Poly(ethylene terephthalate) (PET) is known for its various useful characteristics, including its applicability in cardiovascular applications, more precisely as synthetic bypass grafts for large diameter (≥ 6 mm) blood vessels. Although it is widely used, PET is not an optimal material as it is not interactive with endothelial cells, which is required for bypasses to form a complete endothelium. Therefore, in this study, poly(alkylene terephthalate)s (PATs) have been studied. They were synthesized via a single-step solution polycondensation reaction, which requires mild reaction conditions and avoids the use of a catalyst or additives like heat stabilizers. A homologous series was realized in which the alkyl chain length varied from 5 to 12 methylene groups (n = 5-12). Molar masses up to 28,000 g/mol were obtained, while various odd-even trends were observed with modulated differential scanning calorimetry (mDSC) and rapid heat-cool calorimetry (RHC) to access the thermal properties within the homologous series. The synthesized PATs have been subjected to in vitro cell viability assays using Human Umbilical Vein Endothelial Cells (HUVECs) and Human Dermal Microvascular Endothelial Cells (HDMECs). The results showed that HUVECs adhere and proliferate most pronounced onto PAT(n = 9) surfaces, which could be attributed to the surface roughness and morphol. as determined by at. force microscopy (AFM) (i.e. Rq = 204.7 nm). HDMECs were investigated in the context of small diameter vessels and showed superior adhesion and proliferation after seeding onto PAT(n = 6) substrates. These preliminary results already pave the way towards the use of PAT materials as substrates to support endothelial cell adhesion and growth. Indeed, as superior endothelial cell interactivity compared to PET was observed, time-consuming and costly surface modifications of PET grafts could be avoided by exploiting this novel material class. Glycerol and alkanediols are being studied as alternative solvents to extract phytochems. from plant material, often as hydrogen bond donors in deep eutectic solvents (DESs). Many of those alcs. are liquid at room temperature, yet studies of their use as extraction solvents are scarce. In this work, glycerol and a series of alkanediols (1,2-ethanediol, 1,2-propanediol, 1,3-propanediol, 1,3-butanediol, 1,2-pentanediol, 1,5-pentanediol, and 1,2-hexanediol) were studied for the extraction of phenolic compounds from Juglans regia L. leaves, a rich source of this class of bioactive compounds The extraction yield was quantified, and the bioactivity of both extracts and pure solvents was evaluated by measuring the anti-inflammatory and cytotoxic activities. The solvents showing the best combined results were 1,2 and 1,3-propanediol, as their extracts presented a high amount of phenolic compounds, close to the results of ethanol, and similar cytotoxicity against cervical carcinoma cells, with no impact on non-tumor porcine liver cells in the studied concentration range. On the other hand, none of the extracts (and solvents) presented anti-inflammatory activity. In this study, dried PE fruit powder was extracted, tested for biol. activities, and loaded into transfersomes for hair follicle targeting. Before lyophilization, PE fruit powder was extracted using 2 solvent systems, water and 30% ethanol. The PE 30% ethanolic extract had higher antioxidant activity and total phenolic content than the PE aqueous extract However, the cytotoxicity of the PE 30% ethanolic extract was higher than that of PE aqueous extract As a result, the PE aqueous extract was analyzed using ultra-performance liquid chromatog. and found that the major component of the PE aqueous extract was gallic acid. Afterward, the PE aqueous extract was tested for its potential to activate the expression of genes involved in hair growth promotion in human keratinocytes. At a non-toxic concentration (10 μg/mL), this extract promoted various growth factors comparable to 1% minoxidil. PE-loaded transfersomes were prepared to deliver the PE aqueous extract to the hair follicle. The particle size and polydispersity index of PE-loaded transfersomes were 228 nm and 0.25, resp. After 3 mo of storage, the particle size at 4°C and 30°C was 218 nm and 241 nm, resp., which was comparable to its initial size. However, at 40°C, the particle size dramatically increased (315 nm). The fluorescent agent, rhodamine B, was used to evaluate the potential of transfersomes to target hair follicles. Rhodamine B transfersomes had better penetration and accumulation in hair follicles than rhodamine B solution To conclude, the PE aqueous extract, mainly composed of gallic acid, can activate hair growth gene expression. Herein, fabrication of colonic enzyme-responsive dextran based oligoester crosslinked nanoparticles is reported for the controlled release of 5-fluorouracil (5-FU) – an anticancer drug. The 5-FU drug loaded nanoparticles (DNPs, size ∼237 ± 25 nm, ζ-potential -17.0 ± 3 mV) were developed by the in-situ crosslinking of dextran with a bifunctional telechelic oligoester followed by the phys. drug encapsulation via nanopptn. Drug encapsulation efficiency and drug loading capacity of DNPs were found to be ∼76% (±0.1) and ∼8% (±0.1), resp. The DNPs were demonstrated to release the encapsulated drug selectively in the presence of dextranase enzyme. The in vitro release kinetics assay revealed that the DNPs released about 75% (±4) of the entrapped drug within 12 h of incubation with dextranase enzyme. No drug was released in a control experiment where DNPs were exposed to pH conditions encountered in the stomach and small intestine. Moreover, the treatment of HCT116 colon cancer cell line with the developed DNPs highlighted its biocompatibility as well as dextranase triggered cytotoxicity. The developed system offers an avenue to reduce the non-specific cytotoxicity of the encapsulated 5-FU, and a colon specific delivery of the encapsulated drug in response to the dextranase enzyme.
