Smoke in the air – Associations between experimentation with cannabis and the use of tobacco and nicotine products among 15-16-year-old school students

Background: Cannabis is the most popular illicit drug among adolescents in developed countries, including Finland. The aim of this study was to estimate 1) how cannabis experimentation among Finnish adolescents changed between 2003 and 2019, and 2) whether the associations between cannabis experiments and factors related to it, especially the use of tobacco and nicotine products, changed during the same time period. Methods: The analyses are based on the European School Survey Project on Alcohol and Other Drugs data, collected from Finnish adolescents aged 15 to 16 in 2003, 2007, 2011, 2015, and 2019 (N = 20,630). Results: Experimentation with cannabis among Finnish adolescents has increased since the beginning of our follow-up. At the same time, alcohol use and smoking have decreased markedly, and attitudes toward cannabis use have become more relaxed. The association between smoking and cannabis experimentation has become weaker over time. However, the use of tobacco and nicotine products, especially polytobacco, is still a strong risk factor for experimentation with cannabis. The higher the number of tobacco or nicotine products used, the higher the probability for cannabis experimentation. Conclusions: Experimentation with cannabis has previously been concentrated predominantly on adolescents who smoke, but recently non-smokers are increasingly trying cannabis. It is possible that alternative ways of using cannabis may have increased its use. Despite the strict cannabis policy in Finland, its use has increased, which may be an indication that youth cultures and images of different substances play a significant role in adolescents experimenting with cannabis.

Monolignol oxidoreductases are members of the berberine bridge enzyme-like (BBE-like) protein family (pfam 08031) that oxidize monolignols to the corresponding aldehydes. They are FAD-dependent enzymes that exhibit the para-cresolmethylhydroxylase-topology, also known as vanillyl oxidase-topology. Recently, we have reported the structural and biochemical characterization of two monolignol oxidoreductases from Arabidopsis thaliana, AtBBE13 and AtBBE15. Now, we have conducted a comprehensive site directed mutagenesis study for AtBBE15, to expand our understanding of the catalytic mechanism of this enzyme class. Based on the kinetic properties of active site variants and molecular dynamics simulations, we propose a refined, structure-guided reaction mechanism for the family of monolignol oxidoreductases. Here, we propose that this reaction is facilitated stepwise by the deprotonation of the allylic alcohol and a subsequent hydride transfer from the C alpha-atom of the alkoxide to the flavin. We describe an excessive hydrogen bond network that enables the catalytic mechanism of the enzyme. Within this network Tyr479 and Tyr193 act concertedly as active catalytic bases to facilitate the proton abstraction. Lys436 is indirectly involved in the deprotonation as this residue determines the position of Tyr193 via a cation-pi interaction. The enzyme forms a hydrophilic cavity to accommodate the alkoxide intermediate and to stabilize the transition state from the alkoxide to the aldehyde. By means of molecular dynamics simulations, we have identified two different and distinct binding modes for the substrate in the alcohol and alkoxide state. The alcohol interacts with Tyr193 and Tyr479 while Arg292, Gln438 and Tyr193 form an alkoxide binding site to accommodate this intermediate. The pH-dependency of the activity of the active site variants revealed that the integrity of the alkoxide binding site is also crucial for the fine tuning of the pK(a) of Tyr193 and Tyr479. Sequence alignments showed that key residues for the mechanism are highly conserved, indicating that our proposed mechanism is not only relevant for AtBBE15 but for the majority of BBE-like proteins.

Au coated ZnO/MWCNTs nanocomposites film-induced four-wave-mixing effect for multi-wavelength generation in erbium-doped fiber laser

High optical nonlinearity of nanostructured materials has been attested as a reliable technique for the generation of a multi-wavelength fiber laser. Herein, we experimentally demonstrate a simple and efficient method for generating continuous-wave (CW) multi-wavelength erbium-doped laser (EDFL) using a nonlinear effect of four-wave mixing (FWM) in a highly nonlinear nanocomposite film. A nanocomposite film containing gold nanostars coated zinc-oxide nanorods/multi-walled carbon nanotubes (ZnO-NRs@Au NSs/MWCNTs) embedded in polyvinyl alcohol (PVA) polymer was prepared, characterized, and then incorporated into the laser cavity to induce the FWM effect. Benefiting from the FWM processes, the induced intensity-dependent gain alleviates the intense gain-competition in the EDFL and increases the number of lasing wavelengths. A five-lasing line with a side mode suppression ratio (SMSR) of 36.3 dBm has been observed by appropriately adjusting the pump strength to 300 mW. Repeated measurements verified the high stability of the laser operating at room temperature with less than 3 dBm fluctuations on output power and 0.21 nm lasing wavelength drifts for all wavelengths cases. To the best of the author’s knowledge, this is the first demonstration of multi-wavelength fiber laser utilizing ZnO-NRs@Au NSs/MWCNTs nanocomposite. The results reveal the excellent nonlinear optical properties of the prepared nanocomposite and lay a foundation for its application in the generation of a multi-wavelength fiber laser.

Association of Opioid Use Disorder Treatment With Alcohol-Related Acute Events

Importance Persons with opioid use disorder (OUD) and co-occurring alcohol use disorder (AUD) are understudied and undertreated. It is unknown whether the use of medications to treat OUD is associated with reduced risk of alcohol-related morbidity. Objective To determine whether the use of OUD medications is associated with decreased risk for alcohol-related falls, injuries, and poisonings in persons with OUD with and without co-occurring AUD. Design, Setting, and Participants This recurrent-event, case-control, cohort study used prescription claims from IBM MarketScan insurance databases from January 1, 2006, to December 31, 2016. The sample included persons aged 12 to 64 years in the US with an OUD diagnosis and taking OUD medication who had at least 1 alcohol-related admission. The unit of observation was person-day. Data analysis was performed from June 26 through September 28, 2020. Exposures Days of active OUD medication prescriptions, with either agonist (ie, buprenorphine or methadone) or antagonist (ie, oral or extended-release naltrexone) treatments compared with days without OUD prescriptions. Main Outcomes and Measures The primary outcome was admission for any acute alcohol-related event defined by International Classification of Diseases, Ninth Revision and International Statistical Classification of Diseases and Related Health Problems, Tenth Revision codes. Conditional logistic regression was used to compare OUD medication use between days with and without an alcohol-related event. Stratified analyses were conducted between patients with OUD with and without a recent AUD diagnostic code. Results There were 8 424 214 person-days of observation time among 13 335 participants who received OUD medications and experienced an alcohol-related admission (mean [SD] age, 33.1 [13.1] years; 5884 female participants [44.1%]). Agonist treatments (buprenorphine and methadone) were associated with reductions in the odds of any alcohol-related acute event compared with nontreatment days, with a 43% reduction for buprenorphine (odds ratio [OR], 0.57; 95% CI, 0.52-0.61) and a 66% reduction for methadone (OR, 0.34; 95% CI, 0.26-0.45). The antagonist treatment naltrexone was associated with reductions in alcohol-related acute events compared with nonmedication days, with a 37% reduction for extended-release naltrexone (OR, 0.63; 95% CI, 0.52-0.76) and a 16% reduction for oral naltrexone (OR, 0.84; 95% CI, 0.76-0.93). Naltrexone use was more prevalent among patients with OUD with recent AUD claims than their peers without AUD claims. Conclusions and Relevance These findings suggest that OUD medication is associated with fewer admissions for alcohol-related acute events in patients with OUD with co-occurring AUD.

Preteen Behaviors and Sexual Orientation of High School Students Who Report Depressive Symptoms, United States, 2015-2017

Unhealthy preteen behaviors are associated with adolescent depression. However, little is known about preteen factors among sexual minority young people, a group at increased risk for teen depression and suicide. We completed weighted multivariate logistic regression analyses on data from the national 2015 and 2017 Youth Risk Behavior Survey of 30 389 high school students in the United States. Preteen sex, cigarette smoking, and alcohol and marijuana use were significantly more prevalent among lesbian, gay, or bisexual (LGB) and questioning students who reported depressive symptoms than among their heterosexual peers (adjusted prevalence ratio [APR] range, 1.33-2.34; all significant at P < .05). The only exception was that marijuana use among questioning students was not significantly different from use among heterosexual peers (APR = 1.34; P = .11). Assessment of preteen sex and substance use-especially among LGB and questioning young people, who are more prone to depressive symptoms and more likely to initiate risky preteen behaviors than their heterosexual counterparts-will facilitate a life course approach to sexual minority mental health that should begin by early adolescence. Novel microencapsulated yeast for the primary fermentation of green beer: kinetic behavior, volatiles and sensory profile

The development of innovative and more cost-effective approaches of making beer throughout continuous fermentation process remains a challenging problem, which is worthy of serious exploration. The current work focuses on the application of a commercial brewing yeast (S. cerevisiae Nottingham Ale), entrapped into chitosancalcium alginate double layer microcapsules, for the production of a Pale Ale beer. During the primary alcoholic fermentation, the consumption rate of fermentable brewing sugars and dissolved O-2, estimated by the Gompertz equation, was halved in the beer obtained by encapsulated yeast in comparison with the free cell. The physical-chemical parameters of beer (i.e. pH, alcohol content, color and bitterness) were not remarkably affected by the different yeast-inoculating form. However, the volatile profiles identified by means of HS-SPME-GC-MS analysis, significantly differed in terms of terpenes, esters and alcohols content, thus proving that the yeast inoculating form may typify the odor and flavor descriptors of the green beer.

Fine-tuning the architecture of loose nanofiltration membrane for improved water flux, dye rejection and dye/salt selective separation

Herein, loose nanofiltration membranes were prepared using non-solvent induced phase separation (NIPS) method. In NIPS, the non-solvent controls the morphology of the skin layer of the asymmetric membranes formed, while the solvent tunes the morphology of the lower layer. The membranes were made from polyethersulfone in n-methyl pyrrolidone (NMP) with the addition of dicarboxylic acids (glutaric acid, azelaic acid). Aliphatic alcohols (1-butanol, 1-octanol) were added to the solvent to tailor the skin layer pore morphology. Cross-sectional electron microscope images revealed the acids created nano-voids and shorter micro-voids. The addition of alcohols extended the micro-voids and improved symmetry. Pore size distribution results showed that the addition of the alcohols did indeed reduce the pore size as hypothesised. The performance of the modified polyethersulfone membranes was greatly improved in terms of pure water flux and salt passage without sacrificing the dye rejection. The loose nanofiltration membranes prepared in this study showed good dye/salt separation capability compared to most of the results reported in literature. The membranes were compacted with de-ionised water at an applied pressure of 4 bar for 5 h and rejections tests were done at 2 bar. It is hereby reported that the membranes achieved a high salt passage (Na2SO4 rejection varying between <3% and similar to 12%), as well as sustainable high Congo red rejection of 97%. The Role of Behavioral and Neurocognitive Functioning in Substance Use Among Youth with Perinatally Acquired HIV Infection and Perinatal HIV Exposure Without Infection

This study examined associations of self-regulatory behavior and cognitive functioning with substance use (SU) to inform interventions for youth with perinatal HIV infection (YPHIV) or exposure but uninfected (YPHEU). Youth aged 7-15 years (YPHIV, n = 390; YPHEU, n = 211) were followed longitudinally with cognitive testing and behavioral questionnaires including self-report of alcohol, marijuana, tobacco, and other SU. Cox proportional hazards analyses were used to examine correlates of initiating each substance for those without prior use at baseline and generalized estimating equation analyses were used to address associations of cognitive/behavioral measurements with SU prevalence for the entire sample. Lower self-reported self-regulation skills, but higher cognitive functioning abilities, were associated with initiation and prevalent use of alcohol and marijuana regardless of HIV status. Our findings suggest SU screening tools and self-regulation interventions developed for general adolescent populations should be implemented for those with PHIV, who may be at heightened risk for SU-related health consequences.

Interplay between personality and antisocial peer affiliation as prospective predictors of marijuana use and academic achievement in the transition into and out of college

Marijuana use has increased in recent cohorts and is associated with several negative outcomes, including poorer academic achievement. Less is known about how personality and peer factors in the first two years of college work together to predict problematic marijuana use and potentially co-occurring academic problems three years later. The longitudinal College Experiences Study (N = 209) was used to address this (N = 209, similar to 90% white, similar to 40% male); this study collected data from students in their first year of college, as well as similar to 1 year later (retention = 85%), and again similar to 4 years after the initial data collection (retention = 80%). Longitudinal data were analyzed via the traditional cross-lagged panel (CLPM) and the random-intercept CLPM approach. Results were consistent in that there was strong stability in problematic marijuana use, constraint, and antisocial peer affiliation across time, which were predominately influenced by stable, trait-like influences. These factors were also highly correlated but there was less evidence that one predicted the other over time. Nonetheless, greater constraint at Wave 1 was associated with significantly greater cumulative GPA in the transition out of college (beta s = 0.43-0.44). Results support the importance of early prevention and intervention of problematic marijuana use, as well as the possibility of using personality-targeted approaches in the first year of college to promote growth in behaviors related to constraint (e.g., staying organized, risk avoidant) in an effort to improve academic success and correlated substance use outcomes by the transition out of college.
