded together with β-1,3-glycosidic bound, the other is 1-O-β-D-galactopyranosyl-D-fructose, D-fructose and β-D-galactose bounded together with β-1,1-glycosidic bound, which were accomplished in seven steps from D-fructose and β-D-galactose and every step of yield above 75%. This synthetic route provided a practical and effective synthetic strategy for galacto-oligosaccharides, starting from com. available monosaccharides. Then we evaluated on their prebiotic properties in the search for potential agents of regulating and improving the intestinal flora of human. The result showed that the prebiotic properties of Lactulose-derived oligosaccharides was much better than Lactulose. Among them, 3-O-β-D-galactopyranosyl-D-fructose displayed the most potent activity of proliferation of L. acidophilus.

The therapeutic potency of platinum-based anticancer drugs can be substantially improved through the use of polymeric nanocarrier systems to target cancer cells efficiently. We synthesized a series of glyco-block copolymers with three different sugars able to self-assemble into nanoparticles when conjugated with 1,2-diamino-cyclopentane platinum(II) (DACP-Pt). The polymers are based on (2-(D-glucosyloxy)ethyl methacrylate (glucose; GlcMA), 2-(D-galactosyloxy)ethyl methacrylate (galactose; GalMA), 1-O-methacryloyl-β-D-fructopyranose (fructose; FrucMA1), and 3-O-methacryloyl-β-D-fructopyranose (fructose; FrucMA3)). They are all readily taken up intracellularly by the breast cancer cell lines MCF-7 and MDA-MB-231 and the ovarian cancer cell line A2780. All cell lines expressed a high preference for the fructose coated nanoparticles. The nanocarriers were themselves nontoxic, but exhibited high cytotoxicity and increased efficacy when conjugated with the DACP-Pt drug.

Naturally occurring fructosamines are of high clin. significance due to their potential use in diabetes mellitus monitoring (quantification of fructosylated Hb, HbA1c) or for the study of their reactivity in consecutive reactions and harmfulness towards the organism. Here the authors report the specific synthesis of the fructosylated dipeptide L-valyl-L-histidine (Fru-Val-His) and fructosylated L-valine (Fru-Val). Both are basic tools for the development and validation of enzymic HbA1c assays. The two fructosamine derivatives were synthesized via a protected glucosone intermediate which was coupled to the primary amine of Val or Val-His, performing a reductive amination reaction. Overall yields starting from fructose were 36% and 34% for Fru-Val and Fru-Val-His, resp. Both compounds were achieved in purities and > 90%. A HILIC-ESI-MS/MS method was developed for routine anal. of the synthesized fructosamines, including starting materials and intermediates. The presented method provides a well-defined and efficient synthesis protocol with purification steps and characterization of the desired products. The functionality of the fructosylated dipeptide has been thoroughly tested in an enzymic HbA1c assay, showing its concentration-dependent oxidative degradation by fructosyl-peptide oxidases (FPOX).

Free sugars were rapidly converted into the corresponding di-O-isopropylidene derivatives and Me O-benzylidene glycosides in excellent yields and purity in a deep eutectic solvent made from choline chloride and malonic acid (ChCl:MA). Reaction conditions were mild; work-up was easy, and further purification was not necessary. Given the inexpensive, nontoxic, and recyclable nature of ChCl:MA, this protocol is environmental friendly.
