Structure-activity relationship of the left side-chain on Ph substitutions were explored which delivered many potent ALK inhibitors. Among them, I showed favorable pharmacokinetic profiles in rats and dogs together with significant antitumor efficacy in EML4-ALK fusion xenograft model.
