Alterations in Insulin Levels in Adults with Prenatal Alcohol Exposure

Objective Evidence suggests that prenatal alcohol exposure (PAE) may adversely impact insulin production and signaling but there is limited information on the range of these effects and their future health consequences. Method A prospective cohort of predominantly African-American individuals identified while in utero and followed into adulthood were used to evaluate differences in various indicators of diabetes, including fasting plasma glucose, hemoglobin A1c (HbA1c), and insulin levels. The homeostatic model assessment of insulin resistance (HOMA-IR) was also computed. Body mass index (BMI) was calculated and normal defined as < 25 kg/m(2). Participants were categorized as having PAE (n = 39) if their mothers drank at least 1 ounce of absolute alcohol per week or more during the 1(st) trimester of pregnancy and as Controls (n = 22) if their mothers reported abstaining from alcohol consumption during pregnancy. Results Mean age of the sample was 36.0 +/- 1.5 years. Indices of glucose metabolism, including fasting plasma glucose and hemoglobin A1c levels, did not vary by group status but insulin levels and HOMA-IR values varied by group status and BMI level. PAE individuals with a normal BMI had lower insulin levels than Controls. However, in PAE subjects, there was a steeper increase in insulin levels relative to their BMI than in Control subjects. A cluster of 5 PAE cases had low levels of insulin and 4 of the 5 had severe cognitive impairment. Conclusions The bidirectional effects on insulin level and insulin resistance associated with PAE may indicate differential rates of diabetes disease impact or differential PAE impact in the brain and peripheral areas involved in insulin production and signaling. Single-wall carbon nanotube-containing cathode interfacial materials for high performance organic solar cells

Water/alcohol soluble cathode interfacial materials (CIMs) are playing important roles in optoelectronic devices such as organic light emitting diodes, perovskite solar cells and organic solar cells (OSCs). Herein, n-doped solution-processable single-wall carbon nanotubes (SWCNTs)-containing CIMs for OSCs are developed by dispersing SWCNTs to the typical CIMs perylene diimide (PDI) derivatives PDIN and PDINO. The Raman and X-ray photoelectron spectroscopy (XPS) measurement results illustrate the n-doped behavior of SWCNTs by PDIN/PDINO in the blend CIMs. The blended and n-doped SWCNTs can tune the work function and enhance the conductivity of the PDI-derivative/SWCNT (PDI-CNT) composite CIMs, and the composite CIMs can regulate and down-shift the work function of cathode, reduce the charge recombination, improve the charge extraction rate and enhance photovoltaic performance of the OSCs. High power conversion efficiency (PCE) of 17.1% and 17.7% are obtained for the OSCs based on PM6:Y6 and ternary PM6:Y6:PC71BM respectively with the PDI-CNT composites CIMs. These results indicate that the n-doped SWCNT-containing composites, like other n-doped nanomaterials such as zero dimensional fullerenes and two dimensional graphenes, are excellent CIMs for OSCs and could find potential applications in other optoelectronic devices.

Genetic polymorphisms in pri-let-7a-2 are associated with ischemic stroke risk in a Chinese Han population from Liaoning, China: a case-control study

Ischemic stroke is a complicated disease, and its pathogenesis has been attributed to the occurrence of genetic polymorphisms. Evidence has suggested that the microRNA let-7a is involved in the pathogenesis of ischemic stroke. Pri-miRNA is the primary transcript, which undergoes several processing steps to generate pre-miRNA and, later, mature miRNAs. In this case-control study, we analyzed the distribution of pri-let-7a-2 variants in patients at a high risk for ischemic stroke and the interactions of pri-let-7a-2 variants and environmental factors. Blood samples and clinical information were collected from 1086 patients with ischemic stroke and 836 healthy controls between December 2013 and December 2015 at the First Affiliated Hospital of China Medical University. We found that the rs1143770 CC genotype and the C allele were associated with a decreased risk of ischemic stroke, whereas the rs629367 CC genotype was associated with an increased risk for ischemic stroke. Moreover, these two single-nucleotide polymorphisms were in linkage disequilibrium in this study sample. We analyzed gene-environment interactions and found that rs1143770 exerted a combined effect on the pathogenesis of ischemic stroke, together with alcohol use, smoking, and a history of hypertension. Therefore, the detection of pri-let-7a-2 polymorphisms may increase the awareness of ischemic stroke risk. This study was approved by the Institutional Ethics Committee of the First Affiliated Hospital of China Medical University, China (approval No. 2012-38-1) on February 20, 2012, and was registered with the Chinese Clinical Trial Registry (registration number: ChiCTR-COC-17013559) on December 27, 2017.

An investigation into Sunitinib malate nanoparticle production by US- RESOLV method: Effect of type of polymer on dissolution rate and particle size distribution

The present study practiced ultrasonic-assisted rapid expansion of a supercritical CO2 solution into a liquid solvent (US-RESOLV) process for the synthesis of nanoparticles (NPs) of Sunitinib malate for the first time. Hydrophilic polymers including hydroxypropyl methylcellulose (HPMC), poly (vinyl alcohol) (PVA), chitosan (CHI) and polyethylene glycol (PEG) were utilized for modifying the size distribution and increasing the dissolution rate of the Sunitinib malate NPs. Also, we determined the impact of pressure, nozzle diameter as well as temperature on morphology, size distribution and dissolution rate of produced particles. In order to accomplish the objectives of the present study, PEG was the best polymer for controlling the particle size distribution and HPMC served as the best polymeric stabilizer for increasing the dissolution rate of the drug particles. The synthesized particles were much smaller (< 600 nm) in comparison to the unprocessed particles of Sunitinib malate. The combined effects of the smaller sizes of the particles, ultrasonication and the presence of polymers could enhance dissolution rate of the Sunitinib malate in the aqueous media. Characterization of a novel type III alcohol dehydrogenase from Thermococcus barophilus Ch5

The genome of the hyperthermophilic and piezophilic euryarchaeaon Thermococcus barophilus Ch5 encodes three putative alcohol dehydrogenases (Tba ADHs). Herein, we characterized Tba ADH(547) biochemically and probed its catalytic mechanism by mutational studies. Our data demonstrate that Tba ADH547 can oxidize ethanol and reduce acetaldehyde at high temperature with the same optimal temperature (75 degrees C) and exhibit similar thermostability for oxidization and reduction reactions. However, Tba ADH(547) has different optimal pH for oxidation and reduction: 8.5 for oxidation and 7.0 for reduction. Tba ADH(547) is dependent on a divalent ion for its oxidation activity, among which Mn2+ is optimal. However, Tba ADH(547) displays about 20% reduction activity without a divalent ion, and the maximal activity with Fe2+. Furthermore, Tba ADH(547) showcases a strong substrate preference for 1-butanol and 1-hexanol over ethanol and other alcohols. Similarly, Tba ADH(547) prefers butylaldehyde to acetaldehyde as its reduction substrate. Mutational studies showed that the mutations of residues D195, H199, H262 and H274 to Ala result in the significant activity loss of Tba ADH(547), suggesting that residues D195, H199, H262 and H274 are responsible for catalysis. Effects of chemogenetic manipulation of the nucleus accumbens core in male C57BL/6J mice

Binge drinking is a widespread public health concern with limited effective treatment options. To better select pharmaceutical targets, it is imperative to expand our knowledge of the underlying neural mechanisms involved in binge drinking. Our previous experiments in C57BL/6J female mice found that increasing activity in the nucleus accumbens (NAc) core using excitatory Designer Receptors Exclusively Activated by Designer Drugs (DREADDs) reduced binge-like drinking. These results differed from what has been found in males; however, it is unclear whether differences in experimental procedures or sex underlie these discrepancies. We matched the conditions used in our female study and asked whether bidirectional manipulation of NAc core activity has different effects on binge-like drinking in males. Male C57BL/6J mice were stereotaxically injected with AAV2 hSyn-HA hM3Dq (excitatory), -hM4Di (inhibitory), or -eGFP bilaterally into the NAc core. We tested the effects of altering NAc activity on binge-like ethanol intake using Drinking in the Dark (DID). During the first week, mice were pre-treated with vehicle to establish baseline ethanol intake. In week 2, mice were treated with 1 mg/kg CNO prior to DID to determine the effects of DREADD-induced changes in NAc core activity on ethanol intake. Decreasing activity via CNO/hM4Di significantly decreased binge-like drinking in male mice relative to eGFP and hM4Di groups. We also measured intake of sucrose, quinine, and water after CNO treatment and found that increasing NAc core activity via CNO/hM3Dq increased quinine intake, and increased water intake over time. We did not observe significant differences in the GFP or hM4Di groups. This work suggests there exist apparent sex-related differences in NAc core contributions to binge-like alcohol drinking, thus demonstrating the need for inclusion of both sexes in future work. Time series Fourier transform infrared spectroscopy for characterization of water vapor sorption in hydrophilic and hydrophobic polymeric films

This work investigates the nature of the molecular interactions between water vapor and polymers using time series Fourier transform infrared (FTIR) spectroscopy. A detailed analysis of the frequency shifts and relative peak intensities led to the conclusion that polyvinyl alcohol (PVOH) sorbed a large quantity of water vapor molecules, resulting in swelling and dissolving of polymer crystallites. Difference spectra were calculated to investigate spectral changes occurring upon sorption by dividing the spectra of polymers during the sorption time series by the spectrum of the dry sample and subsequently subtracting the water vapor spectrum. Based on the absorbance area of the OH stretching vibration region (4000-3000 cm(-1)) in difference spectra, the amount of water sorbed was significantly higher in poly-L-lactic acid (PLLA) and polyvinyl chloride (PVC) than in polyethylene (PE) and polytetrafluoroethylene (PTFE), increasing with the hydrophilicity of the surface. The OH stretching band of difference spectra shifted from 3499 cm(-1) for PVC, to 3416 cm(-1) for PE and finally to 3387 cm(-1) for PTFE, indicating a more strengthened hydrogen-bonding network in the PTFE matrix upon water vapor sorption. Cheers together, but not alone: Peer drinking moderates alcohol consumption following interpersonal stress

In view of the importance of the need to belong in motivating behavior, we examined whether interpersonal and academic stress differentially influence social and solitary alcohol consumption and whether social and solitary alcohol consumption differentially predict next-day interpersonal and academic stress. Based on research suggesting that drinking with friends is related to increased alcohol consumption following belongingness threat, we also examined whether peer consumption moderates associations between daily interpersonal stress and social drinking. Each day for 30 days, 1641 undergraduates reported stress, alcohol consumption, and peer consumption. Academic stress was related to lower levels of social and solitary alcohol consumption. Interpersonal stress was associated with greater social alcohol consumption, but only when students reported being around others who were drinking greater than average peer quantities. However, although social drinking was related to lower next-day academic stress, it was unrelated to next-day interpersonal stress. Findings are consistent with the notion that individuals’ perceptions of peers’ alcohol use might serve as a signal to join in this behavior to reduce belongingness threats associated with interpersonal stressors, although this may not be an effective strategy.

Reducing anionic surfactant adsorption using polyacrylate as sacrificial agent investigated by QCM-D

Surfactant losses by adsorption to rock surfaces make surfactant-based enhanced oil recovery economically less feasible. We investigated polyacrylate (PA) as a sacrificial agent in the reduction of anionic surfactant adsorption with focus on calcite surfaces by using quartz crystal microbalance with dissipation monitoring. It was found that the adsorption of the anionic surfactant alcohol alkoxy sulfate (AAS) followed a Langmuir adsorption isotherm, and the adsorbed amount reached saturation above its critical micellar concentration. Adsorption of PA was a much slower process compared to AAS adsorption. Increasing the calcium ion concentration also increased the amount of AAS adsorbed as well as the mass increase rate of PA adsorption. Experimental results combined with density functional theory calculations indicated that calcium cation bridging was important for anionic surfactant AAS and PA adsorption to calcite surfaces. To effectively reduce the amount of surfactant adsorption, it was needed to preflush with PA, rather than by a simultaneous injection. Preflushing with 30 ppm of PA gave a reduction of AAS adsorption of 30% under high salinity (HS, 31,800 ppm) conditions, compared to 8% reduction under low salinity (LS, 3180 ppm) conditions. In the absence of PA, the amount of adsorbed AAS was reduced by already 50% upon changing from HS to LS conditions. Lower calcium ion concentrations, as under LS conditions, contributed to this observation. On different mineral surfaces, PA reduced the AAS adsorption in the order of alumina > calcite > silica. These results offer important insights into mitigating surfactant adsorption using PA polyelectrolyte as sacrificial agent and contribute to improved flooding strategies with reduced surfactant loss.
