Enzyme cascades are efficient tools to perform multi-step synthesis in one-pot in a green and sustainable manner, enabling non-natural synthesis of valuable chemicals from easily available substrates by artificially combining two or more enzymes. Preparation of 3D Printed Chitosan/Polyvinyl Alcohol Double Network Hydrogel Scaffolds

In this work, a 3D printed double-network (DN) hydrogel scaffold is designed with chitosan (CS) and polyvinyl alcohol (PVA) as the framework matrix. The addition of PVA into the CS-based hydrogel clearly enhances the mechanical properties and lowers the swelling behaviors of the hydrogels. The crosslinking of CS with genipin can perform the pre-crosslinking to improve the viscosity and 3D printability of the hydrogel precursor, while increasing the PVA content results in lowering the viscosity and 3D printability of the pre-crosslinked hydrogel. The antibacterial property results of the DN hydrogel display that the hydrogel have favorable long-lasting antibacterial ability. The appropriate pre-crosslinked hydrogel with the CS/PVA mass ratio of 3:10 and pre-crosslinking time of 7 h is used for 3D printing to prepare the 3D printed porous DN hydrogels. Moreover, the anti-tumor drug doxorubicin (DOX) is loaded into the 3D printed porous DN hydrogels and the in vitro release study displays the sustainable drug release behavior. And the DOX release from hydrogel scaffold can be adjusted by the pH value of release environment. All of the results indicate that the porous DN CS/PVA hydrogel scaffolds have great application potential for tissue regeneration.

In vivo safety assessment, biodistribution and toxicology of polyvinyl alcohol microneedles with 160-day uninterruptedly applications in mice

Dissolving microneedles (DMNs) are widely used in drug delivery systems since they are based on one-step application, which is simple and convenient for patients, especially for the patients such as diabetes who need daily or long-term self-administration. In general, the matrix materials of DMNs are water-soluble materials that can release the encapsulated drugs gradually by dissolving in the skin without generating sharp needle waste. However, the matrix materials of DMNs will also leave in the skin after application. Thus, it is vital to evaluate whether the matrix material of DMNs dissolved in the skin will cause health risks such as toxicity to the body or some skin-related complications to patients who frequent or long-term administration. In this work, PVA, as one of the typical matrix materials of DMNs, was selected to prepare the DMNs to research the safety of PVA-based MNs to the body after being dissolved in the skin. Briefly, in a 160 – days trial, the healthy mice were daily administrated by PVA MNs. The results showed that PVA materials mainly accumulated in the skin tissues of mice after dissolving and the concentration of PVA in the insertion sites gradually decreased and was almost undetectable at 6 days after administration. The observation of general conditions, blood hematological analysis and histological examinations of the mice demonstrated that the PVA-based MNs do not cause appreciable toxicity to the healthy mice after daily insertion in a 160 – days trial. Altogether, these results encourage further studies of PVA MNs for biomedical applications and support translation of PVA-based DMNs from pre-clinical development into clinical trials.

The present study aimed to evaluate the effect of two ethanol vapor doses (250 and 500 ppm), 1-methylcyclopropene (1-MCP) (0.650 ppm) with or without ethylene application (150 ppm) on the aerobic and anaerobic metabolism and quality of ‘Elstar’, a fast ripening apple cultivar, and ‘Nicoter’, a slowly ripening apple cultivar, over 14 d of holding at room temperature (20 +/- 2 degrees C). Fruit were subjected to treatments for 24 h and analyses performed after 0, 7 and 14 d holding at room temperature (20 +/- 2 degrees C). For both cultivars studied, ethanol vapor treatments, especially 500 ppm, inhibited the ripening of apples even when combined with 150 ppm of ethylene, but not as much as 1-MCP treatment in ‘Nicoter’ apples. Fruit treated with ethanol vapor maintained lower electrolyte leakage, higher flesh firmness, greener color and had more healthy fruit. However, its application increased the pyruvate decarboxylase (PDC) and alcohol dehydrogenase (ADH) activity, acetaldehyde and ethyl acetate accumulation, but in concentrations below the odor threshold reported in the literature. The 1-MCP treatment increased decay incidence, reducing the healthy fruit amount in ‘Nicoter’ apples after 14 d, but maintained a very low ethylene production and respiration rate, which allowed higher acidity maintenance after 14 d holding at room temperature. There was no incidence of external and internal physiological disorders in both cultivars.

Effect of High-Dose Baclofen on Agitation-Related Events Among Patients With Unhealthy Alcohol Use Receiving Mechanical Ventilation A Randomized Clinical Trial

IMPORTANCE Unhealthy alcohol use can lead to agitation in the intensive care unit (ICU). OBJECTIVE To assess whether high-dose baclofen reduces agitation-related events compared with placebo in patients with unhealthy alcohol use receiving mechanical ventilation. DESIGN, SETTINGS, AND PARTICIPANTS This phase 3, double-blind, placebo-controlled, randomized clinical trial conducted in 18 ICUs in France recruited adults receiving mechanical ventilation who met criteria for unhealthy alcohol use. Patients were enrolled from June 2016 to February 2018; the last follow-up was in May 2019. INTERVENTIONS Baclofen (n = 159), adjusted from 50 to 150 mg per day based on estimated glomerular filtration rate, or placebo (n = 155) during mechanical ventilation up to a maximum of 15 days before gradual dose reduction over 3 to 6 days. MAIN OUTCOMES AND MEASURES The primary end point was the percentage of patients with at least 1 agitation-related event over the treatment period. Secondary outcomes included duration of mechanical ventilation, length of ICU stay, and 28-day mortality. RESULTS Among 314 patients who were randomized (mean age, 57 years; 60 [17.2%] women), 313 (99.7%) completed the trial. There was a statistically significant decrease in the percentage of patients who experienced at least 1 agitation-related event in the baclofen group vs the placebo group (31 [19.7%] vs 46 [29.7%]; difference, -9.93%[95% CI, -19.45% to -0.42%]; adjusted odds ratio, 0.59 [95% CI, 0.35-0.99]). Of 18 prespecified secondary end points, 14 were not significantly different. Compared with the placebo group, the baclofen group had a significantly longer median length of mechanical ventilation (9 vs 8 days; difference, 2.00 [95% CI, 0.00-3.00]; hazard ratio [HR] for extubation, 0.76 [95% CI, 0.60-0.97]) and stay in the ICU (14 vs 11 days; difference, 2.00 [95% CI, 0.00-4.00]; HR for discharge, 0.70 [95% CI, 0.54-0.90]). At 28 days, there was no significant difference in mortality in the baclofen vs placebo group (25.3% vs 21.6%; adjusted odds ratio, 1.24 [95% CI, 0.72-2.13]). Delayed awakening (no eye opening at 72 hours after cessation of sedatives and analgesics) occurred in 14 patients (8.9%) in the baclofen group vs 3 (1.9%) in the placebo group. CONCLUSIONS AND RELEVANCE Among patients with unhealthy alcohol use receiving mechanical ventilation, treatment with high-dose baclofen, compared with placebo, resulted in a statistically significant reduction in agitation-related events. However, considering the modest effect and the totality of findings for the secondary end points and adverse events, further research is needed to determine the possible role of baclofen in this setting and to potentially optimize dosing.

Release of catechin from Azivash gum-polyvinyl alcohol electrospun nanofibers in simulated food and digestion media

In this study, the electrospinning process was applied for the encapsulation of catechin in nanofibers from Azivash (Corchorus olitorius. L) gum-polyvinyl alcohol. Public implementation of a web-based program for veterans with risky alcohol use and PTSD: A RE-AIM evaluation of VetChange

Background: Evidence-based web and mobile interventions are available for a range of mental health concerns. Little is known about how self-administered web interventions are used outside of controlled research trials, and there is a critical need to empirically examine real-world public implementation of such programs. To this end, the aim of the current study was to evaluate and describe outcomes of a nationwide public implementation of VetChange, a self-administered web intervention for veterans with problematic alcohol use and symptoms of PTSD. Method: The study used the RE-AIM framework to organize outcomes along five key dimensions: reach, effectiveness, adoption, implementation, and maintenance. This naturalistic observation study included program use information for all who registered an account with VetChange during the 2-year study period and who self-identified as a returning veteran. We collected program use data automatically via normal website operation; a subset of program users provided additional self-report outcome data. The study used linear multilevel mixed modeling to evaluate changes in alcohol use and PTSD symptoms over a six-month postregistration period. Results: VetChange successfully reached a large, geographically diverse sample of returning veterans with risky drinking and PTSD symptoms. Despite variable, overall modest, rates of intervention use over time, registered users demonstrated significant improvements in drinking, PTSD, and quality of life, and participants maintained these outcomes at a six-month follow-up. Conclusions: Given the observed low cost per acquisition, positive clinical outcomes, and the potential to produce long-term cost savings through reduced health care burden associated with chronic alcohol use disorder and PTSD, this study demonstrates how web-based interventions can provide public health benefits and reduce long-term health care costs.

Differential Microwave Resonator Sensor for Real-Time Monitoring of Volatile Organic Compounds

In this work, a noninvasive microwave gas sensor suitable for accurate and real-time tracking of volatile organic compounds concentration is presented. The proposed differential sensor was comprised of two independent split-ring resonators in power splitter/combiner configuration to enhance the robustness of the sensor to changes in environmental parameters. The designwas fabricatedon a Rogers RT/duroid 5880 high-frequency substrate and operated at f(s) = 5.12 GHz and f(r) = 5.65 GHz, referring to the sensing and reference resonant frequencies, respectively. A thin layer of polydimethylsiloxane was used as an interface layer in the sensing resonator region with the strongest electric field to enable gas sensing with high sensitivity. The performance of the sensor was validated by introducing different volatile organic compounds, such as acetone, methanol, ethanol, and isopropyl alcohol. Time-based responses of the sensor demonstrated the capability of the sensor to monitor different concentrations of acetone vapor ranging from low, 387 to 1935 ppm, with the sensitivity of 0.02 mdB/ppm and high concentrations, 3878 to 62048 ppm, with sensitivity of 0.0125 mdB/ppm.

Live predator stress in adolescence results in distinct adult behavioral consequences and dorsal diencephalic brain activation patterns

Exposure to traumatic events during childhood increases the risk of adult psychopathology, including anxiety, depression, alcohol use disorders and their co-morbidity. Early life trauma also results in increased symptom complexity, treatment resistance and poor treatment outcomes. The purpose of this study was to establish a novel rodent model of adolescent stress, based on an ethologically relevant life-threatening event, live predator exposure. Rats were exposed to a live predator for 10 min. at three different time points (postnatal day (PND)31, 46 and 61). Adult depression-, anxiety-like behaviors and ethanol consumption were characterized well past the last acute stress event (two weeks). Behavioral profiles across assessments were developed to characterize individual response to adolescent stress. CNS activation patterns in separate groups of subjects were characterized after the early (PND31) and last predator exposure (PND61). Subjects exposed to live-predator adolescent stress generally exhibited less exploratory behavior, less propensity to venture into open spaces, a decreased preference for sweet solutions and decreased ethanol consumption in a two-bottle preference test. Additional studies demonstrated blunted cortisol response and CNS activation patterns suggestive of habenula, rostromedial tegmental (RMTg), dorsal raphe and central amygdala involvement in mediating the adult consequences of adolescent stress. Thus, adolescent stress in the form of live-predator exposure results in significant adult behavioral and neurobiological disturbances. Childhood trauma, its impact on neurodevelopment and the subsequent development of mood disorders is a pervasive theme in mental illness. Improving animal models and our neurobiological understanding of the symptom domains impacted by trauma could significantly improve treatment strategies.
